  • Thirty-One Years of Research and Development in the Vine Cacti Pitaya in Israel
Improving Pitaya Production and Marketing THIRTY-ONE YEARS OF RESEARCH AND DEVELOPMENT IN THE VINE CACTI PITAYA IN ISRAEL Yosef Mizrahi Department of Life Sciences, Ben-Gurion University of the Negev Beer-Sheva, 8441901, Israel E-mail: [email protected] ABSTRACT Taxonomy: The vine cacti have three genera and many species in each one of them. The genera are: Hylocereus, Selenicereus and Epiphyllum. Only Selenicereus megalanthus combine characteristics of the first two genera. Genetics and breeding: Selenicereus megalanthus is tetraploids (4n) while all others are diploids (2n). We obtained inter-clonal, interspecies, and intergeneric hybrids with much improved characteristics, to the point that today we have new excellent hybrids which can provide fruits from July to May. Physiology: All plants are of the CAM (crassulacean acid metabolism) photosynthetic pathway; hence use 10% of water other plants are using in the same environment. CO2 enrichment increases both the vegetative and reproductive productions. Nitrogen fertilization should accompany the CO2 enrichment to get maximum efficiency. Stomata were found on the fruit surface functioning through the CAM pathway. Stomatal density is much higher on the fruit scale than on the fruit surface. Cytokinins induces and gibberelliic acid (GA3) delays flowering. Scale shrinking is the major reason to shorten the shelf life. Root system is very shallow maximum depth, 40cm. Uses: There are more uses to the plants parts other than the fruit, such as fruit pigments as coloring agents, edible flowers and more. Keywords: cacti, pitaya, dragon fruit, taxonomy, genetics, breeding, physiology, cytokinins, GA3 , flowering, post-harvest INTRODUCTION The vine-cacti pitaya of the Cactaceae, subfamily Cactoideae, tribe Hylocereeae is known to have been used for thousands of years by the indigenous people of the Americas (Ortiz-Hernández and Carrillo-Salazar 2012).

FUNGAL SUPPRESSIVE ACTIVITIES OF SELECTED RHIZOSPHERIC STREPTOMYCES SPP. ISOLATED FROM HYLOCEREUS POLYRHIZUS KAMALANATHAN RAMACHANDARAN FACULTY OF SCIENCE UNIVERSITY OF MALAYA KUALA LUMPUR 2014 FUNGAL SUPPRESSIVE ACTIVITIES OF SELECTED RHIZOSPHERIC STREPTOMYCES SPP. ISOLATED FROM HYLOCEREUS POLYRHIZUS KAMALANATHAN RAMACHANDARAN DISSERTATION SUBMITTED IN FULFILMENT OF THE REQUIREMENTS FOR THE DEGREE OF MASTER OF SCIENCE INSTITUTE OF BIOLOGICAL SCIENCES FACULTY OF SCIENCE UNIVERSITY OF MALAYA KUALA LUMPUR 2014 ABSTRACT Actinomycetes, mainly Streptomyces spp., have been extensively studied as potential biocontrol agents against plant pathogenic fungi. This study was aimed at isolating and screening Streptomyces strains from rhizosphere soils of Hylocereus polyrhizus collected in Kuala Pilah for potential in vitro antifungal activity. A total of 162 putative strains of actinomycetes was isolated from moist-heat treated soil plated on starch- casein-nitrate agar, humic-acid-vitamin agar and raffinose-histidine agar. Based on the ability to produce abundant aerial mycelium, 110 strains were categorised as Streptomycete-like. Seven main groups based on aerial mycelium colour observed in this study were grey (41.4%), white (37.7%), brown (8.0%), orange (4.3%), yellow (4.3%), green (2.5%) and black (1.9%). Three pathogenic fungi, namely, Fusarium semitactum, Fusarium decemcellulare and Fusarium oxysporum were isolated from the diseased stem regions of Hylocereus polyrhizus. The actinomycetes were screened for in vitro antagonistic activity against the isolated pathogenic fungi. In the qualitative screening, 23 strains were able to inhibit at least one of the three pathogenic fungi. In the quantitative screening, three strains, C17, C68 and K98, showed the highest antagonistic activity (70-89%) against all the fungal pathogens.

Characterising plant pathogen communities and their environmental drivers at a national scale A thesis submitted in partial fulfilment of the requirements for the Degree of Doctor of Philosophy at Lincoln University by Andreas Makiola Lincoln University, New Zealand 2019 General abstract Plant pathogens play a critical role for global food security, conservation of natural ecosystems and future resilience and sustainability of ecosystem services in general. Thus, it is crucial to understand the large-scale processes that shape plant pathogen communities. The recent drop in DNA sequencing costs offers, for the first time, the opportunity to study multiple plant pathogens simultaneously in their naturally occurring environment effectively at large scale. In this thesis, my aims were (1) to employ next-generation sequencing (NGS) based metabarcoding for the detection and identification of plant pathogens at the ecosystem scale in New Zealand, (2) to characterise plant pathogen communities, and (3) to determine the environmental drivers of these communities. First, I investigated the suitability of NGS for the detection, identification and quantification of plant pathogens using rust fungi as a model system.

However, its scientific report of disease is still lacking, primarily on anthracnose disease. This study was then conducted to investigate the distribution of anthracnose disease on dragon fruit and to correlate its occurrence with weather and cultural data. Approach: Survey and sampling were conducted on dragon fruit-growing areas in Peninsular Malaysia since December 2007 until August 2008 to measure the Disease Incidence (DI) and Disease Severity (DS). The diseased stem and fruit were sampled and brought to laboratory for isolation and identification. DI data were plotted with DS and then correlated using Pearson correlation with weather and cultural data. Results: Of the 43 surveyed-farms in 11 states, DI and DS were successfully recorded on three dragon fruit species from 36 farms (83.72%).

Their infections with phytopathogens, however, not only have an impact on the plant physiology and appearance but also may lead to air contamination and, in consequence, affect human health. Present research is a case study of Echinopsis oxygona, known as the Easter Lily cactus, infected with a fungal pathogen. The phenotypic and molecular studies were conducted to identify the etiological agent of the lesions. Colony appearance and growth on various media, as well as the presence of fungal propagation structures were evaluated. Internal transcribed spacer (ITS) sequences from the isolated cultures were obtained, and the BLAST analysis was performed to estimate genetic similarity. The phenotypic and molecular tests allowed to identify the pathogen as Fusarium oxysporum and, to our knowledge, it is the first report on E. oxygona (as a pot plant) infected with this species in Poland. This fungus is a soil-born species and a well-known toxin producer.
