In this study, natural attenuation and biostimulation of PCBs and microbial communities responsible for PCB transformations were investigated in Ohio River sediments. Natural attenuation of PCBs was negligible in sediments, which was likely attributed to low temperature conditions during most of the year, as well as low amounts of available nitrogen, phosphorus, and organic carbon.

MOORE' AND PETER HIRSCH2 Department of Microbiology and Public Health, Michigan State University, East Lansing, Michigan 48823 Received for publication 21 December 1971 The genetic relatedness of a number of budding and prosthecate bacteria was determined by deoxyribonucleic acid (DNA) homology experiments of the di- rect binding type. Strains of Hyphomicrobium sp. isolated from aquatic habi- tats were found to have relatedness values ranging from 9 to 70% with strain "EA-617," a subculture of the Hyphomicrobium isolated by Mevius from river water. Strains obtained from soil enrichments had lower values with EA-617, ranging from 3 to 5%. Very little or no homology was detected between the amino acid-utilizing strain Hyphomicrobium neptunium and other Hyphomi- crobium strains, although significant homology was observed with the two Hyphomonas strains examined. No homology could be detected between pros- thecate bacteria of the genera Rhodomicrobium, Prosthecomicrobium, Ancal- omicrobium, or Caulobacter, and Hyphomicrobium strain EA-617 or H. nep- tunium LE-670. The grouping of Hyphomicrobium strains by their relatedness values agrees well with a grouping according to the base composition of their DNA species. It is concluded that bacteria possessing cellular extensions repre- sent a widely diverse group of organisms. The 16S rRNA gene sequence analysis revealed that strain SA-279T formed a distinct lineage within the family Ancalomicrobiaceae and shared the highest pairwise similarity values with Pinisolibacterravus E9T (96.4 %) and Ancalomicrobiumadetum NBRC 102456T (94.2 %). Cells of strain SA-279T were rod-shaped, motile, oxidase and catalase positive, and capable of forming rosettes. Its predominant fatty acids were C18 : 1!7c (69.0 %) and C16 : 1!7c (22.7 %), the major respiratory quinone was Q-10, and the main polar lipids were phosphatidylethanolamine, phosphatidylmonomethylethanolamine, phosphatidylcholine, phosphatidylglycerol, an unidentified aminophospholipid and an unidentified lipid. The G+C content of the genomic DNA of strain SA-279T was 69.2 mol%. On the basis of the phenotypic, chemotaxonomic and molecular data, strain SA-279T is considered to represent a new genus and species within the family Ancalomicrobiaceae, for which the name Siculibacillus lacustris gen. nov., sp. nov. is proposed. The type strain is SA-279T (=DSM 29840T=JCM 31761T). The order Rhizobiales (class Alphaproteobacteria) currently another new strain, SA-279T, was characterized in detail, contains more than 15 families, such as ‘Aurantimonada- which was isolated from the same locality.

Up to date, only few reports have described the microbiological aspects behind biovermiculation geomicrobiology of Italian sulfuric acid caves despite their overall abundance. Here, we present the frst characterization of biovermiculation microbial populations from the Santa Cesarea Terme (Apulia, Italy) using next-generation sequencing and feld emission scanning electron microscopy (FESEM) approaches. We focused our analysis on biovermiculations from Fetida Cave located along the Adriatic Sea coastline. Tis cave is at sea level, and moving from the entrance to its inner part, it is possible to observe a decrease of marine infuence accompanied by a corresponding increase in the acidic efect of the upwelling waters.

    Studying new eukaryotic niches of bacterial life Marine Biological Laboratory Microbial Diversity Course 2005 Torben Stührmann Max Planck Institute for Marine Microbiology Department of Molecular Ecology Celsisus Str. 1 28379 Bremen-Germany Abstract Biologists are becoming increasingly aware that a variety of animals and plants bears symbiotic microorganisms. The formation of alliances between animals and microorganisms contribute to their nutrition or defence against predators and parasites. This study has the aim to search for new eukaryotic niches of bacterial live (symbiosis or commensalisms). As study object two different eukaryotic species from varying environments were taken. For the first study a brown insect larva was obtained from the Sippewissett Salt marshes which was not further characterized. The second study object was the jellyfish Cyanea capillata (the Lion’s Mane). This species was highly abundant in the Cape Cod area during the course period (June-August 2005). Symbiotic interactions between the cnidarians and Cyanobacteria are known but there is nothing known about bacterial interactions among the Scyphozoa. Microscopic- (Scanning electron microscopy, Light microscopy), cultivation- and molecular- techniques (16S rRNA clone libraries, Fluorescence in situ hybridization FISH) were applied for studying bacterial-eukaryotic interactions. Only few indications were found with FISH suggesting a eukaryotic niche for bacteria within the Scyphozoa. A rod shaped morphotype detected with a FISH-probe specific for Gammaproteobacteria was detected in the mouth tissue. Other approaches were not applied or failed. The 16S rRNA library of brown larvae revealed a high predominance of Gammaproteobacteria (Thiomicrospira, Halomonas, Marinobacter and Vibrio spp.). Epsilon-, alpha- proteobacteria, Bacteriodetes and Cyanobacteria were only less frequent.

With it´s numerous ecological niches such as mud-volcanoes, cold seeps and hot and low-temperature influenced vents it could provide a harbourage for a diverse microbial consortia. By several molecular and microbial approaches, basalt and sediment samples collected in the vicinity of low- temperature influenced diffuse vents along the Mid-Atlantic-Ridge, were investigated. Our study revealed that a diverse microbial population inhabiting the analysed rock samples, dominated by Proteobacteria . The isolated organisms are distinguishable from the overlaying deep sea water and by characterisation of selected isolates high tolerances against a broad range of temperatures, pH-values and salt concentrations were observed.

This investigation confirms that the genus is polyphyletic based on an extensive analysis of the 16S rRNA gene sequences of several named species of the genus. The analyses indicate that some Prosthecomicrobium species are more closely related to non-prosthecate genera, including Devosia, Labrenzia, Blastochloris, Methylosinus, Mesorhizobium and Kaistia, than they are to other species of the genus Prosthecomicrobium. For this reason, two of the Prosthecomicrobium clades which are polyphyletic with the type species, Prosthecomicrobium pneumaticum, are renamed as new genera. The currently named species Prosthecomicrobium enhydrum, Prosthecomicrobium mishustinii, Prosthecomicrobium consociatum and Prosthecomicrobium litoralum are reclassified in two new genera, Vasilyevaea gen. nov. and Bauldia gen. nov. with four new combinations: Vasilyevaea enhydra comb. nov. (the type species) and Vasilyevaea mishustinii comb.

These strains were compared genotypically by DNA/DNA reassociation and 16S rDNA based phylogenetic analyses. Genotypic comparisons were complemented with phenotypic characterizations. Together, these studies clearly indicate each Prosthecobacter strain represents a novel species of bacteria. We propose three new species of Prosthecobacter, P. dejongeii strain FC1, P. vanneervenii strain FC2, and P. debontii strain FC3; P. fusiformis is reserved for the type strain of the genus, strain FC4. Additionally, we propose the genera Prosthecobacter and Verrucomicrobium, currently members of the order Verrucomicrobiales, to comprise a novel higher order taxonomic group, the division Verrucomicrobia div. nov. and the class Verrumicrobiae class nov.
