Biological Activity: In a functional ELISA, immobilized recombinant Human ACVR1B (0.2μg/mL) will bind recombinant Human -1 (Cat no.: PHC8014, PHC8015, PHC8011, PHC8013) with a linear range of 0.8–100 ng/mL.

Biological Function: ACVR1B or ALK-4 is a type I transmembrane serine/threonine receptor consisting of a 103 aa extracellular domain, a 23 aa transmembrane domain and a 356 aa cytoplasmic domain. ACVR1B acts as a transducer of Activin or activin like ligand signals. Activin binds to either type II receptor ACVR2A or ACVR2B which then forms a stable complex with ACVR1B. In the process ACVR1B is phosphorylated within its cytoplasmic domain by the type II receptor and the cascade is initiated. ACVR1B also transduces signals of , GDF-1, and Vg1; however, unlike activin, they require other coreceptor molecules such as the protein Cripto.

Sterility: Filtered prior to lyophilization through a 0.22 micron sterile filter.

Production: Recombinant Human ACVR1B is expressed in Human Embryonic Kidney 293 cells and purified via sequential chromatography.

Reconstitution We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Recommendation: Reconstitute lyophilized Human ACVR1B in sterile, distilled water to a concentration of 0.1–0.5 mg/mL. Stock solutions should be apportioned into working aliquots and stored at ≤ –20°C. Further dilution should be made in medium or buffered solution containing carrier protein, such as PBS with 0.1% BSA.

Storage: Lyophilized Human ACVR1B should be stored at 2°C to 8°C, preferably desiccated. Store reconstituted Human ACVR1B at ≤ –20°C (not in a frost-free freezer). Keep freeze-thaw cycles to a minimum.
