Kappa opioid receptor (KOR) antagonists have recently shown potential for treating drug addiction and 1,2,3 4 8 depression. Arodyn (Ac[Phe ,Arg ,D-Ala ]Dyn A(1-11)-NH2), an acetylated dynorphin A (Dyn A) analog, has demonstrated potent and selective KOR antagonism, but can be rapidly metabolized by proteases. Cyclization of arodyn could enhance metabolic stability and potentially stabilize the bioactive conformation to give potent and selective analogs. Accordingly, novel cyclization strategies utilizing ring closing metathesis (RCM) were pursued. However, side reactions involving olefin isomerization of O-allyl groups limited the scope of the RCM reactions, and their use to explore structure-activity relationships of aromatic residues. Here we developed synthetic methodology in a model dipeptide study to facilitate RCM involving Tyr(All) residues. Optimized conditions that included microwave heating and the use of isomerization suppressants were applied to the synthesis of cyclic arodyn analogs.

Methods: Participants underwent a one-to-one semi-structured interview exploring the subjective effects of ibogaine. We employed interpretative phenomenological analysis to identify relevant phenomenological categories, including physical sensations, perceptual (visual, auditory, and olfactory), emotional, cognitive, and spiritual. Participants also compared ibogaine with other drugs used in life, including psychedelics like ayahuasca, psilocybin mushrooms, and lysergic acid diethylamide. Results: The findings reveal that the subjective experience with ibogaine has similarities with other psychedelic substances, but also important differences. These include very strong and unpleasant physical effects as well as, at least in this patient population, a very difficult and challenging experience. Conclusions: Overall, the descriptions involve heightened memory retrieval, specially related to drug abuse and the perception of one’s own future with or without drug use.

    1 (A Brief History and) Motivation of an Entheogenic Chemist Casey William Hardison - POWd (Civ) Abstract Casey Hardison was arrested spring 2004 for the production of psychedelic-type drugs, i.e., LSD, 2C- B and DMT. In the three years since, not one person from ‘authority’ had bothered to ask him what motivated him to synthesise psychedelic drugs. It was as if the a priori assumption that ‘all illegal drugs are bad’ had provided the answer. Hence, the Judge asserted that Hardison did it for “that basest of human emotion, greed” as though the psychospiritual benefits of an alchemical path dedicated to expanding consciousness and personal transformation, through insights integrated into action, upon which he had expounded at great lengths during trial, were some elaborate “portmanteau defence”, just some ruse to get him out of the dock. It was not, it was a committed stand for ‘cognitive liberty’ and for a world full of people who understand the fine line between alone and all one. MINDSET I was born in Washington State on the edge of Western exploration in the New World in the summer of 1971. I came of age in and around the communal rooms of AA, NA, ACA, Alanon and Alateen throughout the Pacific and Mountain West. My father is 33 years sober. His father died 14 years sober in 1982. I too wrestled my psychospiritual demons through alcohol and Cannabis which gratefully led me to the rooms of AA and NA where, at the age of 14, I declared myself an alcoholic and an addict. I delved headlong into the 12 Steps and promptly saw that I had wrapped myself in a shame-bound identity (‘ism’ - internalised shame manifested).

Two aromatic amino acids, Tyr1 and Phe3 or Phe4, are important structural elements in opioid peptides because they interact with opioid receptors. The usefulness of an artificial amino acid residue 2,6-dimethylphenylalanine (Dmp) was investigated as an aromatic amino acid surrogate for several opioid peptides, including enkephalin, dermorphin, deltorphin, endomorphin, dynorphin A, and nociceptin peptides. In most peptides, substitutions of Phe3 byaDmpresidueproducedanalogswithimproved receptor-binding affinity and selectivity, while the same substitution of Phe4 induced markedly reduced receptor affinity and selectivity. Interestingly, replacement of Tyr1 by Dmp produced analogs with unexpectedly high affinity or produced only a slight drop in receptor affinity and bioactivity for most peptides. Thus, Dmp is also a useful surrogate for the N-terminal Tyr residue in opioid peptides despite the lack of a phenolic hydroxyl group, which is considered necessary for opioid activity.
