At least four can- didate species, Capoeta sp4, Capoeta sp5, Capoeta sp6, and Capoeta sp7, are awaiting description. Capoeta capoeta comprises a species complex with distinct genetic line- ages. The divergence times of the three main Capoeta clades are estimated to have occurred around 15.6–12.4 Mya, consistent with a Mio- Pleistocene origin of the di- versity of Capoeta in Iran. The changes in Caspian Sea levels associated with climate fluctuations and geomorphological events such as the uplift of the Zagros and Alborz Mountains may account for the complex speciation patterns in Capoeta in Iran. -

Dr. Ramazan DEMİRDAĞ Bu tez kapsamında, Siraz ( Capoeta umbla) karaciğer dokusunda karbonik anhidraz (CA, karbonat hidroliyaz, E.C. 4.2.1.1) enzimi üzerine bazı metallerin ve pestisitlerin in vitro etkileri incelendi. Siraz (C.umbla) balığının karaciğer dokusundan karbonik anhidraz enzimi ilk kez tarafımızdan afinite kromatografisi (Sepharose 4B–L–tirozin– sulfanilamid) yöntemi ile saflaştırıldı. Afinite kromatografisi neticesinde enzim 2026.32 EÜ/mg spesifik aktiviteyle ve % 59.97 verimle 212.40 kat saflaştırıldı. Saflaştırılan enzimin saflığını kontrol etmek amacıyla Sodyum dodesil sülfat- poliakrilamid jel (SDS-PAGE) elektroforezi yapıldı ve tek bant elde edildi. Siraz ( C.umbla) karaciğer dokusu CA enzimi için yapılan kinetik çalışmalarda; p- nitrofenil asetat substratı kullanılarak Km ve Vmax Lineweaver-Burk grafiklerinden hesaplandı. Tris-SO4 tamponunda optimum pH 8.5, optimum iyonik şiddet 0,02 M Tris-SO4, optimum sıcaklık 35ºC olarak, stabil pH ise Tris-SO4 tamponunda pH 7.0 olarak bulundu. -

Örnekleme, Melet Irmağı’nın üzerinde yapımı planla- nan Ordu Barajı ve HES için önerilen mevkiiden Karadeniz sahiline kadar olan mansap kıs- mında yapılmıştır. Bu araştırmada, 3 familyaya ait 7 tür tespit edilmiştir. Bu türlerden Barbus tauricus (Kessler, 1877) (% 50) baskın tür olup, bunu sırasıyla Vimba vimba (Linnaeus, 1758) (% 14), Capoeta banarescui (Turan, Kottelat, Ekmekçi ve İmamoğlu, 2006) (% 14) Neogobius kessleri (Günther, 1861) (% 9), Neogobius fluviatilis (Pallas, 1814) (% 8), Leuciscus cephalus (Linnaeus, 1758) (% 4) ve Mugil cephalus (Linnaeus, 1758) (% 1) türleri takip etmektedir. Bu türlerden Capoeta banarescui ve Neogobius kessleri Orta Karadeniz Bölgesi için yeni kayıttır. Bu etkiler, bazen balık miktarının azalması, bazen de bir türün hiç avlanamaması ya da neslinin tükenmesi şeklinde olabilmektedir. Nehir üzerindeki bu tür engeller nehir sürekliliğini de etkilemektedir. Bir nehir sisteminin doğrusal ve yatay sürekliliği ekolojik bakımından hayati önemi haizdir. Çeşitli maksatlarla hidrolik yapıların inşasının zorunlu olduğu durumlarda, ekosisteme verilen zararın asgarî düzeye indirilmesi için balık geçitleri inşa edilmelidir. Balık popülasyonları, bütün biyolojik işlevlerini destekleyen sucul yaşam alanının özelliklerine doğrudan bağımlıdır. Göçmen balıklar, üreme, yavru balıkların büyümesi ve cinsel olgunlaşma gibi hayatlarının belirli dönemlerinde farklı ortamlara ihtiyaç duymaktadır.
