Economically, apple is the fourth most important fruit crop after citrus, grapes and banana. Apples are consumed fresh, directly after harvest or after a storage period for up to 6 months or even longer. Apples can also be processed to produce, e.g., juice, sauce, slices, vinegar and cider. Most of the cultivated apples belong to the species Malus × domestica Borkh. in the Rosaceae family. More than 7500 apple cultivars have been described from different countries. However, only a few of them have sufficient quality and productivity. Many cultivars are limited by different diseases that reduce the apple quality and market acceptability. Research attempts have recently been focused specifically on some traits which are economically very important, e.g. disease tolerance, fruit texture and quality. This introductory paper forms part of a PhD study that aims to quantify the storage disease tolerance of some apple cultivars by performing inoculation tests with fungal spores on harvested fruits.

  • SSR-Based Genetic Mapping of Qtls Determining Chilling Requirements
SSR-based genetic mapping of QTLs determining chilling requirements for time of initial vegetative budbreak in domesticated apple (Malus x domestica Borkh.) cultivar ‘Anna’ x ‘Austin’. by Paidashe Hove A thesis submitted in partial fulfilment of the requirements for the degree of MAGISTER SCIENTIAE (M.Sc.) Department of Biotechnology University of the Western Cape Bellville Supervisor: Dr. D. J. G. Rees Co-supervisor: Prof. B. Ndimba June 2012 ABSTRACT The Rosaceae family contains major temperate crops such as the domesticated apple (Malus x domestica Borkh.), peach (Prunus persica L. Batsch) and European pear (Pyrus communis L.). However, despite its evident economic importance, it is generally poorly studied in genomic terms, relative to the other major crop groups. Microsatellite and Diversity Array Technology (DArT) genetic markers have been exploited in this work and are essential tools in genetic map construction and marker-assisted selection (MAS) of high quality apples and other rosaceous crops. Microsatellites are advantageous in that they are co-dominant, highly polymorphic, abundant, transferable and reliably reproducible; hence their use in this study. In order for budbreak to take place in a timely and homogenous fashion, apple trees need a period of exposure to low temperatures. Within orchards the application of chemicals that induce budbreak in unsuitable environments is required to produce apples from cultivars that require high chilling levels. However, this and other practices using chemicals in orchards tend to pollute the environment. One of the solutions to this problem is to breed low chill apples such as ‘Anna’ cultivar, which was used as one of the parents in this study.

  • ABSTRACT JOHNSON, KENDALL A. Characterization and Fungicide
ABSTRACT JOHNSON, KENDALL A. Characterization and Fungicide Efficacy of North Carolina Colletotrichum Populations Causing Glomerella Leaf Spot and Fruit Rot on Apple. (Under the direction of Dr. Sara Villani). Glomerella leaf spot (GLS) and fruit rot (GFR) is historically caused by members of the Colletotrichum gloeosporioides species complex. Although the disease was first reported in the 1970s in the United States, the disease has reemerged on apples in North Carolina (NC) causing economically devastating losses. These pathogens cause necrotic leaf lesions and rots of fruit that will ultimately lead to premature defoliation, a decrease in yield, and unsaleable fruit. According to previous research, ‘Red Delicious’ and ‘Fuji’ have shown partial or complete resistance to GLS and GFR. Cultivars from the ‘Golden Delicious’ lineage are the most susceptible which includes cvs. ‘Golden Delicious’, ‘Gala’, and ‘Pink Lady’ which are also amongst consumer favorites and most commonly grown in NC. Currently there is a paucity of information regarding the management of the disease, but fungicides, cultural practices, and the planting of resistant cultivars are primarily used to reduce disease within an orchard. Fungicide use is limited due to the risk of resistance, but has found to be the most effective form of control for this disease. The objectives of this research were to 1) characterize Colletotrichum species based on morphological and molecular techniques and evaluate in vitro QoI fungicide sensitivity shifts in Colletotrichum populations and 2) determine the most efficacious fungicide and timing for the management of GLS and GFR. Three hundred ninety-six isolates were collected from symptomatic apple leaves and fruit from a total of 17 orchards in North Carolina and Georgia in 2016.
