The Speciation History of Heliconius: Inferences from Multilocus DNA Sequence Data
The speciation history of Heliconius: inferences from multilocus DNA sequence data by Margarita Sofia Beltrán A thesis submitted for the degree of Doctor of Philosophy of the University of London September 2004 Department of Biology University College London 1 Abstract Heliconius butterflies, which contain many intermediate stages between local varieties, geographic races, and sympatric species, provide an excellent biological model to study evolution at the species boundary. Heliconius butterflies are warningly coloured and mimetic, and it has been shown that these traits can act as a form of reproductive isolation. I present a species-level phylogeny for this group based on 3834bp of mtDNA (COI, COII, 16S) and nuclear loci (Ef1α, dpp, ap, wg). Using these data I test the geographic mode of speciation in Heliconius and whether mimicry could drive speciation. I found little evidence for allopatric speciation. There are frequent shifts in colour pattern within and between sister species which have a positive and significant correlation with species diversity; this suggests that speciation is facilitated by the evolution of novel mimetic patterns. My data is also consistent with the idea that two major innovations in Heliconius, adult pollen feeding and pupal-mating, each evolved only once. By comparing gene genealogies from mtDNA and introns from nuclear Tpi and Mpi genes, I investigate recent speciation in two sister species pairs, H. erato/H. himera and H. melpomene/H. cydno. There is highly significant discordance between genealogies of the three loci, which suggests recent speciation with ongoing gene flow. Finally, I explore the phylogenetic relationships between races of H. melpomene using an AFLP band tightly linked to the Yb colour pattern locus (which determines the yellow bar in the hindwing). -

Der Hauptzweck dieser Arbeit ist die Identifizierung einer Schmetterlingspopulation der Gattung Ithomiinae die in einem subtropischen Waldstück vorkommt welches sich in den östlichen Ausläufern der Westkordillere Kolumbiens im Departemento von Caldas befindet, und zweitens die Flora zu kennen, in der diese Population vorkommt. Ergebnisse. Es wurden Insgesamt 42 Ithomiinae Arten und 23 Gattungen beschrieben. Kenntnisse. Es wurde festgestellt, dass einige Arten in mimetischen und kryptischen Komplexen mit anderen Schmetterlingsfamilien und Arten der Region vorkommen. Schlussfolgerungen. Die Bedeutung der ökologischen Dynamik welche sich aus dieser Gemeinschaft ergibt wird hervorgehoben. Außerdem soll das Verzeichnis der vorkommenden typischen Arten der Kaffeeanbauzone Kolumbiens ergänzt werden. Este trabajo tiene como propósito identificar una población de mariposas Ithomiinae que habita un fragmento de bosque subtropical ubicado en el piedemonte este de la cordillera Occidental de Colombia en el departamento de Caldas y, en segunda instancia, conocer la flora en la que conviven. Resultados. Se listan un total de 42 especies pertenecientes a 23 géneros de Ithomiinae. -

Six species are recognized: Phalacropsyl- la allos Wagner, P. hamata Tipton and Mendez, P. morlani Eads and Campos, P. nivalis Barrera and Traub, P. oregonensis Lewis and Maser, and P. paradisea Rothschild. Phalacropsylla hamata is designated herein as a junior synonym of P. paradisea. The distribution of P. paradisea is more extensive than previously thought, extending from Arizona through southern Colorado, into New Mexico, Texas, and northern Mexico (State of Nuevo León). -

Chong Saint Joseph’s Institution, 38 Malcolm Road, 308274 Singapore [email protected] ABSTRACT. The moth fauna of Bukit Timah Nature Reserve, Singapore, was studied using light trapping. Specimens and photographs were sorted into morphospecies including macro and micro moths. A total of 399 species has been found, of which nearly 200 have been identified to species level. Several are notably rare or otherwise of interest. The figures do not reach an asymptote, suggesting that the total moth fauna may be considerably greater. The nature reserve may be too small to yield statistically significant differences in the moth fauna between forest vegetation zones. Keywords. Biodiversity, insects, Lepidoptera, survey techniques, tropical rain forest Introduction Although Alfred Russel Wallace investigated extensively the fauna and flora of the Malay Archipelago between 1854 and 1862, collecting at least 110,000 insects, including more than 3,000 Lepidoptera (Wallace, 1869), his emphasis was not upon moths. Some moths were mentioned and/or collected by him but not in significant numbers (Wallace, 1869). Except for several months after his first arrival, in 1854, when he was based near what is now Bukit Timah Nature Reserve (BTNR), his trips to Singapore seem to have been largely for practical reasons, such as replenishing supplies, rather than focussing on nature. This is understandable in the context that, relative to the richness of the surrounding islands, Singapore was likely to yield fewer and fewer new species for a given time or effort. -

Bushley August 2009 © 2009 Kathryn E. Bushley EVOLUTION OF NONRIBOSOMAL PEPTIDE SYNTHETASE PROTEINS INVOLVED IN SECONDARY METABOLISM IN FUNGI Kathryn E. Bushley, Ph. D. Cornell University 2009 Nonribosomal peptide synthetases (NRPSs) are multimodular enzymes which biosynthesize peptides (NRPs) independently of ribosomes. Three core domains (adenylation (A), thiolation (T), condensation (C)) comprise a functional module for NRP biosynthesis. Although NRPSs produce a diversity of bioactive compounds, little is known about the evolutionary relationships of genes encoding NRPSs and the mechanisms by which they evolve. The objectives of this research were to perform phylogenomic analyses to identify major NRPS subclasses and determine evolutionary relationships and to elucidate fine-scale evolutionary mechanisms giving rise to the diverse NRPS domain structures in fungi. Chapter 2 is a published manuscript on ferrichrome synthetases tracking the evolution of domain architectures of these relatively conserved enzymes across fungi. Results supported the hypothesis that ferrichrome synthetases evolved by tandem duplication of complete modules (A-T-C) (single or double units) and loss of single A domains or complete A-T-C modules. A mechanism for evolution of iterative biosynthesis is proposed. Protein modeling of the A domain substrate binding pockets refined characterization of key residues involved in substrate specificity, by identifying novel sites. Chapter 3 reports a fungal kingdom-wide phylogenomic study of NRPSs, with the objective of identifying subclasses.
