EvoDevo (2016) 7:16 DOI 10.1186/s13227-016-0054-6 EvoDevo RESEARCH Open Access The developmental and genetic bases of apetaly in Bocconia frutescens (Chelidonieae: Papaveraceae) Cristina Arango‑Ocampo1, Favio González2, Juan Fernando Alzate3 and Natalia Pabón‑Mora1* Abstract Background: Bocconia and Macleaya are the only genera of the poppy family (Papaveraceae) lacking petals; how‑ ever, the developmental and genetic processes underlying such evolutionary shift have not yet been studied. Results: We studied floral development in two species of petal-less poppies Bocconia frutescens and Macleaya cordata as well as in the closely related petal-bearing Stylophorum diphyllum. We generated a floral transcriptome of B. frutescens to identify MADS-box ABCE floral organ identity genes expressed during early floral development. We performed phylogenetic analyses of these genes across Ranunculales as well as RT-PCR and qRT-PCR to assess loci- specific expression patterns. We found that petal-to-stamen homeosis in petal-less poppies occurs through distinct developmental pathways. Transcriptomic analyses of B. frutescens floral buds showed that homologs of all MADS-box genes are expressed except for the APETALA3-3 ortholog. Species-specific duplications of other ABCE genes inB. frute- scens have resulted in functional copies with expanded expression patterns than those predicted by the model. Conclusions: Petal loss in B. frutescens is likely associated with the lack of expression of AP3-3 and an expanded expression of AGAMOUS. The genetic basis of petal identity is conserved in Ranunculaceae and Papaveraceae although they have different number of AP3 paralogs and exhibit dissimilar floral groundplans. -

On the research Front, a new weevil (Rhinocyllus conicus) introduced as a bio-control For weed thistles was discovered impacting the already threatened Pitcher’s thistle (Cirsium pitcheri). Garden scientists have been working For over 15 years to recover this important dune species. In Colorado, experiments were set up to test new post-wildFire restoration methods as part oF our Post-wildfire restoration plots – Pine Ridge, CO ongoing work with the Bureau of Land Management. From exploring biodiversity benefits of green roofs in Chicago, to understanding the impacts of climate change on rare plants in the Western U.S., and From developing sustainable pollination strategies For crops to developing tools to better conserve Fungi, Garden scientists and students are positively impacting plants and plant communities around the world. Training the next generation oF plant scientists and conservation biologists continues to be an important activity in the department. The joint CLM interns collecting seed in NM graduate program with Northwestern University is growing and thriving. Many oF our students received prestigious Fellowships and awards in 2012. Over 120 interns contributed to important stewardship activities on public lands as part oF our Conservation and Land Management Intern Program. Students From middle school through to post-doctoral researchers worked alongside Garden scientists as part oF the Science Career Continuum. Garden scientists contributed their expertise to conservation meetings and workshops in Taiwan and Korea and beyond. -

A key goal in evolutionary biology is to discover and characterize changes in the genetic programming of development that drive the modification and diversification of morphology. Here we present a synopsis of reproductive architecture in Papaveraceae s.l., a lineage in which the evolution of inflorescence determinacy, flower structure and symmetry, and effloration sequence produced unique reproduc- tive syndromes. This study aims to develop a new method for isolation of benzophenanthridine alkaloids from Macleaya leaves, which do not use toxic or ecologically dangerous solvents or reagents. According to this method, the extraction of alkaloids from the plant material is performed with 90% ethanol by percolation at 40±2 °C until 5–8 parts (V/M) of extract is obtained. The extract is acidified with sulphuric acid to pH 4.0–4.5 for sedimentation of ballast substances. After filtration or centrifugation, sulphuric acid is added to the extract to obtain the concentration of 0.1–0.12 M. Crystallisation of bisulphates of alkaloids occurs during 14 days at room temperature, then is finished in 7 days at 2–8 °C. The crude product is separated by filtration, washed with 96% ethanol and dried. Upon purification, alkaloids pass into a concentrated aqueous solution in the form of sulphates. A major part of impurities is removed by sedimentation, the remainder – by sorption on activated carbon. Than alkaloids are crystallised again in the form of bisulphates or other salts as required. The procedure has been created to obtain the sum of benzophenanthridine alkaloids of Macleaya in form of salicylates. This substance, named "Sanguirisal", is proposed as an active substance for preparation of pharmaceutical forms for topic admin- istration, being much more lipophilic than sanguiritrine. -
