Di bawah bimbingan ARIDA SUSILOWATI dan HENTI HENDALASTUTI RACHMAT. Raru (Cotylelobium melanoxylon Pierre) dan (Cotylelobium lanceolatum Craib) merupakan pohon yang tumbuh pada daerah beriklim tropis dan tumbuh tersebar di Pulau Sumatera dan Kalimantan. Pemanenan pohon yang tidak terkontrol telah menyebabkan penurunan populasi alami yang cepat menyebabkan potensi penurunan keanekaragaman genetik. Informasi terkait dengan genetika raru di Sumatera Utara masih sangat terbatas, termasuk metode ekstraksi DNA yang sesuai. Oleh karena itu, penelitian ini dilakukan untuk mendapatkan informasi tentang ekstraksi DNA menggunakan metode CTAB dan penanda referensi untuk studi genetik populasi. Ekstraksi DNA dilakukan melalui metode CTAB (Cetyl Trimetyl Ammonium Bromide). DNA hasil ekstraksi selanjutnya diamplifikasi dengan PCR menggunakan tiga penanda standar barkoding yaitu ITS, trnH-psbA dan trnL-trnF. Hasil penelitian menunjukkan bahwa metode CTAB menghasilkan kualitas DNA yang kurang baik dan pengenceran 10% memberikan hasil amplifikasi produk PCR terbaik. Primer trnL-trnF mampu mengamplifikasi DNA raru dengan suhu annealing 50ºC dan trnH- psbA 54ºC. Cotylelobium melanoxylon Pierre and Cotylelobium lanceolatum Craib or locally known as raru,is tropical tree species and commonly distributed on the Sumatera and Kalimantan Island. Uncontrolled harvesting of wild-grown trees has led to rapid decrease of their natural populations and causing potential decrease in genetic diversity. The information on genetic aspect on raru in North Sumatera still limited, including it extraction methods.Therefore, this research was conducted to get information about optimal DNA isolation using CTAB methods and reference marker for further used in genetic study. DNA extraction was conducted through CTAB (Cetyl Trimetyl Ammonium Bromide) method. The isolation product then amplified using three barcoding standard marker those were ITS, trnH-psbA and trnL-trnF. The result showed that CTAB method was not able to yield good quality DNA and 10% dilution produced the best amplification of PCR product. The trnL-trnF primer was able to amplified DNA of raru with 50ºC annealing temperature and trnH-psbA with 54ºC annealing temperature. Penulis merupakan anak tunggal dari pasangan Bapak Ridwan Gea dan Ibu Bakhtiar Gea. Pada Tahun 2009 penulis lulus dari SDN 070981 Fodo. Penulis kemudian melanjutkan studi ke SMP Negeri 1 Gunungsitoli dan lulus pada tahun 2012. Penulis lulus pada tahun 2015 dari MA Negeri Gunungsitoli. Pada tahun 2015, penulis diterima di Universitas Sumatera Utara (USU) melalui jalur Seleksi Nasional Masuk Perguruan Tinggi Negeri (SNMPTN) sebagai mahasiswa di Program Studi Kehutanan Fakultas Kehutanan, Universitas Sumatera Utara. Selain mengikuti perkuliahan, penulis menerima beasiswa BIDIKMISI sejak tahun 2015. Penulis juga aktif mengikuti kegiatan organisasi sebagai anggota divisi DANUS (Dana dan Usaha) dalam organisasi BKM Baytul Asyjar dan GORGA (Gerakan Observasi Rimbawan Giat Alam) dari tahun 2017-2018 di Fakultas Kehutanan, Universitas Sumatera Utara serta mengikuti kegiatan organisasi PMIN-Medan dan menjabat sebagai Sekretaris Bidang Pembinaan dan Pengembangan Anggota dari tahun 2017-2018. Penulis melakukan Praktik Pengenalan Ekosistem Hutan (P2EH) di Kawasan Hutan Dengan Tujuan Khusus (KHDTK) Pondok Buluh pada tahun 2016. Ganesan The position of Singapore on the Sunda continental shelf is a special one, with the principal island originally about 540 km2 in extent, together with some 60 smaller islands at the southern exit to the Malacca Strait and near the confluence of the South China Sea and Karimata Strait just west of Borneo. Geographically at the equatorial extremity of the Malay Peninsula, Singapore is separated from the Riau islands to its south (principally the Karimun Islands, Batam and Bintan) by the Singapore Strait which includes the deeply scoured 204-m ‘Singapore Deeps’, a likely subsidence basin resulting from tectonic movements (Bird et al., 2006). While this Strait may seem able to restrict the dispersal of some organisms with interglacial and post- Pleistocene high sea levels, it probably was not an effective dispersal barrier during episodes of lowest sea levels, such as during the Last Glacial Maximum (LGM) at 18 ka. Then, it must have been in the path of a key land bridge between mainland Southeast Asia farther north and likewise exposed links to Java and other areas to the south (Ho, 1960; Morley & Flenley, 1987; Heaney, 1991; Voris, 2000; Bird et al., 2005). By contrast, the Johor Strait that separates Singapore from south Peninsular Malaysia is just about 10 m deep and 600 m wide at its narrowest. The Sundaland region on the Sunda continental shelf has seen climate shifts since even before the Pleistocene, sometimes associated with tectonic events.

    Plant Formations in the Malayan BioProvince Peter Martin Rhind Malayan Mixed Lowland Dipterocarp Forest These forests occur from sea level to an altitude of about 750 m. In Endau-Rompin they can be broadly divided into two types. One is characterized by Dipterocarpus and Shorea, and mainly occurs at altitudes below 250 m, and is common, for example, in the valleys of the Endau River and its tributaries. The other is characterized by Dryobalanus aromatica, which occurs normally at altitudes below 300 m; it is common, for example, in the northeast and eastern areas of the Kinchin and Lamakoh valleys. Both these forests contain massive trees with canopies up to 30 m high and emergents reaching 45-50 m, and have many common trees. The main dipterocarps are Dipterocarpus baudi, D. concavus, D. costulatus, D. crinitus, D. gracilis, D. kerrii, Shorea acuminata, S. blumutensis, S. maxwelliana, S. ovalis, S. paevifolia, S. pauciflora, S. singkawang and the endemic S. bentongensis (Dipterocarpaceae). Dryobalanops aromatica is distributed gregareously in various river valleys while the endemic Hopea johorensis (Dipterocarpaceae), has a more patchy distribution but is often found in treefall gaps. Other large trees include Artocarpus lanceifolius, Coelostegia borneensis, Dialium platysepalum, Dillenia excelsa, Dyera costulata, Endospermum malaccensis, Koompassia malaccensis, Parkia speciosa, Pouteria malaccensis, Scaphium macropodum, Swintonia floribunda var. penangiana and the endemic Sarcotheca laxa var. sericea (Oxalidaceae). Second tier species include Agostistachys borneensis, Aporusa microstachya, Elateriospermum tapos, Pimelodendron griffithianum, Horsfieldia superba, Knema furfuracea, Myristica iners, Xerospermum noronhianum and the endemic Dillenia albiflos (Dilleniaceae) and Schoutenia furfuracea (Tiliaceae). Palms are also significant components with Oncospermum horridula being the most common species.

Distribution of dipterocarps species would be affected by some factors especially edaphic, climate, and altitude. In Indonesia the dipterocarps species distribution could be shown from islands groups, number of species and forest types. Based on the observation of herbarium collection in Herbarium Bogoriense the distribution of the most dipterocarps species was in the altitude of 0-500 m and 500-1000 m on the dipterocarps forest type.     Maintenance and conservation of Dipterocarp diversity in tropical forests _______________________________________________ Mohammad Nazrin B Abdul Malik A thesis submitted in partial fulfilment of the degree of Doctor of Philosophy Faculty of Science Department of Animal and Plant Sciences November 2019 1 i Thesis abstract Many theories and hypotheses have been developed to explain the maintenance of diversity in plant communities, particularly in hyperdiverse tropical forests. Maintenance of the composition and diversity of tropical forests is vital, especially species of high commercial value. I focus on the high value dipterocarp timber species of Malaysia and Borneo as these have been extensive logged owing to increased demands from global timber trade. In this thesis, I explore the drivers of diversity of this group, as well as the determinants of global abundance, conservation and timber value. The most widely supported hypothesis for explaining tropical diversity is the Janzen Connell hypothesis. I experimentally tested the key elements of this, namely density and distance dependence, in two dipterocarp species. The results showed that different species exhibited different density and distance dependence effects. To further test the strength of this hypothesis, I conducted a meta-analysis combining multiple studies across tropical and temperate study sites, and with many species tested. It revealed significant support for the Janzen- Connell predictions in terms of distance and density dependence. Using a phylogenetic comparative approach, I highlight how environmental adaptation affects dipterocarp distribution, and the relationships of plant traits with ecological factors and conservation status. This analysis showed that environmental and ecological factors are related to plant traits and highlights the need for dipterocarp conservation priorities.

They are both utility and amenity. Trees can evoke awe, mysticism, and reverence. Trees represent great public and private values. Trees most noticed and celebrated by people and communities are the one-tenth of one-percent of trees which approach the limits of their maximum size, reach, extent, and age. These singular, historic, culturally significant, and massive extreme trees become symbols and icons of life on Earth, and our role model in environmental stewardship and sustainability. What Is A Tree? Figure 1 is a conglomeration of definitions and concepts about trees from legal and word definitions in North America. For example, 20 percent of all definitions specifically state a tree is a plant. Concentrated in 63% of all descriptors for trees are four terms: plant, woody, single stem, and tall. If broad stem diameter, branching, and perennial growth habit concepts are added, 87% of all the descriptors are represented. At its most basic level, defining a tree is not species based, but is a structural definition. A tree is represented by a type of plant architecture recognizable by non-technical people. The most basic concepts for defining a tree are — a large, tall, woody, perennial plant with a single, unbranched, erect, self-supporting stem holding an elevated and distinct crown of branches, and which is greater than 10 feet in height and greater than 3 inches in diameter.
