Mikroskop elektron merupakan instrumen yang secara luas digunakan untuk melihat morfologi spesimen biologi dan material. Namun perlakuan untuk investigasi spesimen biologi tidak sama dengan material anorganik pada umumnya. Hal ini dikarenakan sensitivitas terhadap radiasi, kondisi vakum, adanya kandungan air, dan rendahnya kontras yang dihasilkan dari spesimen biologi. Sifat tersebut menjadi tantangan tersendiri dalam menjaga spesimen biologi agar tidak rusak maupun mengalami perubahan yang signifikan ketika dikarakterisasi dengan mikroskop elektron. Pada tulisan ini diurai mengenai teknik preparasi sebelum karakterisasi, teknik pelapisan dengan cairan ionik, dan prosedur memperoleh multiwarna untuk spesimen biologi dengan menggunakan instrumen berbasis mikroskop elektron. Microscope electron is an instrument that widely used to observe the morphology of biological specimens and materials. Nevertheless, the treatment for investigation of biological specimen is different than others inorganic materials in general. This because of the sensitivity towards radiation, vacuum condition, water containing, and low contrast of biological specimen. These properties became a challenge about how to develop technique that suitable to prevent the destruction or collapse of biological specimen. Here we discuss about some preparation techniques before characterization, coating technique with ionic liquid, and procedure to obtain multicolor graphic using microscope electron based instrument. Pendahuluan Mikroskop elektron adalah instrumen yang telah secara luas digunakan dalam mengobservasi material anorganik seperti logam, alloy, komposit maupun organik, seperti tumbuhan, sel, jaringan, virus, atau bakteri. Tidak seperti mikroskop optik pada umumnya, mikroskop ini menggunakan elektron, dalam menghasilkan pencitraan. Elektron dihasilkan dari pistol elektron dengan energi berkisar antara ~500 eV dan 30 keV [1]. Semua mikroskop elektron membutuhkan kondisi vakum baik untuk kepentingan pergerakan elektron maupun meminimalisir hamburan lain dari sampel. Prototipe dari mikroskop elektron pertama kali dibuat oleh Ernst Ruska and Max Knoll pada tahun 1931, yang dapat melihat objek dengan perbesaran hingga seratus kali [2].
