However, recent evidence suggests that MTHFD2 has a non-enzymatic function which may underlie the dependence of cancer cells on this protein. Understanding this non-enzymatic function is important for optimal targeting of MTHFD2 in cancer. Methods: To identify potential non-enzymatic functions of MTHFD2, we defined its interacting proteins using co-immunoprecipitation and mass spectrometry and integrated this information with large-scale co-expression analysis, protein dynamics, and gene expression response to MTHFD2 knockdown. Results: We found that MTHFD2 physically interacts with a set of nuclear proteins involved in RNA metabolism and translation, including components of the small ribosomal subunit and multiple members of the RNA-processing hnRNP family. Interacting proteins were also in general co-expressed with MTHFD2 in experiments that stimulate or repress proliferation, suggesting a close functional relationship. Also, unlike other folate one-carbon enzymes, the MTHFD2 protein has a short half-life and responds rapidly to serum. Finally, shRNA against MTHFD2 depletes several of its interactors and yields an overall transcriptional response similar to targeted inhibition of certain ribosomal subunits. Conclusions: Taken together, our findings suggest a novel function of MTHFD2 in RNA metabolism and translation.

Moran1,2,3 Cancer results from processes prone to selective pressure and dysregulation acting along the sequence-to-phenotype continuum DNA-RNA-protein-disease. However, the extent to which cancer is a manifestation of the proteome is unknown. Here we present an integrated omic map representing non-small cell lung carcinoma. Dysregulated proteins not previously implicated as cancer drivers are encoded throughout the genome including, but not limited to, regions of recurrent DNA amplification/deletion. Clustering reveals signatures composed of metabolism proteins particularly highly recapitulated between patient-matched primary and xenograft tumours. Interrogation of The Cancer Genome Atlas reveals cohorts of patients with lung and other cancers that have DNA alterations in genes encoding the signatures, and this was accompanied by differences in survival. Background: Long-term observations and studies have found that the occurrence and development of lung adenocarcinoma (LUAD) is associated with certain metabolic changes and that metabolic disorders are directly related to carcinogenic gene mutations. We attempted to establish a prognostic model for LUAD based on the expression profiles of metabolic genes. Method: We analyzed the gene expression profiles of patients with LUAD obtained from The Cancer Genome Atlas (TCGA). Univariate Cox regression was used to assess the correlation between each metabolic gene and survival. The survival-related metabolic genes were fit into the least absolute shrinkage and selection operator (LASSO) to establish a prognostic model for LUAD. After 100,000 times of calculations and model construction, we successfully established a prognostic model consisting of 16 genes that can classify patients with LUAD into high-risk and low-risk groups. Further, the protein-protein interaction (PPI) network was built to determine the hub gene from16 metabolic genes.
