Emerging Hemp Diseases in New Jersey

Dr. Andrew Wyenandt Richard Buckley Dr. Nimmi Rajmohan Diseases on Hemp

• Yellow leaf spot- Septoria sp

• Hemp leaf spot- Bipolaris sp

• Powdery Mildew Hemp trial site at Snyder farm Yellow leaf spot - Septoria sp. B

• Common leaf spot fungus on Hemp A

• Irregular leaf spots with yellow margins C • Symptoms begin on lower mature leaves

Photos: Nimmi Raj • Leads to defoliation and stunted growth

• Observed in the field

. Leaf spots on Hemp at Snyder Hemp leaf spot - Bipolaris sp. A B C

• New pathogen of Hemp

• Symptoms begin as tiny specks or dots

D E F • Spots coalesce to form irregular shape

• Observed in the field Photos: Nimmi Raj Hemp leaf spot at RAREC Photo credit: Dr Raul Cabrera Powdery Mildew A B

• White powdery growth on leaf surface C • Host specific

• Identified as Golovinomyces sp.

• Mostly observed in greenhouses Powdery mildew on Hemp leaves Managing Diseases in Hemp

Must Have Recommendation Use clean seeds and cuttings; Good sanitation Sanitize tools; Remove plant debris

Increase spacing between Good drainage plants; Improve air circulation

Weeds can be alternate fungal Good weed control hosts

Good soil health Test soil fertility The New RU Hemp Testing Lab

New Jersey Agricultural Experiment Station in concert with: – The New Use Agriculture and Natural Plant Products Program – The Center for Agricultural Food Ecosystems (RUCAFE) – School of Biological & Environmental Sciences – Rutgers University, New Brunswick, NJ Rutgers Hemp Analytics Lab:

All incoming hemp samples must: 1. Be accompanied with a completed intake form 2. Be logged in upon arrival 3. Must be accompanied by a hemp permit 4. We ask for a 24-hour notification. 5. Samples needed to be received Monday & Friday only 6. Traceability and record keeping critical

Already isolating glandular trichomes for CBDs & THC content • Above: Our Agilent 6540 Q-TOF Mass Spectrometer with an electrospray ionization (ESI) source, and software and libraries for metabolomics and protein/peptide mapping (for biochemistry, metabolomic and quality control studies) • Left: UPLC-QqQ/Ms System: Agilent 1290 Infinity II UPLC interfaced with an Agilent 6470 Triple Quadrupole Mass Spectrometer with an electrospray ionization (ESI) source; (for bioanalysis) Illustrative example of quantitation of hemp bioactives

LC/MS TIC of (A) untreated and (B) decarboxylated high-Δ9-THCA; (C) untreated and (D) decarboxylated high-CBDA; and (E) partly decarboxylated high-CBGA strains

Accurate quantitative analysis of natural products (even when in very low concentrations) in plant or in biological samples: Agilent 1290 Infinity II UHPLC interfaced with an Agilent 6470 Triple Quadrupole MS with ESI. Chemically characterizing the aroma, flavor and fatty acids in Hemp: By GC/MS • GC/MS, SPME or Headspace Volatile Capture and Analysis used to determine aroma composition and fatty acid composition in hemp and can analyze THC and CBD. • SCFA and aromatic compounds can rapidly be analyzed using our Shimadzu GCMS-TQ8040 Triple Quadrupole MS interfaced with a Shimadzu AOC6000 autosampler allowing us to us to detect very low concentrations.

Retention time

For CBD and THC: MW of CB/MW of CB- A = 0.877

Memmert ICH 110 Stability Chamber

The Climate Chamber ICH has been specially designed for testing pharmaceutical products according to ICH - WHO - EMA - ASEAN - GMP - GLP – GCCP guidelines.

This equipment was built precisely to conduct stability testing (according to ICH Q1A) for accelerated stability studies and for long-term storage/stability studies (can control temperature and relative humidity). Now you/we can provide support by conducting such plant-based/food- based/cosmetic-based/nutraceuticals correctly to support your efforts to examine chemistry, overall product quality (texture, consistency, color, aroma and more in addition to CBD and THC).

This is particular useful when doing product formulations for hemp and CBD. The RU Hemp Analytics Lab: Open For Research & Collaboration *We now have grower and processor/handler permit from NJDA *We also established a new analytical lab for hemp and CBD testing and its operational *Our instrumentation can quantitate: • delta9, delta8, total THC, and CBDs, as well as minor CBs (UPLC- Special thanks to: QqQ/MS) and novel CB metabolites • New Jersey Agricultural Experiment Station (UPLC-QTOF/MS) (W. Cohick, B. Hillman, M. Brennan) • seed oils & terpenes (GC/MS) • Rutgers Cooperative Extension Service *Have pharma grade climate chamber (B. Schilling) • New Jersey Department of Agriculture to evaluate product stability and • Our hemp team colleagues accelerated aging. (R. Cabrera; S. Komar, B. Bamka, N. *Working next on processing and Rajmohan, A. Wyenandt) product quality • Agilent Instruments and Shimadzu *Working to register lab with DEA At SEBS, each Fall, we teach a 3-credit course Hemp & (11:776:390) *Working with Bressan Lab, Purdue For more info: [email protected]
