DATA

Western Blot Simple Western Detection of Human/Mouse Detection of Mouse AlphaA Crystallin/CRYAA by AlphaA Crystallin/CRYAA by Simple Western Blot. Western blot shows WesternTM. Simple Western lane view lysates of mouse tissue. shows lysates of mouse eye lens tissue, PVDF membrane was probed with loaded at 0.2 mg/mL. A specific band was 0.1 µg/mL Goat AntiHuman/Mouse detected for AlphaA Crystallin/CRYAA at AlphaA Crystallin/CRYAA Antigen approximately 28 kDa (as indicated) using Affinitypurified Polyclonal Antibody 1 µg/mL of Goat AntiHuman/Mouse (Catalog # AF4848) followed by HRP AlphaA Crystallin/CRYAA Antigen Affinity conjugated AntiGoat IgG Secondary purified Polyclonal Antibody (Catalog # Antibody (Catalog # HAF017). For AF4848) followed by 1:50 dilution of HRP additional reference, recombinant conjugated AntiGoat IgG Secondary human CRYAA and CRYAB Antibody (Catalog # HAF109). This (5 ng/lane) were included. Bands for experiment was conducted under AlphaA Crystallin/CRYAA were reducing conditions and using the detected at approximately 20 23 kDa 12230 kDa separation system. (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 2.

BACKGROUND AlphaA Crystallin (CRYAA, also known as Crystallin αA chain and HspB4) is a 2122 kDa member of the family of . It is found in lens epithelium where it noncovalently oligomerizes with CRYαB to generate a transparent 3501000 kDa αCrystallin complex. Human CRYAA is 173 amino acids (aa) in length. There is a αCrystallin Hsp domain over aa 63145. Multiple posttranslational modifications may exist. The Cterminal Ser and the Nterminal MetAspVal is occasionally cleaved. There is also phosphorylation at Ser45 and 122, Olinked GlcNAc modification at Ser162, deamidation at Asn101, and acetylation at Lys70. Full length human CRYAA is 54% aa identical to CRYAlphaB, and 95% aa identical to mouse CRYAA.
