Detailed analysis of NR and resin contents revealed different concentrations of various pentacyclic triterpenes such as α-, β-amyrin and taraxasterol, which strongly affect the mechanical properties of the resulting rubber material. Therefore, the reduction of triterpene content would certainly improve the industrial applications of dandelion NR. We developed T. koksaghyz plants with reduced triterpene contents by tissue-specific downregulation of major laticifer-specific oxidosqualene cyclases (OSCs) by RNA interference, resulting in an almost 67% reduction in the triterpene content of NR. Plants of the T1 generation showed no obvious phenotypic changes and the rubber yield also remained unaffected. Hence, this study will provide a solid basis for subsequent modern breeding programs to develop Russian dandelion plants with low and stable triterpene levels. Introduction In Russian dandelion (Taraxacum koksaghyz), high concentrations of secondary metabolites such as natural rubber (NR) are found mainly in the latex of root laticifers [1,2]. NR is an important industrial biopolymer that is used for the production of more than 40,000 consumables, including tires and medical devices [3,4]. To date, the main source for high-quality NR is the rubber tree Hevea brasiliensis, but increased raw material demand has led to the development of Russian dandelion as an alternative and sustainable rubber source. T. koksaghyz is a self-incompatible, sexually reproductive plant. Consequently, the resulting offspring is heterogenic [4,5], leading to trait divergence among individual plants. One such example is the highly variable triterpene content of dandelion-derived NK, which negatively affects NR properties [6,7]. For this reason, genetic engineering of the corresponding triterpene biosynthesis could support the development of more uniform accessions with low and stable triterpene contents in their roots. The central isopentenyl diphosphate (IPP) building block for triterpenes as well as for poly(cis-1,4-isoprenes) such as dolichols or the long-chain NR-forming poly(cis-1,4-isoprene) is synthesized by the mevalonate (MVA) pathway in the latex of laticifers (Figure1)[8,9]. These compounds are extremely common and are found in most plants. There are at least 4000 known triterpenes. Many triterpenes occur freely but others occur as glycosides (saponins) or in special combined forms. Pentacyclic triterpenes have a wide spectrum of biological activities and some of them may be useful in medicine. The therapeutic potential of three pentacyclic triterpenes – lupeol, betuline and betulinic acid – is discussed in this paper. Betulinic acid especially is a very promising compound. This terpene seems to act by inducing apoptosis in cancer cells. Due to its apparent specificity for melanoma cells, betulinic acid seems to be a more promising anti-cancer substance than drugs like taxol. Saponin was administered as strawberry suspension at a dose of 10mg per day (divided into four doses) to ten rabbits, nine of which were fed with food contaminated with 0.5mL bacterial suspension obtained by McFarland standardization (10% Barium sulfate) after starvation for 6hrs . Multiple foci of tubular necrosis and haemorrhages in the kidney, marked hyperplasia of the mucosal layer of the small intestine, and a mild periportal lymphocytic cellular infiltration of the liver of the treated rabbits were observed. Kalunke2 Asparagus racemosus (Shatavari), belongs to the family Asparagaceae and is known as a “curer of hundred diseases” since ancient time. This plant has been exploited as a food supplement to enhance immune system and regarded as a highly valued medicinal plant in Ayurvedic medicine system for the treatment of various ailments such as gastric ulcers, dyspepsia, cardiovascular diseases, neurodegenerative diseases, cancer, as a galactogogue and against several other diseases. In depth metabolic fngerprinting of various parts of the plant led to the identifcation of 13 monoterpenoids exclusively present in roots. LC-MS profling led to the identifcation of a signifcant number of steroidal saponins (33). However, we have also identifed 16 triterpene saponins for the frst time in A. racemosus. In order to understand the molecular basis of biosynthesis of major components, transcriptome sequencing from three diferent tissues (root, leaf and fruit) was carried out. Functional annotation of A. racemosus transcriptome resulted in the identifcation of 153 transcripts involved in steroidal saponin biosynthesis, 45 transcripts in triterpene saponin biosynthesis, 44 transcripts in monoterpenoid biosynthesis and 79 transcripts in favonoid biosynthesis. These fndings will pave the way for better understanding of the molecular basis of steroidal saponin, triterpene saponin, monoterpenoids and favonoid biosynthesis in A. racemosus. In this research, we intended to study the metabolomics of Camellia japonica leaf by using gas chromatography-mass spectroscopy technique. Results: A total of twenty-nine anti-inflammatory compounds, occupying 83.96% of total area, came out in the result. Most of the metabolites are terpenoids leading with triterpenoids like squalene, lupeol, and vitamin E. In this study, the candidate molecules responsible for anti-inflammatory activity were spotted out in the leaf extract and biosynthetic relation or interactions between those components were also established. Conclusion: Finding novel anticancer and anti-inflammatory medicinal compounds like lupeol in a large amount in Camellia japonica leaf is the most remarkable outcome of this gas chromatography-mass spectroscopy analysis. Developing probable pathway for biosynthesis of methyl commate B is also noteworthy. The complete coding sequences of these 2 genes were cloned from leaf mRNA, and their functions were characterized by heterologous expression in yeast. The cycloartenol synthase sequence is very similar to other known OSCs of this type (approximately 80% identity), although the M. ilicifolia friedelin synthase amino acid sequence is more related to β-amyrin synthases (65–74% identity), which is similar to the friedelin synthase cloned from Kalanchoe daigremontiana. Multiple sequence alignments demonstrated the presence of a leucine residue two positions upstream of the friedelin synthase Asp-Cys-Thr-Ala-Glu (DCTAE) active site motif, while the vast majority of OSCs identified so far have a valine or isoleucine residue at the same position. The substitution of the leucine residue with valine, threonine or isoleucine in M. ilicifolia friedelin synthase interfered with substrate recognition and lead to the production of different pentacyclic triterpenes.
