Sample Analysing meat the problem of separating inosine and hypoxanthine pretreatment: in the presence of different mono nucleotides occurs. With the column material NUCLEOSIL 100-5 C18 these compounds can easily be quantified. The retention time of hypoxanthine and inosine monophosphate depends on how long the column is regenerated. A regeneration time of 13-15 min. yields in good separations whereas regeneration times longer than 25 min. leads to overlapping of the peaks of hypoxanthine and UMP.

Conditions: Eluents: A) 0.1 M KH2PO4, 4 mM tetrabutylammonium dihydrogen , pH 6.0 B) 50% A plus 50% methanol Gradient: 0-6 min. 15% B, 6-16 min. from 15% to 90% B, 16-17 min. 90% B, 17-17.5 min. to 15% B, 17.5-31.5 min. 15% B, 32 min.: start of next run Flow rate: 0.5 mL/min
