Drug Exposure Definition and Healthy User Bias Impacts on the Evaluation of Oral Anti Hyperglycemic Therapies by Maxim Eskin A thesis submitted in partial fulfillment of the requirements for the degree of Master of Science in Health Policy Research School of Public Health University of Alberta © Maxim Eskin, 2017 ABSTRACT Accurate estimation of medication use is an essential component of any pharmacoepidemiological research as exposure misclassification will threaten study validity and lead to spurious associations. Many pharmacoepidemiological studies use simple definitions, such as the categorical “any versus no use” to classify oral antihyperglycemic medications exposure, which has potentially serious drawbacks. This approach has led to numerous highly publicized observational studies of metformin effect on health outcomes reporting exaggerated relationships that were later contradicted by randomized controlled trials. Although selection bias, unmeasured confounding, and many other factors contribute to the discrepancies, one critical element, which is often overlooked, is the method used to define exposure. Another factor that may provide additional explanation for the discrepancy between randomized controlled trials and observational study results of the association between metformin use and various health outcomes is the healthy user effect. Aspects of a healthy lifestyle such as following a balanced diet, exercising regularly, avoiding tobacco use, etc., are not recorded in typical administrative databases and failure to account for these factors in observational studies may introduce bias and produce spurious associations. The influence of a healthy user bias has not been fully examined when evaluating the effect of oral antihyperglycemic therapies in observational studies. It is possible that some, if not all, of the benefit, observed with metformin in observational studies may be related to analytic design and exposure definitions. -

Bias in preclinical animal research Susan Bridgwood Green Abstract Background: The public and healthcare workers have a high expectation of animal research which they perceive as necessary to predict the safety and efficacy of drugs before testing in clinical trials. However, the expectation is not always realised and there is evidence that the research often fails to stand up to scientific scrutiny and its 'predictive value' is either weak or absent. Discussion: Problems with the use of animals as models of humans arise from a variety of biases and systemic failures including: 1) bias and poor practice in research methodology and data analysis; 2) lack of transparency in scientific assessment and regulation of the research; 3) long-term denial of weaknesses in cross-species translation; 4) profit-driven motives overriding patient interests; 5) lack of accountability of expenditure on animal research; 6) reductionist-materialism in science which tends to dictate scientific inquiry and control the direction of funding in biomedical research. Summary: Bias in animal research needs to be addressed before medical research and healthcare decision-making can be more evidence-based. Research funding may be misdirected on studying 'disease mechanisms' in animals that cannot be replicated outside tightly controlled laboratory conditions, and without sufficient critical evaluation animal research may divert attention away from avenues of research that hold promise for human health. The potential for harm to patients and trial volunteers from reliance on biased animal data1 requires measures to improve its conduct, regulation and analysis. -

[email protected] Abstract: This target article is concerned with the implications of the surprisingly different experimental practices in economics and in areas of psychology relevant to both economists and psychologists, such as behavioral decision making. We consider four features of ex- perimentation in economics, namely, script enactment, repeated trials, performance-based monetary payments, and the proscription against deception, and compare them to experimental practices in psychology, primarily in the area of behavioral decision making. Whereas economists bring a precisely defined “script” to experiments for participants to enact, psychologists often do not provide such a script, leaving participants to infer what choices the situation affords. By often using repeated experimental trials, economists allow participants to learn about the task and the environment; psychologists typically do not. Economists generally pay participants on the ba- sis of clearly defined performance criteria; psychologists usually pay a flat fee or grant a fixed amount of course credit. Economists vir- tually never deceive participants; psychologists, especially in some areas of inquiry, often do. We argue that experimental standards in economics are regulatory in that they allow for little variation between the experimental practices of individual researchers. -

Mukurunge, N. Tlali Limkokwing University of Creative Technology, Maseru, Lesotho The curriculum in Lesotho at tertiary level does not adequately prepare graduates for the employment world, nor for self-made business people. The main emphasis is theoretical and academic excellence rather than for production in industry or for empowerment with entrepreneurial skills. This is a problem because Lesotho does not have an industrial power base for the economy to employ a big number of tertiary graduates. The industries that offer employment to citizens are mainly the Chinese-owned textile industries and taxi/transport industry, or the South African mines and farms which require cheap labour and not tertiary graduates. Lesotho therefore requires an education that produces graduates who can create employment for other primarily or highly skilled technocrats who will be able to be employed above the level of mere labourers in the sophisticated economy of South Africa. This study therefore sought to establish what the Ministry of Education and tertiary institutions are doing about this scenario, whether they have plans for curriculum review that will be aligned towards producing entrepreneurs and technocrats for the economic development of Lesotho. This research will benefit the authorities responsible for development through small businesses, the employment sector, as well as tertiary institutions in curriculum review. Abstract The aim of the study was to investigate the role that cognitive abilities, rational thinking abilities, cognitive styles and self-control play in explaining the endorsement of epistemically suspect beliefs among university students. A total of 159 students participated in the study. We found that different aspects of rational thought (i.e. rational thinking abilities and cognitive styles) and self-control, but not intelligence, significantly predicted the endorsement of epistemically suspect beliefs. Based on these findings, it may be suggested that intelligence and rational thinking, although related, represent two fundamentally different constructs. -

SAMPLE BIAS, BIAS OF SELECTION AND DOUBLE-BLIND 10.1 SAMPLE BIAS: In statistics, sampling bias is a bias in which a sample is collected in such a way that some members of the intended population are less likely to be included than others. It results in abiased sample, a non-random sample[1] of a population (or non-human factors) in which all individuals, or instances, were not equally likely to have been selected.[2] If this is not accounted for, results can be erroneously attributed to the phenomenon under study rather than to the method of sampling. Medical sources sometimes refer to sampling bias as ascertainment bias.[3][4] Ascertainment bias has basically the same definition,[5][6] but is still sometimes classified as a separate type of bias Types of sampling bias Selection from a specific real area. Research Center for Statistics Maria-Pia Victoria-Feser GSEM, University of Geneva B [email protected] Professor of Statistics Executive Summary (September 2019) Employment I started my career as a lecturer at the London School of Economics (LSE) in 1993, the year I completed history my Ph. D. in econometrics and statistics. In 1997, I was hired as lecturer in the Faculty of Psychology and Educational Sciences (FPSE) of the University of Geneva. Between 1997 and 2001, my position has evolved until I became full professor (50%) in 2001 at HEC-Genève. Since 2018, I have a 100% permanent position at the Geneva School of Economics and Management (GSEM). Awards and I received a total of 3,277,537 CHF of personal research funding (Swiss National Science Foundation grants (SNSF) and private) for seven projects, and over 500,000 CHF of research funding for collaborative projects. I was awarded the Latzis International Prize (1995) for my Ph. D. Thesis, as well as a doctoral (1991) and a professorial (2000) fellowships from the SNSF. Research My main research interest is statistical methodology and its application in different domains. I started with the development of robust methods of estimation and inference for application in economics (welfare analysis, risk analysis) and psychology (psychometric). I am also interested in computational statistics and have developed estimation methods for complex models (latent dependence structures, missing data), time series (signal processing), and model selection in high dimensions. My publications are often co-authored with former Ph. D. students or with academic researchers in other universities and/or outside the methodological statistics field.
