Die Allele (*R1, *R2, *S1 und *S2) des A. gambiae complement-like thioester-containing Protein 1 (TEP1) bestimmen die Fitness der Mücken, welches die männlichen Fertilität und den Resistenzgrad der Mücke gegen Pathogene wie Bakterien und Malaria- Parasiten. Dieser Kompromiss zwischen Reproduktion und Immunnität hat Auswirkungen auf die Größe der Mückenpopulationen und die Rate der Malariaübertragung, wodurch der TEP1 Lokus ein Primärziel für neue Malariakontrollstrategien darstellt. Wie die genetische Diversität von TEP1 die genetische Struktur natürlicher Vektorpopulationen beeinflusst, ist noch unklar. Mouse Tep1 Conditional Knockout Project (CRISPR/Cas9)
https://www.alphaknockout.com Mouse Tep1 Conditional Knockout Project (CRISPR/Cas9) Objective: To create a Tep1 conditional knockout Mouse model (C57BL/6J) by CRISPR/Cas-mediated genome engineering. Strategy summary: The Tep1 gene (NCBI Reference Sequence: NM_009351 ; Ensembl: ENSMUSG00000006281 ) is located on Mouse chromosome 14. 55 exons are identified, with the ATG start codon in exon 2 and the TGA stop codon in exon 55 (Transcript: ENSMUST00000006444). Exon 10 will be selected as conditional knockout region (cKO region). Deletion of this region should result in the loss of function of the Mouse Tep1 gene. To engineer the targeting vector, homologous arms and cKO region will be generated by PCR using BAC clone RP23-321A24 as template. Cas9, gRNA and targeting vector will be co-injected into fertilized eggs for cKO Mouse production. The pups will be genotyped by PCR followed by sequencing analysis. Note: Mice homozygous for a disruption in this gene show no obvious phenotype. No changes are seen in telomerase activity or telomere length. Exon 10 starts from about 19.88% of the coding region. The knockout of Exon 10 will result in frameshift of the gene. The size of intron 9 for 5'-loxP site insertion: 4878 bp, and the size of intron 10 for 3'-loxP site insertion: 990 bp. The size of effective cKO region: ~610 bp. The cKO region does not have any other known gene. Here, we have addressed the role of telomerase activity in the proliferation of human tumor and immortal cells by inhibiting TERT function. Inducible dominant-negative mutants of hTERT dramatically reduced the level of endogenous telomerase activity in tumor cell lines. Clones with short telomeres continued to divide, then exhibited an increase in abnormal mitoses followed by massive apoptosis leading to the loss of the entire population. This cell death was telomere-length dependent, as cells with long telomeres were viable but exhibited telomere shortening at a rate similar to that of mortal cells. It appears that telomerase inhibition in cells with short telomeres lead to chromosomal damage, which in turn trigger apoptotic cell death. These results provide the first direct evidence that telomerase is required for the maintenance of human tumor and immortal cell viability, and suggest that tumors with short telomeres may be effectively and rapidly killed following telomerase inhibition. It is not fully understood whether genetic variations in telomere-related genes are associated with development and progression in PCa patients. METHODS: Six potentially functional single-nucleotide polymorphisms (SNPs) of three key telomere-related genes were evaluated in 1015 PCa cases and 1052 cancer-free controls, to test their associations with risk of PCa. Among 426 PCa patients who underwent radical prostatectomy (RP), the prognostic significance of the studied SNPs on biochemical recurrence (BCR) was also assessed using the Kaplan–Meier analysis and Cox proportional hazards regression model. The relative telomere lengths (RTLs) were measured in peripheral blood leukocytes using real-time PCR in the RP patients. RESULTS: TEP1 rs1760904 AG/AA genotypes were significantly associated with a decreased risk of PCa (odds ratio (OR): 0.77, 95% confidence interval (CI): 0.64–0.93, P = 0.005) compared with the GG genotype. By using median RTL as a cutoff level, RP patients with TEP1 rs1760904 AG/AA genotypes tended to have a longer RTL than those with the GG genotype (OR: 1.55, 95% CI: 1.04–2.30, P = 0.031).

The vault complex consists of three proteins and non-coding vault RNAs. It has been shown that the non-coding vault RNA encodes regulatory small vault RNAs (svRNAs). These svRNAs associate with the RNA-induced silencing complex and regulate gene expression similarly to microRNAs. It is unknown which genes the svRNAs regulate, but they are thought to regulate genes relating to multidrug resistance and intracellular antigen transport. I have selected several genes of interest relating to cancer, multidrug resistance, inflammation, and the autoimmune response to predict whether the svRNAs regulate these genes. Since the svRNAs regulate genes similarly to microRNA, I used microRNA target prediction tools to find potential functional binding sites for the svRNAs in the 3' untranslated regions of the selected gene messenger RNAs. I found several target sites that have a very high potential to be functional binding sites for the svRNAs. These results can be used to conduct experiments to verify that the svRNAs bind to the predicted target sites and regulate the expression of the targeted gene.

The analysis of a large number of individuals provided the power to detect relatively small differences in expression between sexes, and the use of an intercross allowed analysis of the genetic control of sexually dimorphic gene expression. Microarray analysis of 23,574 transcripts revealed that the extent of sexual dimorphism in gene expression was much greater than previously recognized. Thus, thousands of genes showed sexual dimorphism in liver, adipose, and muscle, and hundreds of genes were sexually dimorphic in brain.

Blackburn2 Department of Biochemistry and Biophysics, University of California San Francisco, San Francisco, California 94143-2200, USA Telomerase contains an essential RNA, which includes the template sequence copied by the reverse transcription action of telomerase into telomeric DNA. Using phylogenetic comparison, we identified seven conserved sequences in telomerase RNAs from Kluyveromyces budding yeasts. We show that two of these sequences, CS3 and CS4, are essential for normal telomerase function and can base-pair to form a putative long-range pseudoknot. Disrupting this base-pairing was deleterious to cell growth, telomere maintenance, and telomerase activity. Restoration of the base-pairing potential alleviated these phenotypes. Mutating this pseudoknot caused a novel mode of shifting of the boundaries of the RNA template sequence copied by telomerase. A phylogenetically derived model of yeast TER structure indicates that these RNAs can form two alternative predicted core conformations of similar stability: one brings the CS3/CS4 pseudoknot spatially close to the template; in the other, CS3 and CS4 move apart and the conformation of the template is altered. We propose that such disruption of the pseudoknot, and potentially the predicted telomerase RNA conformation, affects polymerization to cause the observed shifts in template usage.
