The solvation of biomolecules is important in molecular biology .In this theoretical study, we focus on a kind of dammarane sapogenins (PAM120). This molecule optimized in various solvent media such as heptan, carbontetrachloride, toluene, tetrahydrofurane, dichloroethane, ethanol, methanol, dimethylsulfoxide and water using the self-consistent reaction field model. This process depends on either the reaction potential function of the solvent or charge transfer operators that appear in solute-solvent interaction. We performed nonempirical quantum-mechanical calculations at the HF/3-21G, 6-31G , 6-31G*,6-31G** and B3LYP/6-31G** levels of theory in the gas phase and some solvents at 298K.We studied energy ,dipole moment ,charge and so on. Accepted 2 April, 2013 Naturally occurring saponins are glycosides of steroids, alkaloids and triterpenoids. They are widely distributed in nature and reported to be present in 500 genera of plants. A wide variety of plants belonging to family Liliaceae, Dioscoreaceae, Solanaceae, Sapindaceae and Agavaceae are the major source of saponins. They are amorphous substances having high molecular weight and are soluble in water and alcohol to produce foam but organic solvents inhibit their foaming property. Plants saponins are generally extracted into butanol through liquid-liquid partition and separated through column chromatography using silica gel as adsorbent and chloroform: methanol as mobile phase. HPLC, GC, Sephadex LH-20 Chromatography, DCCC, preparative paper chromatography and TLC were also used for the separation and isolation of saponins. The structures of saponins were determined by several spectroscopic techniques, viz., UV, IR, 1H NMR, 13 C NMR and Mass spectroscopy. Saponins possess several biological activities such as antioxidant, immunostimulant, antihepatotoxic, antibacterial, anticarcinogenic, antidiarrheal, antiulcerogenic, antioxytoxic, antihypoglycemic, anticytotoxic and antimolluscicidal. Saponins are biologically synthesized by C5 isoprene units through cytosolic mevalonate pathway. -

POTENTIAL MECHANISMS OF ACTION OF KAEMPFEROL IN THE PREVENTION OF BREAST CANCER A Dissertation Presented to the Faculty of the Graduate School of Cornell University in Partial Fulfillment of the Requirements for the Degree of Doctor of Philosophy by Jinzhou Li August 2015 © 2015 Jinzhou Li POTENTIAL MECHANISMS OF ACTION OF KAEMPFEROL IN THE PREVENTION OF BREAST CANCER Jinzhou Li, Ph.D. Cornell University 2015 Epidemiological studies have shown that regular consumption of fruits and vegetables could reduce the risk of cancer. Phytochemicals in fruits and vegetables have been suggested to be responsible for their health benefits. Asparagus and one of its major phytochemical, Kaempferol, have been reported to have anti-cancer activities. However, the mechanisms of the anticancer activities are not completely understood. Seven varieties of asparagus were compared for their antioxidant content, antioxidant activities, and anticancer activities. The results showed that asparagus are rich in phenolic and flavonoids. They also show antioxidant activities in chemical assays and in vitro assays. Asparagus possesses potent anticancer abilities in inhibiting proliferation of HepG2 human liver cancer cells. Among the different varieties, Apollo has the highest phenolic content, flavonoid content, antioxidant activities, and anticancer activities. We further studied kaempferol’s effects in breast cancer cells growth and metastasis. The anti-proliferative activity and cytotoxicity of kaempferol against MCF-7 human breast cancer cells were measured by the methylene blue assay. All the key proteins regulating cell proliferation through signaling transduction pathways were determined by Western blot assay. Kaempferol exhibited potent anti-proliferative activity against MCF-7 human breast cancer cells in a dose-dependent manner. -

The uptake and metabolism of 2-[14C]-2,4-dichlorophenoxyacetic acid (2,4-D) were investigated in leaf calluses of Nicotiana tabacum, tuber calluses of Dioscorea opposita and calluses derived from zygotic embryos, leaves and petioles of Dioscorea composita. Striking similarities were evident in the patterns of 2,4-D metabolites and their chemical characteristics in the three callus types of D. composita compared, but significant differences were detected among the patterns of rnetabolites in the three species studied. Preliminary investigations on the stability of various metabolites (separated using TLC) by hydrolysis showed that sugar esters appeared to be the major metabolites in tobacco whilst in yams (D. opposita) glycosides were shown to be the main ones, which indicated a similarity between plants of Gramineae and Dioscoreaceae in terms of 2,4-D metabolism. Release of 2,4-D from tobacco callus cells upon their transfer to 2,4-D-free medium was detected and the implications of this are discussed in relation to the cultural conditions necessary to induce morphogenesis in vitro. 1 Chapter 4 Phytomorph and Geomorph Identification © This Chapter is based on three published works: (1) a paper by Hugh O Neall (1944) that identifies two New World plants (sunflower and chili peppers) in the Voynich manuscript; (2) a paper of Tucker and Talbert (2013) which identified 39 plants in the Voynich as indigenous to the New World; (3) a paper by Tucker and Janick (2016) which extended the list to 59 species. Although many of the illustrations of the Voynich Codex on first blush could be considered bizarre or whimsical (See Figure in Chapter 14) most contain morphological structures which permit botanical identification. Many enthusiasts have attempted to analyze the plants of the Voynich Codex, but few are knowledgeable plant taxonomists or botanists, despite their large web presence. Most of the plant identification has been predicated on the conclusion that the Voynich is a 15th century European manuscript (Friedman 1962). The principal reports in a web report by non botanists Edith and Erica Sherwood (http:www.edithsherwood.comn/coyhnich_botanical_plants) who identifies he plants as Mediterranean based on their premise that Voynich is a 15th century Italian manuscript and claims to find signature of Leonardo da Vinci in voynich drawings. We respectfully disagree with both assertions. The first exception to the conclusion that the Voynich plants were European is a short remarkable 1944 paper in Speculum (a refereed journal of the Medieval Academy of America) by the distinguished plant taxonomist, the Rev./Dr. Hugh O’Neill (1894–1969), former Director of the Herbarium (official acronym LCU) at the Catholic University of America (CUA) in Washington, D.C.
