O’Reilly,* Nils Holler,‡ Jerry Adams,* and Andreas Strasser* From *The Walter and Eliza Hall Institute, Melbourne, Victoria 3050, Australia; and the ‡Institute of Biochemistry, University of Lausanne, Epalinges CH-1066, Switzerland Abstract The short life span of granulocytes, which limits many inflammatory responses, is thought to be influenced by the Bcl-2 protein family, death receptors such as CD95 (Fas/APO-1), stress-acti- vated protein kinases such as p38 mitogen-activated protein kinase (MAPK), and proinflamma- tory cytokines like granulocyte colony-stimulating factor (G-CSF). To clarify the roles of these various regulators in granulocyte survival, we have investigated the spontaneous apoptosis of granulocytes in culture and that induced by Fas ligand or chemotherapeutic drugs, using cells from normal, CD95-deficient lpr, or vav-bcl-2 transgenic mice. CD95-induced apoptosis, which required receptor aggregation by recombinant Fas ligand or the membrane-bound ligand, was unaffected by G-CSF treatment or Bcl-2 overexpression. Conversely, spontaneous and drug-induced apoptosis occurred normally in lpr granulocytes but were suppressed by G-CSF treatment or Bcl-2 overexpression. Although activation of p38 MAPK has been implicated in granulocyte death, their apoptosis actually was markedly accelerated by specific inhibitors of this kinase. These results suggest that G-CSF promotes granulocyte survival largely through the Bcl-2–controlled pathway, whereas CD95 regulates a distinct pathway to apoptosis that is not required for either their spontaneous or drug-induced death. -

(UCI) today announced it has entered into an agreement to acquire digital media assets as part of the bankruptcy proceedings of Gawker Media Group, Inc. and related companies that produce content under a series of original brands that reach nearly 50 million readers per month, according to comScore. UCI will acquire the digital media assets for $135 million, subject to certain adjustments, and these assets will be integrated into Fusion Media Group (FMG), the division of UCI that serves the young, diverse audiences that make up the rising American mainstream. The deal, which will be accounted for as an asset purchase, includes the following digital platforms, Gizmodo, Jalopnik, Jezebel, Deadspin, Lifehacker and Kotaku. UCI will not be operating the Gawker.com site. Citation for the Award of the Doctor of Laws (Honoris Causa) to Professor Suzanne Cory AC When Suzanne Cory graduated from the Science Faculty at the University of Melbourne molecular biology was in its infancy. In her enthusiasm for this new science she pursued a PhD in Cambridge at the Medical Research Council’s famed Laboratory of Molecular Biology. Immersed in a culture of discovery, her pioneering PhD studies determined the sequence of a transfer RNA, utilising new technology developed by Fred Sanger, one of the three Nobel Laureates in the institute. After three years’ post‐doctoral work at the University of Geneva, Suzanne Cory returned to Australia with her husband, Jerry Adams, to the Walter and Eliza Hall Institute of Medical Research where they helped to introduce gene‐cloning technology to Australia. Since then, her long association with the Institute, including thirteen years as Director, and with the University of Melbourne, has been instrumental in building the international reputation of both institutions and the strength of the Parkville biomedical research network. Suzanne Cory has made major contributions to advancing the science of immunology, cancer and genetics, in a lifelong dynamic research partnership with Adams. Her work is published widely and cited frequently. The positions held by the many PhD candidates and Post‐Doctoral Fellows she has supervised demonstrate her intergenerational influence in developing scientific leaders for the future. The list of accolades and awards Suzanne Cory has received in recognition of her contributions to science is long and distinguished. She was elected a Fellow of the Australian Academy of Science in 1986 and since has been elected to numerous sister organisations across the globe. -

Celebrating DNA Day 2015: Engaging Teachers to Inspire Students
April 7, 2015 Spring brings a welcome increase in temperatures (and decrease in snowfall!), as well as other changes around NIH. At the end of March, my friend and colleague, Dr. Harold Varmus, ended his tenure as Director of the National Cancer Institute. I have had the pleasure of collaborating with Harold on joint NCI-NHGRI efforts [most notably The Cancer Genome Atlas (TCGA)] and learning from him around the Institute/Center Director’s table. We have also enjoyed having the Varmus research laboratory within the NHGRI Intramural Research Program. His scientific leadership has been invaluable to NCI and NIH, and we wish him all the best in his future endeavors in New York City. Also of note, I just returned from The Tech Museum of Innovation in San Jose, California, where I joined several leading Bay Area genomics leaders for an engaging public program entitled “Big Data, Genomics, and Precision Medicine.” The exhibition Genome: Unlocking Life’s Code is still on display in San Jose, and it will remain there until the end of this month, after which it will travel to my hometown of St. Louis, Missouri. We look forward to additional future events associated with the exhibition as it travels across North America. Hann, 2 and Allan Bradley 1 tlnstitute for Molecular Genetics, Baylor College of Medicine, Houston, Texas 77030 USA; ~Department of Cell Biology, Vanderbilt University, School of Medicine, Nashville, Tennessee 37232-2175 USA To directly assess c-myc function in cellular proliferation, differentiation, and embryogenesis, we have used homologous recombination in embryonic stem cells to generate both heterozygous and homozygous c-myc mutant ES cell lines. The mutation is a null allele at the protein level. Mouse chimeras from seven heterozygous cell lines transmitted the mutant allele to their offspring. The analysis of embryos from two clones has shown that the mutation is lethal in homozygotes between 9.5 and 10.5 days of gestation. The embryos are generally smaller and retarded in development compared with their littermates. Pathologic abnormalities include the heart, pericardium, neural tube, and delay or failure in turning of the embryo. Heterozygous females have reduced fertility owing to embryonic resorption before 9.5 days of gestation in 14% of implanted embryos, c-Myc protein is necessary for embryonic survival beyond 10.5 days of gestation; however, it appears to be dispensable for cell division both in ES cell lines and in the the embryo before that time. Wotgali Village is a slum area. The aim of the current research is to find out the performance of slum base infrastructure using Lakip (Government Agency Accountability Report) of Kotaku (City without Slums) Program. The first step was performed by identifying the initial settlement (base line), then some treatments on the facilities and infrastructure (finish line), performance evaluation stages of the preparation, planning, implementation, and sustainability stages. The research used qualitative and quantitative approach. Qualitative measurement began with a numerical assessment of the results on the level of regional slum. In addition, quantitative data used the Lakip simulation using results of the Kotaku program stage performance. Results of the initial condition reach value of 32%. It is categorized as slight slums with an average sectoral slum of 36.68%. Basic infrastructure development which has final technical reached 24% and is included in slight slum with an average sectoral of 27.50%. The performance of LAKIP Kotaku was in preparation phase 90.46% (very good performance), planning stage was 89.93% (very good performance), implementation stage was 90.25% (very good performance), sustainability stage was 85.21% (better performance). Thus, the achievement of Kotaku program is 85.19% with a range of value of 80-90. -
