    Biosearch Nyika: Malawi 2004 Edited by Marianne J Overton FOREWORD Peter Overton It is ten years since the Biosearch Nyika project was first mooted and agreement with the Director of National Parks and Wildlife obtained for our exploration of the remoter parts of the Nyika National Park. Over this period the teams have focused mainly on the northern part of the park where patrolling has been very limited and our gathering of intelligence has been most helpful to the Nyika management. In 2004 we undertook the most challenging expedition to date, launched from the extreme north of the park at Uledi, a four-hour drive from Thazima. The team‟s first challenge was to cross the unbridged North Rukuru River with all their supplies. They then had to climb up the western escarpment of the Mpanda ridge to a point on the Mpero River, where they set up a Base Camp, from which to launch out on their surveys. The greatest achievement was to climb both Mpanda and Kawozya and discover the remote Bleak House, now derelict but offering stunning views over Lake Malawi and far beyond. At this point they could certainly claim to be in remote country since this old site is much talked about but very rarely seen by visitors. We have yet to have clear information about who built it, when and why. Perhaps it was a holiday „retreat‟ for Livingstonia or a staging post for missionaries who conducted business on the west of the Nyika National Park and into Zambia. In many ways this expedition was the pinnacle of logistical achievement.

    SOUTHERN TANZANIA 2013 by Rondi Salter and Phil Benstead Day One - 15/01/2013 - Dar es Salaam (Kawe) and Bongoyo Island Having left a cold and dreary UK behind the previous evening, we were welcomed by the sticky heat of Dar. After collecting our baggage we made our way to the Mediterraneo Hotel in Kawe. Phil met us at the hotel and we breakfasted on the seafront overlooking Msasani bay. Phil had arrived the night before and had already seen two species of fruit-bat and a few edible bullfrogs. Morning birds seen flitting around in the pretty gardens had included Zanzibar sombre greenbul, red-fronted tinkerbird and a golden oriole. The weather and the gods were kind to us in Dar this year. We got exposed mudflats in the morning and a little later on a trip out to Bongoyo Island just offshore. The exposed mudflats visible from the hotel producing our first views of crab plover, 8-9 reasonably approachable individuals. Also present were some common migrant waders and a single Terek sandpiper. We paddled through bath temperature water out to the speedboat and sped off to the island across a beautifully flat sea. After a round of the ever popular soda Tangawizi (ginger ale) we went for a wander through the islands coral rag thicket ending up at Shark Bay in hot (in both senses of the word) pursuit of a mangrove kingfisher. A mixture of Moray eels, plants such as Pseuderanthemum hildebrandtii (above left), coral rag skinks, butterflies and birds keep us entertained before retreating to the shade for freshly barbequed fish and chips.

    Southern Tanzania Flora & fauna of the Southern Highlands A Greentours Trip Report 8th – 23rd January 2019 Led by Phil Benstead & Peter Steiger 8/1 travel We all travelled by various routes. Six of us converging at Istanbul for the flight on to Dar, where Martin was already comfortably ensconced. The Istanbul team arrived in the early hours of the next day which was rather rude and for those applying for visas on arrival a little tedious. But eventually we were out and changing cash for the tour ahead. Our driver met us and there was a mercifully brief journey to the hotel. We were in bed by 0500 and most of is slipped immediately into a deep sleep. 9/1 early arrival in Dar and transfer to Kawe and Bongoyo Island It was very hard to get up this morning but thankfully we only had a relaxed morning of acclimatisation ahead of us. The heat and humidity were very different from Northern Europe... People drifted in to breakfast as and when and it was sometime after coffee that we realised that Peter and Andreas were probably not in the hotel. Sure enough reception had neglected to tell us that their flight had been cancelled and that they would instead be arriving mid-morning... We pottered around the grounds picking up some very beautiful birds despite the heat. Brown- breasted barbet was a nice surprise and we also had encounters with Zanzibar sombre greenbul, collared sunbirds and white-browed scrub-robin. Martin worked hardest and was rewarded with an amethyst sunbird.

The aim of the present Viola betonicifolia study was to investigate the presence of cyclotides in Sri Lankan Violaceae plants, using combined tools of Violaceae transcriptomics and mass spectrometry. New cyclotides were discovered for the firsttime in the wild floraof Sri Cyclotides Lanka, within Viola betonicifolia, a plant used in traditional medicine as an antimicrobial. Plant extracts prepared LC-MS in small scale from Viola betonicifolia were first subjected to LC-MS analysis. Subsequent transcriptome de novo Transcriptome de novo sequencing – Cyclisation enzyme sequencing of Viola betonicifolia uncovered 25 new (vibe 1 25) and three known (varv A/kalata S, viba 17, viba ¨ Protein disulfide isomerase 11) peptide sequences from Mobius and bracelet cyclotide subfamilies as well as hybrid cyclotides. Among the transcripts, putative linear acyclotide sequences (vibe 4, vibe 10, vibe 11 and vibe 22) that lack a conserved asparagine or aspartic acid vital for cyclisation were also present.

Craik* Institute for Molecular Bioscience, The University of Queensland, Brisbane, QLD 4072, Australia *Corresponding Author: David J Craik Email: [email protected] ABSTRACT This article reviews the class of plant-derived macrocyclic peptides called cyclotides. We include an account of their discovery, characterization and distribution in the plant kingdom, as well as a detailed analysis of their sequences and structures, biosynthesis and chemical synthesis, biological functions and their applications. These macrocyclic peptides are around 30 amino acids in size and are characterized by their head-to-tail cyclic backbone and cystine knot motif, which render them to be exceptionally stable, with resistance to thermal or enzymatic degradation. Routes to their chemical synthesis have been developed over the past two decades and this capability has facilitated a wide range of mutagenesis and structure‒activity relationship studies. In turn, these studies have led both to an increased understanding of their mechanisms of action as well as facilitating a range of applications in agriculture and medicine, as eco-friendly crop protection agents and as drug leads or scaffolds for pharmaceutical design. Our overall objective in this review is to provide readers with a comprehensive overview of cyclotides that we hope will stimulate further work on this fascinating family of peptides. CONTENTS 1.
