Nelson1,4 1Department of Molecular and Human Genetics, Baylor College of Medicine, Houston, Texas 77030, USA; 2Department of Statistics, Rice University, Houston, Texas 77030, USA; 3Center for Genome Information, Department of Environmental Health, University of Cincinnati, Cincinnati, Ohio 45267, USA To facilitate association-based linkage studies we have studied the linkage disequilibrium (LD) and haplotype architecture around five genes of interest for cancer risk: ATM, BRCA1, BRCA2, RAD51, and TP53. Single nucleotide polymorphisms (SNPs) were identified and used to construct haplotypes that span 93–200 kb per locus with an average SNP density of 12 kb. These markers were genotyped in four ethnically defined populations that contained 48 each of African Americans, Asian Americans, Hispanic Americans, and European Americans. Haplotypes were inferred using an expectation maximization (EM) algorithm, and the data were analyzed using DЈ, R2, Fisher’s exact P-values, and the four-gamete test for recombination. LD levels varied widely between loci from continuously high LD across 200 kb to a virtual absence of LD across a similar length of genome. LD structure also varied at each gene and between populations studied. This variation indicates that the success of linkage-based studies will require a precise description of LD at each locus and in each population to be studied. One striking consistency between genes was that at each locus a modest number of haplotypes present in each population accounted for a high fraction of the total number of chromosomes.

Dr. Daniel Woo, M.S., M.D. Dr. Divakar Choubey, Ph.D. ABSTRACT Prostate cancer (PCa) is the most commonly diagnosed visceral malignancy with over one-third of all incident cancers and the second leading cause of cancer deaths in men in the United States. Age, ethnicity and family history are among the classical epidemiological risk factors. Both environmental and genetic factors contribute to the etiopathogenesis of PCa. The region on 19q11-13, identified through linkage and association studies, was previously shown to harbor genes for PCa susceptibility as well as for biologic aggressiveness. Recently, admixture mapping and a number of independently performed genome-wide association studies (GWAS) further identified strong association signals on chromosome 8q24. The only biologically plausible candidate gene for cancers on 8q24 is the myc oncogene, located ~500 kb downstream to the identified region. The myc protein is a key regulator of cell cycle, growth and differentiation, apoptosis, genomic instability and angiogenesis. At the cellular level, myc synergistically interacts with mutL homolog 1 (MLH1), one of the key genes in mismatch repair (MMR) of DNA.

In October 2002, a California jury convicted William Curtis Wilson of “first degree murder with use of a dangerous weapon during commission of an attempted rape and a lewd act upon a child. The court sentenced him to a term of life in prison without possibility of parole.”2 DNA evidence played a central role in obtaining the conviction. Radhakrishna Rao: A Century in Statistical Science Nandini Kannan1 and Debasis Kundu2 Abstract In this article, the authors provide a glimpse into the legendary career of centenarian Profes- sor C. Radhakrishna Rao, a career that is inextricably linked to the history of modern statistics from Pearson and Fisher to the age of big data and Artificial Intelligence. A scientist of extraor- dinary vision, his ground-breaking contributions have influenced not only the field of statistics, but the mathematical, social, biological, and economic sciences. An outstanding researcher, a gifted and inspirational teacher who could make complex concepts come to life in a classroom, and a mentor who demanded excellence from his students, Professor Rao continues to inspire generations of statisticians and scientists around the world. 1 Initial Years: 1941-1950 Calyampudi Radhakrishna Rao was born on September 10, 1920 in Huvanna Hadagali in what was then the Madras Presidency, and is now a town in the Indian state of Karnataka. As the eighth of ten children, he was named Radhakrishna after the Hindu deity Krishna, also the eight-born according to Hindu mythology. His father, C. D. Naidu, an Inspector in the Criminal Investigation Department, recognized Rao's gift for mathematics and encouraged him to pursue a career in research. His mother instilled the discipline and work ethic that have stayed with him throughout his extraordinary career. Identification (or narrowing down) of sources of contributors in a DNA mixture is a complex process, for which the present DNA community is progressively using a probabilistic approach, which involves two steps. First, the DNA mixture profile is deconvoluted into single-source individual profiles with the number of contributors assumed to be known. In the second step, the likelihoods of such deconvoluted sets of profiles are computed under two alternative hypotheses of sources of such a DNA mixture, from which the strength of involvement of (one or more) known persons are assessed. In contrast, interpretation of DNA mixtures without mixture deconvolution is a simpler approach, but in most scenarios yields a diminished strength of DNA evidence. This presentation will address the advantages of the probabilistic approaches of DNA mixture interpretation, explicitly enumerating the assumptions of the probabilistic approaches and parameters required to carry out such analyses. Limitations of these methods will also be specified, particularly in the context of allele sharing between contributors in a mixed DNA sample, uneven contributions of DNA from different donors, and presence of low copy number DNA from (one or more) contributors.

    INTERNATIONAL COMMISSION ON RADIOLOGICAL PROTECTION 28 September 2018 In Memoriam Dr Ranajit Chakraborty (April 17, 1946 – September 23, 2018) Dr. Ranajit Chakraborty, a world-renowned scientist in the field of human radiation genetics, died September 23 at the age of 72. Born in West Bengal India, he received his PhD in Biostatistics from the Indian Statistical Institute in Calcutta. He was the current Director of the Center for Computational Genomics at the Institute of Applied Genetics, and Professor of Molecular and Medical Genetics at the University of North Texas Health Science Center in Fort Worth, Texas. In 1973, Ranajit was hired by the Center for Demographic and Population Genetics (now Human Genetics Center) at the University of Texas School of Public Health. Ranajit left Texas for Ohio in 2001, to become the Director of the Center for Genome Information at the University of Cincinnati. He published extensively on radiation genetics with K. Sankaranarayanan (Sankar) – their seminal papers are referenced extensively, most notably by the United Nations Scientific Committee on the Effects of Atomic Radiation (UNSCEAR) in their 2001 report on Heritable Effects. Among his many collaborations throughout the world, Ranajit contributed to the genetic studies of the children of parents treated for cancer, and served many years with the American National Council on Radiation Protection and Measurements. In 1993, he first joined ICRP as a member of the task group that would go on to produce Publication 79 on the Genetic Susceptibility to Cancer. In 2005, he joined Committee 1 on a full-time basis, providing world-class experience and knowledge to ICRP on a volunteer basis.
