Pro tento cíl byly zvoleny tři lokusy: jaderný úsek ITS a dva chloroplastové úseky trnL – trnF a ndhF. Vhodnost vybraných markerů k rekonstrukci fylogeneze na nízké taxonomické úrovni byla testována prostřednictvím dvou metod: maximální úspornosti a maximální věrohodnosti. Topologie získaných stromů byla velice podobná napříč hodnocenými markery a také použitými metodami. Na všech zkonstruovaných stromech se vyčlenilo několik monofyletických skupin: "Lactuca sensu lato", skupina asijských taxonů, "afro-americká" skupina, skupina taxonů perennis/undulata a sororia/triangulata. Skupina "Lactuca sensu lato" byla tvořena dvěma příbuznými, dobře odlišenými skupinami "Lactuca sensu stricto" a "Scariola". Vnitřní mezidruhové vztahy v rámci těchto hlavních skupin zůstaly částečně nevyřešené. Nevyřešené zůstaly také fylogenetické vztahy mezi hlavními skupinami. Z práce lze zhodnotit, že použité lokusy jsou dostatečné k vyčlenění hlavních evolučních linií v rámci subtribu Lactucinae, avšak nejsou dostatečné na to, aby dokázaly rozřešit vnitřní vztahy mezi jednotlivými taxony a mezi jednotlivými liniemi. Denisa Plucnarová Title: Utilization of selected nuclear and chloroplast DNA markers in the study of phylogeny of selected taxa from the genus Lactuca Type of thesis: Master thesis Department: Department of Botany, Faculty of Science, Palacký University in Olomouc, Supervisor: RNDr. Ľuboš Majeský, Ph.D. Consultant: RNDr. Miloslav Kitner, Ph.D. The year of defense: 2017 Abstract The aim of this master thesis was to evaluate the suitability of selected molecular markers for reconstruction of the phylogeny of the genus Lactuca and to resolve interspecific relationships among selected species of the genus. For this purpose three loci were selected: nuclear locus ITS, and two chloroplast loci trnL – trnF and ndhF. Suitability of the selected markers for reconstruction of phylogeny on the low taxonomic level was tested using two main methods, Maximum parsimony, and Maximum Likelihood. The topology of inferred trees was very similar across all of the tested loci and methods. On all of the inferred trees, several monophyletic groups were consistently present. Namely, the group "Lactuca sensu lato"; group of "Afro- American" taxa, group of "Asian" taxa, perennis/undulata clade, and sororia/triangulata clade. The "Lactuca sensu lato" group consist of two related and clearly separated subgroups, "Lactuca sensu stricto" and "Scariola". Within the inferred groups, the interspecific relationships remained partially unresolved as well as the relationships between the inferred main groups. The results showed that used molecular markers are sufficient for recognition of wider groups/lineages within the subtribe Lactucinae, however, are not sufficient enough for resolving internal relationships within the wider groups and between the main lineages. Do daného rodu se řadí jeden kulturní druh L. sativa L., který má mnoho variet či kultivarů různých barev a tvarů listů, má také hospodářský význam a pěstuje se jako listová zelenina. Mnoho planě rostoucích druhů se vyskytuje po celém světě a to zejména v Evropě, Asii, Severní a Střední Americe a Africe a Australii (Lebeda et al., 2007). Biogeograficky

Dillman, Ecologist USDA Forest Service, Tongass National Forest [email protected] Vascular Plants of Negelle-Borona, Ethiopia, USFS IP Introduction This report provides supplemental information to the Inventory and Assessment of Biodiversity report prepared for the US Agency for International Development (USAID) following the 2011 mission to Negelle- Borona region in southern Ethiopia (Dillman 2012). As part of the USAID supported Pastoralist Livelihood Initiative (PLI), this work focused on the biodiversity of the kallos (pastoral reserves). This report documents the vascular plant species collected and identified from in and around two kallos near Negelle (Oda Yabi and Kare Gutu). This information can be utilized to develop a comprehensive plant species list for the kallos which will be helpful in future vegetation monitoring and biodiversity estimates in other locations of the PLI project. This list also identifies plants that are endemic to Ethiopia and East Africa growing in the kallos as well as plants that are non-native and could be considered invasive in the rangelands. Methods Field work was conducted between November 28 and December 9, 2011 (the end of the short rainy season). The rangeland habitats visited are dominated by Acacia and Commifera trees, shrubby Acacia or dwarf shrub grasslands.

Методом сканирующей электронной микроскопии (СЭМ) исследована скульптура по- верхности семянок 371 вида из 168 родов, 21 трибы, пяти подсемейств семейства Asteraceae. У 75 видов из 45 родов, относящихся к 8 трибам, а также у Symphyllocarpus exilis (вида с неустановленным таксономическим положением в системе семейства) было выявлено наличие папилл. На поверхности семянок большинства видов трибы Cichorieae находятся папиллы разнообразного строения. Только у видов этой трибы папиллы об- разуют гребни (виды подтрибы Hypochaeridinae) и на поверхности экзокарпия многих видов находятся окру- глые гладкие гранулы эпикутикулярного воска, отсутствующие у видов других триб. На поверхности семянок видов триб Anthemideae, Astereae, Athroismeae, Calenduleae, Gnaphalieae, Heliantheae, Senecioneae папиллы имеются только у некоторых видов. У исследованных нами видов триб Barnadesieae, Mutisieae, Cynareae, Vernonieae, Arctotideae, Inuleae, Helenieae, Coreopsideae, Tageteae, Madieae, Millerieae, Eupatorieae папиллы не обнаружены. Длина, форма папилл, их расположение на поверхности семянки различны у разных видов и могут быть использованы как таксономический признак. Ключевые слова: Asteraceae, семянки, папиллы, морфология, сканирующая электронная микроскопия (СЭМ), систематика. Summary. The sculpture of the surface of cypselae (scanning electron microscopy) of 371 species of 168 gene­ ra, 21 tribes, five subfamilies of the family Asteraceae was investigated. 75 species of 45 genera belonging to 8 tribes and the species Symphyllocarpus exilis with uncertain taxonomic position, papillae were detected. Papillae of diverse structure have been revealed on the surface of the cypselae of the majority of Cichorieae species.

The area adjacent the villages is characterized by self-consumption agriculture, with a population that is on average poor, still very tied to traditions and almost entirely unaffected by modernization and technology. The aim of the present study was to investigate and record local knowledge regarding the use of wild and traditionally cultivated plants used for traditional medicine and for other everyday purposes (e.g., food, fibers and timber). Methods: Ten traditional local healers, with solid botanical knowledge, were interviewed between June and August 2014 by means of semi-structured questionnaires. For each mentioned plant species, the Swahili folk name and, when possible, the classification by family, genus and species was recorded as well as the part of the plant used, the preparation method and the main uses (medicine, food or others). Results: In total 196 species were mentioned of which 118 could be botanically classified. The identified species belong to 44 different botanical families, with that of the Leguminosae being the most representative (24 species). The plants were mostly used as medical treatments (33.3% of the species) and foods (36.8%), and to produce wood and fibers (19.4%).

Sequences for two datasets, one of the nuclear rDNA ITS region, the other of five concatenated non-coding chloroplast DNA markers including the petD region and the psbA-trnH, 59trnL(UAA)-trnF, rpl32-trnL(UAG) and trnQ(UUG)-59rps16 spacers, were, with few exceptions, newly generated for 130 samples of 78 species. The sampling spans the entire subtribe Lactucinae while focusing on its Chinese centre of diversity; more than 3/4 of the Chinese Lactucinae species are represented. The nuclear and plastid phylogenies inferred from the two independent datasets show various hard topological incongruences. They concern the internal topology of major lineages, in one case the placement of taxa in major lineages, the relationships between major lineages and even the circumscription of the subtribe, indicating potential events of ancient as well as of more recent reticulation and chloroplast capture in the evolution of the subtribe. The core of the subtribe is clearly monophyletic, consisting of the six lineages, Cicerbita, Cicerbita II, Lactuca, Melanoseris, Notoseris and Paraprenanthes.
