The early detection of the compatibility of the aforementioned scions and rootstocks was being carried out by fusing the calluses of the scion and rootstock in vitro: The unions were obtained by visual and histochemical union analyses (the cell arrangement and the cell wall staining as well as the presence of lipids and phenol components in the callus). Како се већина добија путем хетеровегетативног размножавања, поље истраживања је компатибилност најчешће гајених култивара украсних трешања на нашем поднебљу: Prunus serrulata ʼAmanogawaʼ, Prunus serrulata ʼKanzanʼ, Prunus serrulata ʼKiku-shidare-zakuraʼ, са домаћим подлогама за трешању: Prunus avium, Prunus ʼColtʼ и Prunus mahaleb. Таксон који је коришћен у истраживању као позитивна (компатибилна) контрола је Prunus serrulata, а два потенцијално инкомпатибилна таксона - Prunus serotina и Pyrus communis ʼPyrodwarfʼ. Фузијом калуса подлоге и племке у условима in vitro, на релативно брз и поуздан начин вршена је рана детекција компатибилности поменутих племки и подлога. Резултати о јачини и компатибилности спојева добијени су визуелним (проценат успешно спојених, степен јачине споја) и хистохемијским анализама споја (распоред и обојеност ћелијских зидова и присуство липида и фенолних компоненти у калусу). Од три коришћене хранљиве подлоге (MS, SH и WP), за највећи број таксона, најпогоднија у процесу индукције калуса се показала МS. За обичну трешњу и култиваре Prunus serrulata ʼKanzanʼ и Prunus ʼColtʼ, у процесу индукције калуса из пупољака или вегетативних делова летораста, одговарајућа је била и SH подлога, док се једино за врсту Prunus mahaleb, WP издвојила као погодна хранљива подлога. Са ауксином NAA у концентрацији 2 mgL-1 и цитокинином BAP 0,5 mgL-1 за већину таксона добијени су најбољи резулатати у калогенези. За генотипове Prunus avium, Prunus ʼColtʼ, Prunus serotina и Prunus serrulata ʼKanzanʼ, могуће је користити у истој концентрацији и 2,4-D као замену, док се IBA показао неодговарајућим ауксином за индукцију калуса код истраживаних врста. На спојевима делова добијеног калуса подлоге и племке, визуелно оцењена јачина споја показала је јаке спојеве код комбинација: P. avium/ P. serrulata ʼKanzanʼ, Prunus ʼColtʼ/P. serrulata ʼAmanogawaʼ, Prunus ʼColtʼ/P. serrulata ʼKiku-shidare-zakuraʼ и P. serrulata са сва три култивара. Делимично јаки спојеви остварени су код калуса P. avium/P. serrulata ʼAmanogawaʼ и ʼKiku-shidare-zakuraʼ, Prunus ʼColtʼ/ P. serrulata ʼKanzanʼ и код спојева сва три култивара са касном сремзом (Prunus serotina). Јасне знаке неподударности калуса кроз лако одвајање на споју, показале су, очекивано, комбинације сва три култивара јапанских украсних трешања са култиваром Pyrus communis ʼPyrodwarfʼ и са магривом (Prunus mahaleb). i Анатомске анализе су указале на значајне манифестације у ткиву спојева калуса: бојењем рутенијум црвеном бојом локализоване су веће количине пектинских полисахарида код потенцијално компатибилних спојева, насупрот мањој локализацији ових једињења код инкомпатибилних. Бојењем толуидин и алцијан плавом и сафранином, доста прецизно су локализовани феноли, као и сенесценција ткива, што може да укаже на ране знаке неподударности калуса. Поменуте појаве јасно су забележене код спојева култивара P. serrulata са магривом и патуљастом крушком (ʼPyrodwarfʼ) и местимично на спојевима са култиваром ʼColtʼ и са обичном трешњом (P. avium/P. serrulata ʼAmanogawaʼ и P. avium/P. serrulata ʼKiku-shidare-zakuraʼ) и код спојева са касном сремзом. Хемијским анализама спојева, потврђена је компатибилност из претходне две анализе као и постојање одређеног броја неподударних спојева. Детекција неких полифенола који су показатељи инкопмпатибилности калемљења (p - кумаринска киселина, кемферол, кверцетин, ферулинска киселина, нарингенин и баикалеин), потврдила је на још један, посредан начин, потенцијално инкомпатибилне спојеве. Код магриве, хемијским анализама укупних и појединачних полифенола потврђено је њихово нагомилавање јасно манифестовано потамњивањем ткива, а на спојевима са обичном трешњом (која се код нас најчешће користи као подлога за калемљење јапанских украсних трешања), хемијске анализе су потврдиле постојање одређених сметњи у споју ове врсте са култиварима ʼAmanogawaʼ и ʼKiku-shidare-zakuraʼ. Према свим анализама за ова два култивара боље резултате су дале подлога Prunus serrulata и Prunus ʼColtʼ. Основни таксон Prunus serrulata могао би се користити као алтернативна подлога и/или посредник калемљења између наших подлога и јапанских украсних трешања, а Prunus ʼColtʼ као алтернатива за домаћу трешњу као подлогу, чиме би се превазишла неподударност између поменутих компонената калемљења. Сумирањем резултата спроведених истраживања, може се констатовати да се примењена метода фузије калуса у in vitro условима може сматрати ефикасним начином добијања релатвно брзог одговора о потенцијалној подударности две компоненте калемљења и да је метода применљива и за друге таксоне. Применом ове технике могли би се, макар у одређеном обиму, избећи пољски огледи у којима се одговор о подударности калемљења може чекати више година. We have previously reported that polymerization of a natural extracellular substance (ECS) on the outer surface of animals by electron beam or plasma irradiation, can give rise to a nano-scale layer, termed the “NanoSuit”, which can keep small animals alive under the high vacuum of a scanning electron microscope (SEM). In the present research, we have focused on plants, using petals of cherry blossoms, as experimental specimens and examined their behavior under high vacuum conditions. Experiments on healthy living petals have demonstrated that without any pre- treatment, the overall morphology of specimens is well preserved and intact after imaging in an SEM, suggesting that natural substances on the petal surface behave like animal ECS and form a NanoSuit following irradiation with an electron beam. Furthermore, we have shown that the surface material can be extracted with chloroform and polymerized into a free-standing membrane by plasma irradiation. From our results, we conclude that surface materials, which have the ability to prevent water loss under natural conditions, increase the barrier ability and can protect plants under high vacuum conditions.
