1/22 ISSN 0100-2945 http://dx.doi.org/10.1590/0100-29452017411 GRAPEVINE VIRUS DISEASES: ECONOMIC IMPACT AND CURRENT ADVANCES IN VIRAL PROSPECTION AND MANAGEMENT1 MARCOS FERNANDO BASSO2, THOR VINÍCIUS MArtins FAJARDO3, PASQUALE SALDARELLI4 ABSTRACT-Grapevine (Vitis spp.) is a major vegetative propagated fruit crop with high socioeconomic importance worldwide. It is susceptible to several graft-transmitted agents that cause several diseases and substantial crop losses, reducing fruit quality and plant vigor, and shorten the longevity of vines. The vegetative propagation and frequent exchanges of propagative material among countries contribute to spread these pathogens, favoring the emergence of complex diseases. Its perennial life cycle further accelerates the mixing and introduction of several viral agents into a single plant. Currently, approximately 65 viruses belonging to different families have been reported infecting grapevines, but not all cause economically relevant diseases. The grapevine leafroll, rugose wood complex, leaf degeneration and fleck diseases are the four main disorders having worldwide economic importance. In addition, new viral species and strains have been identified and associated with economically important constraints to grape production. In Brazilian vineyards, eighteen viruses, three viroids and two virus-like diseases had already their occurrence reported and were molecularly characterized. Here, we review the current knowledge of these viruses, report advances in their diagnosis and prospection of new species, and give indications about the management of the associated grapevine diseases. Transmission of virus by the progeny of crosses between XQhinema diversicaudatüm (Nematoda : Dorylaimoidea) from Italy and Scotland Derek J. F. SUMMARY Transmission of the type-British strainsof arabis mosaic (AMV-T) and strawberry latent ringspot viruses (SLRV-T) and a strain of SLRV from Italy (SLRV-Ip) by FI and F2 hybrid Xiphinenza diversicaudatum was examined in the laboratory. The hybrid nematodes were crossbred from populations which readily (Scotland) and only infrequently (Italy) transmitted viruses.The ability of X. diversicaudatum hybrids to transmit viruses was foundto be inherited withthe choice of both maternaland paternal parents affecting the hybrids ability to transmit viruses. It is possible that the genetic influence on the hybrids ability to transmit viruses was cytoplasmically inherited. The principal factor likely to be involved is the ability of X. di ver sic au da tu??^ selectively and specifically to retain virus particles at sites of retention within its feeding apparatus. Three New Species of B Otryotinia on Ranunculaceae1

GROVES3 Research Branch, Department of Agriculture, Ottawa, Canada Received October 10, 1962 Abstract Three new species of Botryotinia on Caltha palustris L., Ranunculus septen- trionalis Poir., and Ficaria verna Huds. (Ranunculaceae) are described as B. calthae Hennebert and Elliott, B. ranunculi Hennebert and Groves, and B. ficariarum Hennebert. Each of the three species has a Botrytis state of the 73. cinerea complex, and they thus constitute additions to the species already segregated from that complex, i.e. Botryotinia fuckeliana, 73. convoluta, B. draytoni, and B. pelargonii. The Botrytis state of B. ficariarum can be distinguished morp hologically. While B. ranunculi is a North American species and B. ficariarum an European one, B. calthae is reported from both continents. Introduction Botryotinia, a genus of the Sclerotiniaceae, was erected by Whetzel (1945) for four species formerly assigned to Sclerotinia Fuckel, but which differ from the true Sclerotinia species mainly in their erumpent, planoconvexoid sclerotia which are firmly attached to the substrate, and in the possession of a conidial state belonging to the form-genus Botrytis Pers. The type species is Botryotinia convoluta (Drayton) Whetzel in which the conidial state is a Botrytis of the cinerea type or B. cinerea sensu lato, and the other species included were Botryotinia fuckeliana (De Bary) Whetzel, of which the conidial state is Botrytis cinerea Pers. or B. cinerea sensu stricto, and Botryotinia ricini (Godfrey) Whetz. and B. porri (v. Beyma) Whetz. In the latter two species the conidial states would not be considered Botrytis species of the cinerea type. -

KASAPOĞLU ULUDAMAR1* Tohid BEHMAND1 İbrahim Halil ELEKCİOĞLU1 Abstract In this study, penetration, population changes and reproduction rates of root lesion nematodes, Pratylenchus neglectus (Rensch, 1924), Pratylenchus penetrans (Cobb, 1917) and Pratylenchus thornei Sher & Allen, 1953 (Tylenchida: Pratylenchidae), at 3, 7, 14, 21, 28, 35, 42, 49 and 56 d after inoculation in chickpea Bari 2, Bari 3 (Cicer reticulatum Ladiz) and Cermi [Cicer echinospermum P.H.Davis (Fabales: Fabaceae)] were assessed in a controlled environment room in 2018-2019. No juveniles were observed in the roots in the first 3 d after inoculation. Although, population density of P. thornei reached the highest in Cermi (21 d), Bari 3 (42 d) and the lowest observed on Bari 2. Pratylenchus neglectus reached the highest population density in Bari 3 and Cermi on day 28. The population density of P. neglectus was the lowest in Bari 2. Also, population density of P. penetrans reached the highest in Bari 3 cultivar within 49 d, similar to P. thornei, whereas Bari 2 and Cermi had low population densities during the entire experimental period. Occurrence of Apple mosaic virus (ApMV), Prune dwarf virus (PDV), Prunus necrotic ringspot virus (PNRSV), Apple chlorotic leaf spot virus (ACLSV), and Plum pox virus (PPV) was investigated by RT-PCR and DAS ELISA detection methods. The de- tection rates of both methods were compared. Screening of occurrence of Strawberry latent ringspot virus (SLRSV), Arabis mosaic virus (ArMV), Tomato ringspot virus (ToRSV) and Petunia asteroid mosaic virus (PeAMV) was performed by DAS-ELISA. In total, 38% of the tested trees by RT-PCR were infected at least with one of the analysed viruses. Among those 30.7% were in- fected with PNRSV and 16.4% with PDV, while ApMV, ACLSV and PPV were detected in few samples. The most widespread mixed infection was the combination of PDV+PNRSV. Observed symptoms characteristic for PPV were confirmed with RT-PCR and D strain was detected. Com- parative analyses showed that detection rates by RT-PCR and DAS ELISA in plums depended on the particular virus tested. -

Culture-based methods for clinical samples are often negative, difficult and time-consuming and mainly identify isolates to the genus level, and sometimes only as Mucorales. Therefore, applying fast and accurate diagnosis methods such as molecular approaches seems necessary. This study aims at isolating Mucorales for determination of Rhizopus genus between the isolates using molecular methods. Methods: In this descriptive observational study, a total of 500 samples were collected from air and different surfaces and inoculated on Sabouraud Dextrose Agar supplemented with chloramphenicol. Then, the fungi belonging to Mucorales were identified and their pure culture was provided. DNA extraction was done using extraction kit and the chloroform method. After amplification, the samples belonging to Mucorales were identified by observing 830 bp bands. For enzymatic digestion, enzyme BmgB1 was applied for identification of Rhizopus species by formation of 593 and 235 bp segments. Results: One hundred pure colonies belonging to Mucorales were identified using molecular methods and after enzymatic digestion, 21 isolates were determined as Rhizopus species. -

Plant Pathology & Quarantine 8(2), 116–121, Doi 10.5943/ppq/8/2/2 Abstract During spring 2017, red delicious and Granny Smith apples with soft rot symptoms were collected from commercial markets in Saudi Arabia. The causal agent was isolated from infected fruits and its pathogenicity was confirmed by inoculation assays. To confirm the identity, total genomic DNA was extracted and multi-locus sequence data targeting two gene markers (ITS, ACT) was sequenced. Phylogenetic analysis confirmed the presence of two distinct clades; Saudi isolate was placed within a clade comprising Rhizopus oryzae and R. delemar reference isolates. Based on morphological characteristics, molecular identification and pathogenicity test, the fungus was identified as Rhizopus oryzae. This is the first report of Rhizopus soft rot caused by R. oryzae from stored apple fruits in Saudi Arabia.
