Abstract

The goal of this project was to inventory the diatom flora of State Parks located in southwestern

Pennsylvania. For the last few years this region has been experiencing a rapid increase in natural gas production. Protecting the health of lakes and rivers is an issue of growing importance in shale gas regions and documenting the current state of the aquatic biota is crucial for any future assessments of human impacts. The diatoms play a major role in aquatic ecosystems and are sensitive and widely used indicators of water quality. Collecting baseline data on the composition of diatom communities in State Parks in southwestern Pennsylvania is an important step towards monitoring environmental health in the region. 153 diatom samples have been collected from in various habitats, such as streams, lakes, and wetlands in twelve State Parks.

The diatoms were identified using light and scanning electron microscopy. A total of 451 diatom taxa were documented in studied water bodies. Two diatom species new for science were discovered; one of these species is likely an Appalachian endemic. The invasive species

Didymosphenia geminata was found in Youghiogheny River (Ohiopyle SP), but it did not form severe blooms impairing river ecosystem. A number of water bodies are characterized by diatom assemblages indicative of moderate to severe impairment such as nutrient enrichment or acid mine drainage.

Objective

Collecting baseline data on the composition of diatom communities in State Parks in southwestern Pennsylvania is an important step in monitoring environmental health in the region.

These data will be useful for future comparisons and impact assessments. They contribute to the knowledge of diatom flora of Pennsylvania and will reveal habitats containing rare, endemic, and possibly invasive diatom species. This work also identifies water bodies with degraded biota and impaired water quality.

Justification

This project belongs to the category of the baseline surveys of aquatic species in State

Parks within the shale gas region. Southwestern Pennsylvania experienced especially fast industrial and agricultural development from mid-1800s to 1950s. The aquatic ecosystems of this of this region were affected by logging, mining, and steel industry. In the last 50-60 years the impact of the industries significantly lessened and many watersheds recovered to various degrees. Many areas are now protected, and a number of State Parks have been established. In the few recent years, the region, however, became one of the “sweet” spots of the shale gas production (MSAC 2011). Although no gas wells are currently drilled in the Pennsylvania State

Parks, their biological communities may still be affected by the drilling and natural gas extraction in surrounding areas. On the other hand, lack of direct impact makes it possible to consider the Parks as refugia where flora and fauna are relatively sheltered from the negative influences.

Diatoms are eukaryotic microbes that are important indicators of environmental health in aquatic ecosystems (Stoermer & Smol 1999). Diatoms possess ornamented silica walls or frustules, which are left behind after the death of the organism. These frustules accumulate in lake sediments and serve as an indispensable tool in reconstructing changes in climate, water quality, and the history of the watersheds. Diatoms are extremely sensitive to changes in ion concentrations, pH, suspended sediment, nutrients, and dissolved organic matter. The high diversity of diatom communities and presence of native and endemic species are indicative of good-quality or undisturbed aquatic ecosystems. Together with fish and benthic macroinvertebrates, diatoms are used by many State and Federal environmental agencies to assess water quality in rivers, lakes, and wetlands (e.g. Potapova & Carlisle 2011). The quality of

4 these assessments depends on the knowledge of regional diatom floras and ecological responses of individual species and communities to a variety of environmental stressors.

These data will be useful for future comparisons and impact assessments. They will contribute to the knowledge of diatom flora of Pennsylvania and will reveal habitats containing rare, endemic, and possibly invasive diatom species. Another goal of this work is to identify water bodies with degraded biota and impaired water quality.

Our previous work in north-central Pennsylvania supported by WRCP showed that diatoms of the State are still poorly known (Potapova 2010). From 310 samples collected in 2009 we found several diatom species new for science. Five of these have been already formally described, while the descriptions of several others will be published in the nearest future

(Potapova 2011a,b, 2012, Enache & Potapova 2012). These new species and several other taxa found in north-central region are possible endemics of Pennsylvania or Eastern North America and are have a high value as indicators of good habitat quality. Such data are important for understanding biogeography of diatoms and other protists and ultimately, for developing better methods of biological assessment and monitoring. There were no diatom surveys in southwestern

Pennsylvania focused on taxonomic treatment of the flora and we can expect that the proposed study will yield at least some discoveries of new species or rare species occurrences.

Methods and Materials

153 diatom samples have been collected from 103 locations in various habitats, such as streams, lakes, and wetlands in twelve State Parks (Figure 2, Table 1). Diatom samples were hand-collected from near-shore benthic habitats; a few phytoplankton samples were collected using plankton net. Basic water quality (pH, conductivity) was measured in the field (Figure 4).

GPS coordinates were recorded. In the laboratory samples were treated by nitric acid to remove organic matter. The samples were rinsed 6 times with distilled water. The diatom slurry was used to prepare permanent diatom slides using the Naphrax mounting medium and stubs for the electron microscopy. The diatoms were identified to species level using historical (Patrick &

Reimer 1966, 1975) and contemporary floras (Krammer & Lange-Bertalot 1986, 1988, 1991,

2004) and many others.

Products delivered in addition to this final report

A database containing all the results of this study is delivered to DCNR in addition to this report. The database contains 2 tables and one query:

1) tbl_Samples is the list of all samples collected in the course of this project together with

information on collection sites with geographic coordinates, sample type (substrate) and

environmental characteristics, such as conductivity and pH.

2) tbl_Counts is the list of all observed taxa in each sample together with semi-quantitative

abundance value given on the scale from 1 (rare) to 5 (very abundant).

3) qry_taxa is the list of all diatom taxa observed in the studied samples together with

number of occurrences (number of samples in which species was found) and average

abundance value.

Results and conclusions

Measured conductivity and pH for the majority of water bodies indicated were within levels typical for Pennsylvania. However, several creeks had clearly elevated levels of conductivity above 500 and even above 1000 S/cm, obviously a legacy of previous mining activities. Some reservoirs and ponds, such as Lake Arthur and Yellow Creek Lake, had extremely high pH at the time of sample collection. These high pH values were caused by the intensive photosynthesis. For instance, in Lake Arthur, a considerable bloom of planktonic cyanobacteria (Gomphosphaeria sp.) was observed (Figure 5).

All 153 samples have been analyzed in the laboratory. A total of 451 diatom taxa were found (Table 2, see also attached database). Some diatoms could not be identified with certainty to species level using existing floras. Two of these diatom species formed large populations and were sufficiently abundant to establish that they indeed represented new species. One of these

10 species, tentatively called Encyonema appalachianum (Figure 6), is likely an Appalachian endemic. It was found in Youghiogheny River and its tributary Meadow Run (Ohiopyle SP,

Figure 7), Laurel Hill Creek and Lake (Laurel Hill SP), Yellow Creek Lake and Little Yellow

Creek (Yellow Creek SP), McCune Run (Keystone State Park), Big Spring Reservoir (Laurel

Ridge State Park), Lake Arthur and its tributary Muddy Creek (Moraine SP), a tributary to

Slippery Rock Creek (McConnell Mill SP), and Traverse Creek (Raccoon Creek SP). A manuscript formally describing this species has been submitted for publication and is currently in review.

Another species new for science belongs to the genus Navicula (Navicula sp. 4 in Figure

8). It has been as well studied in detail and its formal publication is in preparation. This species has been also recently observed in eastern Pennsylvania and it is not clear at this point why it has not been detected before.

Among 451 diatom species, the most common were: Achnanthidium minutissimum (70% samples), Cocconeis placentula (61%), Gomphonema parvulum (40%), Navicula cryptocephala

(39%), Navicula gregaria (36%), Planothidium frequentissimum (36%), and Navicula lanceolata

(33%). A. minutissimum, C. placentula and G. parvulum are known to represent species complexes rather than individual biological species (e.g., Potapova & Hamilton 2007, Jahn

Abarca et al. 2013, Kermarrec et al. 2013) and this are not especially sensitive indicators of environmental conditions. On the other hand, high frequency and abundance of such species as

N. gregaria, N. lanceolata, and P. frequentissimum indicate considerable nutrient enrichment: these species are usually associated with high levels of nitrogen and phosphorus (Potapova &

Charles 2007). Remarkably, almost all 30 most abundant species are known as indicators of moderate to high eutrophication or general pollution, except a few species that are complexes of multiple cryptic species or are associated with acid mine drainage (Eunotia exigua and E. bilunaris). In streams with higher conductivity due to previous mining acitivites, diatoms characteristic for highly mineralized or brackish waters, such as Nitzschia dissipata, were dominant.

Exploratory multivariate analysis of diatom count data (Figure 9) revealed that the largest differences in community structure were caused by pH variation in studied water bodies. The

Eunotia species clustered in the left side of the diagram are indicative of low pH, and the arrow for this factor point in the opposite direction. The centroid for the water bodies affected by the acid mine drainage (AMD) although positioned to the left of the other centroids, was not at the extreme left side of the diagram. This happened because some AMD passive treatment ponds had rather high pH and their diatom communities were thus considerably different from those in

AMD water bodies with low pH. The ordination also elucidated which species were especially abundant in higher-conductivity waters and in various kinds of water bodies.

A few water bodies affected by the acid mine drainage had acidic water with conspicuous deposition of iron hydroxide or yellow boy (Figure 10) and diatom community dominated by a single species, Eunotia exigua. Interestingly, a series of passive treatment ponds at Jennings

Environmental Center demonstrated a successful restoration of water quality. Although the conductivity was high in all treatment ponds, pH was almost circumneutral in lower ponds and diatom community became much more diverse and abundant in comparison to the upper pond.

Some creeks with relatively low pH had rather diverse representatives of the genus Pinnularia, including several species that did not fit any published descriptions, but did not form large enough populations for a morphological detailed study (Figure 11).

The invasive species Didymosphenia geminata was found in Youghiogheny River

(Ohiopyle SP). This species has been observed in the river since 2012. The growth was moderate, so that Didymosphenia did not form conspicuous masses and was found aming a diverse community of other diatoms and green algae (Figure 12). No growth of Didymosphenia was observed in any studied tributaries of the Youghiogheny River.

The conclusion of this work is a documentation of a rich diatom flora found in water bodies of Southwestern Pennsylvania. Although the diatom communities of this region are definitely far from pristine state, they are still highly diverse and provide vast opportunities for environmental monitoring and assessment. Further more detailed investigations of aquatic

17 ecosystems of this region would be beneficial for developing appropriate management guidelines and ultimately led to improved water quality.

Discussion and Management Recommendations

The diatom flora revealed in the course of this study is quite diverse, but is dominated by common pollution-tolerant species, typically found in moderately and even highly polluted rivers. The overall number of species (451) is lower than in previously studied diatom flora of

Northcentral Pennsylvania (622 species, Potapova 2010), although this number may be influenced by lower number of studied samples. In the Southwest 103 locations within the State

Parks were sampled, while in the North-central part of the State we sampled 140 sites on all kind of lands. A conspicuous difference between two floras is almost complete absence of pollution- sensitive species in the Southwest, which may be the legacy of intensive mining in this region in the past. The widespread acid mine drainage continues to affect aquatic communities even in the protected lands (Winter 2008). In North-central Pennsylvania a considerable number of streams had high-quality diatom communities dominated by native and rare species, while such habitats were practically absent in the Southwest.

Although the invasive species Didymosphenia geminata is by now firmly established in

Youghiogheny River, it was not observed to form excessive blooms. Its presence and possible spread nevertheless should be monitored since it will likely become more widespread (Spaulding &

Elwell 2007, Taylor & Bothwell 2014).

Diatoms in artificial lakes located in Southwestern Pennsylvania indicated moderate to high eutrophication. It would be beneficial to monitor lakes’ biological communities (Phyto- and ziooplankton and benthos) on a regular basis to prevent deterioration of water quality. An excessive bloom of cyanobacteria in Lake Arthur was a sure indication of nutrient enrichment directly leading to water-quality problems.

The samples collected in the course of this project and permanent diatom slides made from these samples are now stored in the ANSP Diatom Herbarium. These samples and slides are available for further studies involving various methodologies. For example, the diatoms might be counted in the future following bioassessment protocols, such as 500- or 600-valve counts. The data obtained from such observations might then be combined with data obtained from other surveys and used for a variety of purposes, such as inferring species optima and tolerances, or modeling species distributions. They can also be used as a reference in studies of human impacts and climate change. In fact, we are already starting to use these data as a benchmark for assessing impact of the fracking waste-water treatment facilities located on

Blacklick Creek, Western PA.
