The antimicrobial protein producing Pediococcus spp. was isolated from selected diary samples Chemical preservative and characterised by standard microbiology and molecular biology protocols. The cell free supernatant of Microbiological quality Pediococcus spp. was applied on the selected food samples and monitored on daily basis. Antimicrobial potential Enhanced shelf life of the partially purified protein from this bacterium was tested against clinical isolates by well diffusion assay. Antimicrobial potential The preservation efficiency of bacteriocin producing isolate at various concentrations was tested against selected food samples and compared with two chemical preservatives such as sodium sulphite and sodium benzoate. The bacteriocin was partially purified and the microbiological qualities of the biopreservative treated food samples were assessed. The present study suggested that 100 μg/l of bacteriocin extract demonstrated antimicrobial potential against E. coli and Shigella spp. The treatment with the Pediococcus spp. showed enhanced preservation at 15 mL/kg of selected samples for a period of 15 days in comparison with sodium sulphite and sodium benzoate. -

ABSTRACT DAUGHTRY, KATHERYNE VIRGINIA. Phenotypic and Genotypic Characterization of Lactobacillus buchneri Strains Isolated from Spoiled Fermented Cucumber. (Under the direction of Dr. Rodolphe Barrangou and Dr. Suzanne D. Johanningsmeier). Lactobacillus buchneri is a facultative anaerobe and member of the lactic acid bacteria. L. buchneri has been isolated from various environments, but most commonly from decomposing plant material, such as silage and spoiled food products, including wine, beer, Swiss cheese, mayonnaise, and fermented cucumber. Recently, the metabolic pathway for the conversion of lactic acid to acetic acid and 1,2-propanediol was annotated in this species. Although this metabolic pathway is not common in most lactic acid bacteria, L. buchneri degrades lactate under various conditions. Lactic acid utilization in fermented cucumbers leads to a rise in pH, ultimately spoiling the product. In previous studies, strains of L. buchneri isolated from fermented cucumber spoiled displayed variation in colony morphologies. It was predicted the isolates were phenotypically and genotypically diverse, and that the abilities to degrade lactic acid may be strain specific. To examine this hypothesis, thirty-five L. buchneri cultures isolated from spoiled fermented cucumber and the type strain isolated from tomato pulp were characterized and unique strains were subjected to whole genome sequencing. Each isolate was genotypically and phenotypically characterized using 16S rDNA sequencing, DiversiLab® rep-PCR, colony morphology on MRS agar, carbohydrate profiling, growth rates in MRS media, and the ability to degrade lactic acid in a modified MRS medium. Great diversity in colony morphology revealed variations of color (ranging from opaque yellow to white), texture (brittle, viscous, or powdery), shape (umbonate, flat, circular, or irregular) and size (1 mm- 11mm). -

Sci. 48(2): 128·138 (April2013) Abstract Bacteria representing several genera were isolated from integument and alimentary tracts of live Asian longhorned beetle, Anaplophora glabripennis (Motschulsky), larvae and adults. Insects examined were from infested tree branches collected from sites in New York and Illinois. Staphylococcus sciuri (Kloos) was the most common isolate associated with adults, from 13 of 19 examined, whereas members of the Enterobacteriaceae dominated the isolations from larvae. Leclercia adecarboxylata (Leclerc), a putative pathogen of Colorado potato beetle, Leptinotarsa decemlineata (Say), was found in 12 of 371arvae examined. Several opportunistic human pathogens, including S. xylosus (Schleifer and Kloos), S. intermedius (Hajek), S. hominis (Kloos and Schleifer), Pantoea agglomerans (Ewing and Fife), Serratia proteamaculans (Paine and Stansfield) and Klebsiella oxytoca (Fiugge) also were isolated from both larvae and adults. One isolate, found in 1 adult and several larvae, was identified as Tsukamurella inchonensis (Yassin) also an opportunistic human pathogen and possibly of Korean origin .. We have no evi dence that any of the microorganisms isolated are pathogenic for the Asian longhorned beetle. The pest has many hardwood hosts, particularly maples (Acer spp.), and currently is established in isolated popula tions in at least 3 states- New York, NJ and Massachusetts (USDA-APHIS 201 0). -

The main issue in diced eggplant consists in the oxidation due to polyphenol oxidase (PPO, EC 1.14.18.1), an enzyme producing pigments responsible for the undesirable dark color which spoiled the product. The aim of this work was to study fresh or heat-treated (100 °C for 15 min) onion by-products (paste, juice and bagasse) in order to evaluate in vitro the PPO enzyme inhibition capacity in minimally processed eggplant cv. 'Birgah’. The heated onion juice was the most suitable anti-browning extract among all by-products tested, followed by pasteurised paste and fresh juice (54.2, 41.2 and 37.3 % reduction in PPO activity, respectively). Data suggested that the thermal treatment enhances the effects of the extracts probably due to a synergic action between some endogenous active compounds present in onion and Maillard reaction products obtained during heating. -
