PRODUCT INFORMATION Antigen Description Olfactory receptors interact with odorant molecules in the nose, to initiate a neuronal response that triggers the perception of a smell. The olfactory receptor proteins are members of a large family of G-protein-coupled receptors (GPCR) arising from single coding-exon genes. Olfactory receptors share a 7-transmembrane domain structure with many neurotransmitter and hormone receptors and are responsible for the recognition and G protein-mediated transduction of odorant signals. The olfactory receptor gene family is the largest in the genome. The nomenclature assigned to the olfactory receptor genes and proteins for this organism is independent of other organisms. Immunogen A synthetic peptide of human OR4D1 is used for rabbit immunization. For this study, skin samples were derived from abdominal or breast tissues of healthy women (30-40 years old). Samples were received 1 day post-surgery and used directly for microdissection or cell extraction. Microdissected human skin was cultured at 37°C under 5% CO2, in DMEM (Dutscher, #L0060-500) plus 10% FBS (Sigma Aldrich, #F7524), 1% Penicillin/Streptomycin (Sigma Aldrich, #P0781-100), and 1% of minimum medium non-essential amino acids 10X (Gibco, #11140-035). For keratinocyte culture, cells were isolated from human skin epidermis. Briefly, skin samples were cut into small pieces and immersed in Thermolysin (Sigma Aldrich, #T7902-100mg) at 4°C overnight. Epidermis was then carefully detached from dermis before being incubated with Trypsin-EDTA for 15 minutes at 37°C. Trypsin inhibitor was next added, the mixture centrifuged and the supernatant discarded before adding KGM2 Medium (Promocell, #C- 20011) with 1% Penicillin/Streptomycin (Sigma Aldrich, #P0781-100). NHEK were then maintained in culture at 37 °C under 5 % CO2 and 95 % relative humidity.

Although deletions involving 17q22 have been reported in the literature, only four of the eight patients reported were identified by array-comparative genomic hybridization (array-CGH) or flourescent in situ hybridization. Here, we describe five new patients with 1.8–2.5-Mb microdeletions involving 17q22 identified by array-CGH. We also present one patient with a large karyotypically visible deletion involving 17q22, fine-mapped to B8.2 Mb using array-CGH. We show that the commonly deleted region in our patients spans 0.24 Mb and two genes; NOG and C17ORF67. The function of C17ORF67 is not known, whereas Noggin, the product of NOG, is essential for correct joint development. In common with the 17q22 patients reported previously, the disease phenotype of our patients includes intellectual disability, attention deficit hyperactivity disorder, conductive hearing loss, visual impairment, low set ears, facial dysmorphology and limb anomalies. All patients displayed NOG-related bone and joint features, including symphalangism and facial dysmorphology.

Surgical intervention to address massive aortic root dilatation is uncommon as dissection rarely occurs, and death invariably results from congestive heart failure and recurrent respiratory infections. Case presentation: An 11 month old female with a prenatal diagnosis of aortic root dilatation was aggressively treated both medically and surgically. Genetic testing revealed a partial deletion in FBN1 exons 46–50 and an addition 17q11 microdeletion. Valve-sparing aortic root remodeling was successful at eliminating her risk of a type A aortic dissection. Prior to surgery, however, her root caused significant left atrial and left lower pulmonary vein compression, which has not completely resolved. Conclusions: Aortic dilatation occurs rapidly despite aggressive medical management. Root remodeling and atrioventricular valve repair are possible, but the durability of the latter is uncertain. It is possible that our patient’s combined gene mutations are exacerbating her disease. Abstract The expression of human olfactory receptors in non-olfactory tissues has been documented since the early nineties, however, until recently their functional roles were largely unknown. Many studies have demonstrated that these G-protein coupled receptors (GPCRs) are actively involved in various cellular processes. Here, we summarized current evidence describing the most prominent expression and functional data for these ectopic olfactory receptors. Further studies focused on discovering their ligands, both agonists and antagonists, will be necessary to fully characterize molecular mechanisms underlying their functional roles in human physiology and pathophysiology.
