Pembuatan preparat tulang rangka ikan dilakukan secara fisik dan kimiawi. Tahapan fisik diawali Sisik ikan dihilangkan dengan menggunakan pisau atau pinset otot di bedah, pembedahan otot ikan dilakukan sampai tulang-tulang pada ikan nampak baik, Tahapan kimiawi diawali dengan merendam preparat tulang rangka kedalam formalin 10% selama tujuh hari. Selanjutnya dilakukan perendaman dalam larutan etanol 100% selama 24 jam. Kata Kunci: Komperatif, Osteocranium. This sh is part of an important group of freshwater cyprinids collectively known as mahseers that inhabit mountainous rivers and lakes of the Himalayan belt from Afghanistan to Indonesina and Myanmar. Mahseers form a signicant stock of indigenous sh in India, Nepal, Bangladesh and Pakistan with potential for the freshwater aquaculture industry. Malaysia’s three species of mahseer – T. tambroides, T. douronensis and T. tambra – live in headwaters and the fast- owing, cool and clear water of upland rivers. As with mahseers in other countries, the natural stock of Malaysian mahseers has rapidly declined in recent years due to environmental changes, human disturbance of aquatic ecosystems and overshing. Following the decline in populations of this species in the wild, its market prices rose as high as $80 and $240/kg when sold as food or ornamental sh, respectively.

However, the phylogeny proposed novel taxonomic findings within Capoeta. Results of GMYC, bPTP, and phylogenetic analyses were similar and suggested that species diversity in Iran is currently underestimated. At least four can- didate species, Capoeta sp4, Capoeta sp5, Capoeta sp6, and Capoeta sp7, are awaiting description. Capoeta capoeta comprises a species complex with distinct genetic line- ages. The divergence times of the three main Capoeta clades are estimated to have occurred around 15.6–12.4 Mya, consistent with a Mio- Pleistocene origin of the di- versity of Capoeta in Iran. The changes in Caspian Sea levels associated with climate fluctuations and geomorphological events such as the uplift of the Zagros and Alborz Mountains may account for the complex speciation patterns in Capoeta in Iran.

The present investigation provides data on genetic structure of some species of Nemachilidae including Paraschistura bampurensis , Oxynoemacheilus kiabii and Turcinemacheilus saadii from inland waters of Iran. These species were sequenced for a 652 base pair region of the mitochondrial cytochrome oxidase subunit I gene. All species were identified by cytochrome oxidase subunit I gene sequence. The sequences of P. bampurensis showed that specimens collected from two streams (Shapour and Fahlian, south-west of Iran), with 1.89% within-species Kimura two parameter distance, shared haplotype in neighbor joining tree.

Geiger2 1) Department of Biology, College of Sciences, Shiraz University, Shiraz, Iran 2) Zoological Research Museum Alexander Koenig, Leibniz Institute for Animal Biodiversity, Adenauerallee, Germany ABSTRACT This study provides new data on chromosomal characteristics and DNA barcoding of three endemic loaches of Iran: spiny southern loach Cobitis linea (Heckel, 1847), Persian stream loach Oxynoemacheilus persa (Heckel, 1848) and Tongiorgi stream loach Oxynoemacheilus tongiorgii (Nalbant & Bianco, 1998). The chromosomes of these fishes were investigated by examining metaphase chromosome spreads obtained from epithelial gill and kidney cells. The diploid chromosome numbers of all three species were 2n=50. The karyotypes of C. linea consisted of 4M + 40SM + 6ST, NF=94; of O. persa by 20M + 22SM + 8ST, NF=90 and of O. tongiorgii by 18M + 24SM + 8ST, NF= 92. Sex chromosomes were cytologically indistinguishable in these loaches. Maximum likelihood-based estimation of the phylogenetic relationships based on the COI barcode region clearly separates the three Iranian loach species of the Kor River basin. All species distinguished by morphological characters were recovered as monophyletic clades by the COI barcodes. Corresponding author: *E-mail: [email protected] Abstract The phylogenetic relationships of Oxynoemacheilus bergianus from three Iranian inland waters, i.e. the Caspian Sea, Namak and Urmia Lake basins were investigated using osteological characters. 21 osteological characters were used and Garra rufa was the out-groups. A phylogenetic analysis based on the described characters resulted four most-parsimonious cladograms (36 steps, CI=0.69, RI=0.5). The strict consensus tree showed that the populations of O. bergianus are monophyletic group. It was confirmed that the populations of Qareh- chai, Qezel-ozan and Sefid rivers form a sister group to other examined populations of O. bergianus.
