TLR : Toll-Like Receptor i Universitas Sumatera Utara ABSTRAK Salah satu bakteri pada biofilm yang terbentuk pada infeksi endodontik adalah Fusobacterium nucleatum. Dari aspek mikrobiologis, infeksi endodonti dapat dirawat dengan prosedur kemikal dan mekanikal (cleaning and shaping). Irigasi kombinasi EDTA dan NaOCl adalah metode yang paling sering direkomendasikan. Namun kelemahan utama NaOCl adalah sitotoksisitasnya ketika ekstrusi ke dalam jaringan periradikuler dan kombinasi tersebut menghasilkan erosi yang kuat pada dentin. Akibat kelemahan bahan irigasi tersebut, penggunaan bahan alami sebagai alternatif bahan irigasi saluran akar diharapkan lebih biokompatibel dan salah satu bahan alami yang dapat dikembangkan sebagai bahan irigasi adalah buah lerak (Sapindus rarak DC). Penelitian ini menggunakan sampel koloni bakteri Fusobacterium nucleatum dan dibiakkan dengan media Crystal Violet Erythtromycin (CVE) Agar. Ekstrak buah lerak dengan konsentrasi 6,25, 12,5%, 25% dan dibandingkan dengan NaOCL 2,5% dan EDTA 17% dengan waktu 24 jam, 48 jam, 72 jam untuk diuji antiadhesi, biomassa dan porositas setelah diinteraksikan dengan bakteri F.Nucleatum. Berdasarkan analisis statistik kruskal wallis dan one way anova serta uji lanjutan nya dengan LSD dan mann whitney test, hasil penelitian ini menunjukkan bahwa ekstrak buah lerak dari berbagai konsentrasi (6,25%, 12,5%, 25%) sama baiknya dengan larutan NaOCl 2,5%+EDTA 17% yang menjadi gold standar dalam irigasi saluran akar gigi dalam mencegah adhesi dan menghambat perkembangan bakteri Fusobacterium nucleatum. Secara klinis NaOCl 2,5%+EDTA 17% memiliki tingkat porositas paling tinggi dan berbeda signifikan dengan ekstrak lerak 6,25%, 12,5% dan 25%. Sehingga dapat di asumsikan bahwa ekstrak lerak dalam berbagai konsentrasi (6,25%, 12,5%, 25%) tidak menyebabkan tingkat porositas yang berlebihan dibandingkan dengan NaOCl 2,5%+EDTA 17% yang menjadi gold standar irigasi saluran akar gigi. Ekstrak buah lerak memiliki kemampuan dalam mencegah adhesi dan menghambat perkembangan bakteri F.nucleatum serta secara klinis tidak menyebabkan porositas yang berlebihan pada dinding saluran akar gigi. Kata kunci : Ekstrak buah lerak, antiadhesi, Fusobacterium nucleatum, irigasi saluran akar, porositas saluran akar ii Universitas Sumatera Utara ABSTRACT One of the bacteria in biofilms formed in endodontic infections is Fusobacterium nucleatum. From the microbiological aspect, endodontic infections can be treated with chemical and mechanical procedures (cleaning and shaping). Combination of EDTA and NaOCl irrigation is the most commonly recommended method. However, the main disadvantage of NaOCl is its cytotoxicity when extrusion into periradicular tissue and this combination results in strong erosion of dentin. As a result of the weakness of the irrigation material, the use of natural material as an alternative to root canal irrigation is expected to be more biocompatible and one of the natural materials that can be developed as irrigation is lerak fruit (Sapindus rarak DC). This study used a sample of the bacterial colony Fusobacterium nucleatum and was cultured with Crystal Violet Erythtromycin (CVE) agar. Lerak fruit extract with a concentration of 6.25, 12.5%, 25% and compared with 2.5% NaOCL and 17% EDTA with 24 hours, 48 hours, 72 hours to be tested for anti-adhesion, biomassa and porosity after being interacted with F.Nucleatum bacteria. Based on the statistical analysis of kruskal wallis and one way anova and their follow-up tests with LSD and mann whitney test, the results of this study showed that lerak extract from various concentrations (6.25%, 12.5%, 25%) was as good as NaOCl solution 2.5% + 17% EDTA which became the gold standard in irrigation of root canals in preventing adhesion and inhibiting the development of the bacterium Fusobacterium nucleatum. Clinically, 2.5% NaOCl + 17% EDTA has the highest porosity level and is significantly different with 6.25%, 12.5% and 25% lerak extracts. So it can be assumed that lerak extract in various concentrations (6.25%, 12.5%, 25%) does not cause excessive porosity compared to 2.5% NaOCl + 17% EDTA which is the gold standard of tooth root canal irrigation. Lerak fruit extract has the ability to prevent adhesion and inhibit the development of F.nucleatum bacteria and clinically does not cause excessive porosity in the root canal wall of the tooth. Thesis submitted in accordance with the requirements of the University of Liverpool for the degree of Doctor in Philosophy by Hannah Betts July 2013 - 2 - - 3 - Abstract Due to forest degradation and loss, the use of ecological restoration techniques has become of particular interest in recent years. One such method is the Framework Species Approach (FSA), which was developed in Queensland, Australia. The Framework Species Approach involves a single planting (approximately 30 species) of both early and late successional species. Species planted must survive in the harsh conditions of an open site as well as fulfilling the functions of; (a) fast growth of a broad dense canopy to shade out weeds and reduce the chance of forest fire, (b) early production of flowers or fleshy fruits to attract seed dispersers and kick start animal-mediated seed distribution to the degraded site. The Framework Species Approach has recently been used as part of a restoration project in Doi Suthep-Pui National Park in northern Thailand by the Forest Restoration Research Unit (FORRU) of Chiang Mai University. FORRU have undertaken a number of trials on species performance in the nursery and the field to select appropriate species. However, this has been time-consuming and labour- intensive. It has been suggested that the need for such trials may be reduced by the pre-selection of species using their functional traits as predictors of future performance. Here, seed, leaf and wood functional traits were analysed against predictions from ecological models such as the CSR Triangle and the pioneer concept to assess the extent to which such models described the ecological strategies exhibited by woody species in the seasonally-dry tropical forests of northern Thailand.

Ordo, famili maupun spesies yang dibahas dalam Bahan Ajar ini sebagian besar didasarkan pada manfaat dalam bidang ekonomi, obat, bahan pangan, maupun dalam ekologi. Selain manfaat faktor lain yang juga dipertimbangkan dalam pemilihan ordo maupun famili adalah penyebarannya (yang dipilih terutama yang banyak di temukan di Indonesia). The present study was carried out to evaluate the safety of ethanol extract of lerak (sapindus rarak. DC) fruits. Methods: Lerak fruits were extracted by continuous extraction using the Soxhlet apparatus and ethanol 96 %. Extraction results was continued to acute toxicity by using a fixed dose method and sub-chronic toxicity was performed according to the OECD guideline. In an acute toxicity study, a single dose of 2000 and 5000 mg/kg bw of lerak fruits extract was administered p. o (orally) to healthy female Wistar rats following a sighting study. The animals were observed for mortality and clinical signs for 24 hour and then daily for 14 days. In the sub-chronic toxicity study, the extract was administered orally at doses of 50, 100 and 500 mg/kg bw /day. The animals were given by ethanol extract of lerak fruits once daily for 28 days, the administrations were stopped on the 28th day, while for the satellite group still observed until 14 days during the post observation period for assessment of reversibility, persistence or delayed occurrence of toxicity.

Pengenalan tumbuhan secara taksonomi merupakan merupakan salah satu langka untuk meningkatkan rasa kepedulian mahasiswa terhadap lingkungan sekitar khususnya tumbuhan. Bahan Ajar ini terdiri dari 14 bab yang membahas tumbuhan Spermatophyta atau tumbuhan berbiji. Spermatophyta terdiri dari 2 devisi yaitu Phinophyta dan Magnoliophyta dibahas dalam Bahan Ajar ini, namun karena luasnya cakupan taksonomi maka pembahasan disesuaikan dengan silabus yang telah disusun. Bahan Ajar ini lebih banyak membahas Magnoliophyta khususnya karena devisi tersebut mendominasi tumbuhan di permukaan bumi saat ini. Magnoliophyta terdiri dari 10 kelas. Setiap anak kelas Magnoliophyta dibahas mulai dari ordo, famili dan jenis serta manfaatnya. Ordo, famili maupun spesies yang dibahas dalam Bahan Ajar ini sebagian besar didasarkan pada manfaat dalam bidang ekonomi, obat, bahan pangan, maupun dalam ekologi.
