It has been well characterized that long-term insect-bacterial mutualisms cause genome reduction resulting in extremely small genomes, some even approaching sizes more similar to organelles than bacteria. While several symbioses have been described, each provides a limited view of a single or few stages of the process of reduction and the minority of these are of extracellular symbionts. This dissertation aims to address the knowledge gap in the genome evolution of extracellular insect symbionts using the stink bug – Pantoea system. Specifically, how do these symbionts genomes evolve and differ from their free- living or intracellular counterparts? In the introduction, we review the literature on extracellular symbionts of stink bugs and explore the characteristics of this system that make it valuable for the study of symbiosis. We find that stink bug symbiont genomes are very valuable for the study of genome evolution due not only to their biphasic lifestyle, but also to the degree of coevolution with their hosts. i In Chapter 1 we investigate one of the traits associated with genome reduction, high mutation rates, for Candidatus ‘Pantoea carbekii’ the symbiont of the economically important pest insect Halyomorpha halys, the brown marmorated stink bug, and evaluate its potential for elucidating host distribution, an analysis which has been successfully used with other intracellular symbionts. -

To address whether nitrogen-provi- sioning symbionts might be important in the early stages of ant trophic shifts, we studied bacteria from the Argentine ant, Linepithema humile – an invasive species that has transitioned towards greater consumption of sugar-rich, nitrogen-poor foods in parts of its introduced range. Bacteria were present at low densities in most L. humile workers, and among those yielding quality 16S rRNA amplicon sequencing data, we found just three symbionts to be common and dominant. -

Feeding on a variety of carbohydrate and protein sources, they have an infrabuccal filter that limits passage of large food particles to their gut. A variety of yeasts have been found associated with the infrabuccal pocket and the nests of these ants. The yeast Schwanniomyces polymorphus is associated with the carpenter ant Camponotus vicinus. To examine a possible nutritional association between this yeast and ant, we reared small sub-colonies of defaunated and non-defaunated C. vincus brood on several artificial diets where various nutritional components were removed. Part of the testing involved exposure of brood to these diets and cells of S. polymorphus. The strain of A. bogorensis used was isolated from spoiled commercial fruit-flavored drinking water. The cranberry juice was analyzed for polyphenols, organic acids, and carbohydrates using high-performance liquid chromatography and liquid chromatography–mass spectrometry techniques. The adhesive abilities of bacterial cells in culture medium supplemented with cranberry juice were determined using luminometry and microscopy. The viability of adhered and planktonic bacterial cells was determined by the plate count method, and the relative adhesion coefficient was calculated. This strain of A. bogorensis was characterized by strong adhesion properties that were dependent upon the type of surface. The highest level of cell adhesion was found on the polystyrene. However, in the presence of 10% cranberry juice, attachment of bacterial cells was three times lower. Chemical analysis of juice revealed the presence of sugars, organic acids, and anthocyanins, which were identified as galactosides, glucosides, and arabinosides of cyanidin and peonidin. -

Community composition can differ across host species with increasing dietary and genetic divergence, yet gut microbiota can also vary between conspecific hosts and across an individual’s lifespan. Through exploration of such intraspecific variation and its correlates, molecular profiling of microbial communities can generate and test hypotheses on the causes and conse- quences of symbioses. In this study, we used 454 pyrosequencing and TRFLP to achieve these goals in an herbivorous ant, Cephalotes varians, exploring variation in bacterial communities across colonies, populations and workers reared on different diets. C. varians bacterial communities were dominated by 16 core species present in over two-thirds of the sampled colonies. Core species comprised multiple genotypes, or strains and hailed from ant-specific clades containing relatives from other Cephal- otes species. Yet three were detected in environmental samples, suggesting the poten- tial for environmental acquisition. In spite of their prevalence and long-standing relationships with Cephalotes ants, the relative abundance and genotypic composition of core species varied across colonies. Diet-induced plasticity is a likely cause, but only pollen-based diets had consistent effects, altering the abundance of two types of bacte- ria. -

Brown1,2 and Jennifer J. Wernegreen1,2* Abstract Background: Symbiotic associations between gut microbiota and their animal hosts shape the evolutionary trajectories of both partners. The genomic consequences of these relationships are significantly influenced by a variety of factors, including niche localization, interaction potential, and symbiont transmission mode. In eusocial insect hosts, socially transmitted gut microbiota may represent an intermediate point between free living or environmentally acquired bacteria and those with strict host association and maternal transmission. Results: We characterized the bacterial communities associated with an abundant ant species, Camponotus chromaiodes. While many bacteria had sporadic distributions, some taxa were abundant and persistent within and across ant colonies. Specially, two Acetobacteraceae operational taxonomic units (OTUs; referred to as AAB1 and AAB2) were abundant and widespread across host samples. Dissection experiments confirmed that AAB1 and AAB2 occur in C. chromaiodes gut tracts. We explored the distribution and evolution of these Acetobacteraceae OTUs in more depth. We found that Camponotus hosts representing different species and geographical regions possess close relatives of the Acetobacteraceae OTUs detected in C. chromaiodes. Phylogenetic analysis revealed that AAB1 and AAB2 join other ant associates in a monophyletic clade. This clade consists of Acetobacteraceae from three ant tribes, including a third, basal lineage associated with Attine ants. This ant-specific AAB clade exhibits a significant acceleration of substitution rates at the 16S rDNA gene and elevated AT content. -

However, most studies on the gut microbiota only take into exam individual behavior without considering social dynamics. Contrarily, animals and humans live in complex societies where they constantly adjust physiology and behavior to social interactions. To improve our understanding of how microbes and hosts interact and produce functional individual, social and collective phenotypes, we need to broaden our experimental approaches to a group-level dimension. The ideal models for this purpose are social animals living in stable symbioses with microbes, such as eusocial insects. In our research, we investigate Camponotus carpenter ants and their obligate bacterial symbiont Blochmannia from a behavioral ecology perspective. We aim to create ant colonies including differential proportions of bacteria-free individuals by suppressing Blochmannia with antibiotics. Then, using a machine learning-based video tracking system, we will study network features and group-level behavior of such experimental colonies. Diversity and Organization of the Ground Foraging Ant Faunas of Forest, Grassland and Tree Crops in Papua New Guinea

Abstract Thirty samples of ants were taken in each of seven habitats: primary forest, rubber plantation, coffee plantation, oilpalm plantation, kunai grassland, eucalypt savannah and urban grassland. Sixty samples were taken in cocoa plantations. A total of 156 species was taken, and the frequency of occurrence of each in each habitat is given. Eight stenoecious species are suggested as habitat indicators. Habitats fell into a series according to the similarity of their ant faunas: forest, rubber and coffee, cocoa and oilpalm, kunai and savannah, urban. This series represents an artificial, discontinuous succession from a complex stable ecosystem to a simple unstable one. Availability of species suitably preadapted to occupy habitats did not appear to limit species richness. Habitat heterogeneity and stability as affected by human interference did seem to account for inter-habitat variability in species richness. Species diversity was compared between habitats using four indices: Fisher et al.; Margalef; Shannon; Brillouin. Correlation of diversity index with habitat hetero- geneity plus stability was good for the first two, moderate for Shannon, and poor for Brillouin. Greatest diversity was found in rubber, the penultimate in the series of habitats according to hetero- geneity plus stability ('maturity'). Equitability exceeded the presumed maximum in rubber, and was close to the maximum in all habitats. -

LeBoeuf Abstract Trophallaxis is a complex social fluid exchange emblematic of social insects and of ants in particular. Trophallaxis behaviors are present in approximately half of all ant genera, distributed over 11 subfamilies. Across biological life, intra- and inter-species exchanged fluids tend to occur in only the most fitness-relevant behavioral contexts, typically transmitting endogenously produced molecules adapted to exert influence on the receiver’s physiology or behavior. Despite this, many aspects of trophallaxis remain poorly understood, such as the prevalence of the different forms of trophallaxis, the components transmitted, their roles in colony physiology and how these behaviors have evolved. With this review, we define the forms of trophallaxis observed in ants and bring together current knowledge on the mechanics of trophallaxis, the contents of the fluids transmitted, the contexts in which trophallaxis occurs and the roles these behaviors play in colony life. We identify six contexts where trophallaxis occurs: nourishment, short- and long-term decision making, immune defense, social maintenance, aggression, and inoculation and maintenance of the gut microbiota. Though many ideas have been put forth on the evolution of trophallaxis, our analyses support the idea that stomodeal trophallaxis has become a fixed aspect of colony life primarily in species that drink liquid food and, further, that the adoption of this behavior was key for some lineages in establishing ecological dominance.
