Avoid repeated freezing and thawing Background Information Growth hormone-inducible transmembrane protein (GHITM), also known as transmembrane BAX inhibitor motif containing protein 5 (TMBIM5), is a protein that in humans is encoded by the GHITM gene on chromosome 10. GHITM, also known asMICS1, TMBIM5 orDERP2, is a mitochondrial protein which localizes in the inner membrane.GHITM is involved in mitochondrial morphology in specific cristae structures and the apoptotic release of cytochrome c from the mitochondria. The gene of GHITM maps to chromosome 10q23.1, and encodes a 345-amino- acid protein with a calculated molecular mass of 37 kDa. The apparent molecular weight has been reported to be 42 kDa, the increased size in the protein may be due to post-translational modifications. GHITM can be cleaved into smaller forms of 23-27 kDa. However, little progress has been made in defining human- specific in utero neurodevelopmental pathology due to ethical and technical challenges associated with accessing human prenatal brain tissue. Here we utilized human induced pluripotent stem cells (hiPSCs) to generate reproducible organoids that recapitulate dorsal forebrain development including early corticogenesis. We systemically exposed organoid samples to chemically defined “enviromimetic” compounds to examine the developmental effects of various narcotic and neuropsychiatric-related risk factors within tissue of human origin. In tandem experiments conducted in parallel, we modeled exposure to opiates (μ-opioid agonist endomorphin), cannabinoids (WIN 55,212-2), alcohol (ethanol), smoking (nicotine), chronic stress (human cortisol), and maternal immune activation (human Interleukin-17a; IL17a). Human-derived dorsal forebrain organoids were consequently analyzed via an array of unbiased and high-throughput analytical approaches, including state-of-the-art TMT-16plex liquid chromatography/mass- spectrometry (LC/MS) proteomics, hybrid MS metabolomics, and flow cytometry panels to determine cell-cycle dynamics and rates of cell death. This pipeline subsequently revealed both common and unique proteome, reactome, and metabolome alterations as a consequence of enviromimetic modeling of narcotic use and neuropsychiatric-related risk factors in tissue of human origin.

SUPPLEMENTAL DATA Figure S1. Poly I:C induces IFN-β expression and signaling. Fibroblasts were incubated in media with or without Poly I:C for 24 h. RNA was isolated and processed for microarray analysis. Genes showing >2-fold up- or down-regulation compared to control fibroblasts were analyzed using Ingenuity Pathway Analysis Software (Red color, up-regulation; Green color, down-regulation). The transcripts with known gene identifiers (HUGO gene symbols) were entered into the Ingenuity Pathways Knowledge Base IPA 4.0. Each gene identifier mapped in the Ingenuity Pathways Knowledge Base was termed as a focus gene, which was overlaid into a global molecular network established from the information in the Ingenuity Pathways Knowledge Base. AN ANALYSIS OF APOPTOTIC AND AUTOPHAGIC GENES IN NOVEL DROSOPHILA MODELS OF PARKINSON DISEASE by © Peter Githure M'Angale A Thesis submitted to the School of Graduate Studies in partial fulfillment of the requirements for the degree of Doctor of Philosophy Department of Biology Memorial University of Newfoundland January 2017 St. John's Newfoundland and Labrador Abstract Parkinson disease (PD) is the most common neurodegenerative movement disorder in humans and is strongly associated with the selective loss of dopaminergic (DA) neurons. The clinical features include motor dysfunction, resting tremor and in some cases non- motor features such as autonomic, cognitive and psychiatric disorders. The neuropathologic hallmarks are Lewy bodies and Lewy neurites in surviving neurons which are composed of proteinaceous inclusions comprised of α-synuclein, ubiquitin and other proteins. Pathological mechanisms implicated in PD include aberrant protein aggregation, mitochondrial dysfunction, oxidative stress, and failed cellular processes such as apoptosis, autophagy, proteasomal pathway, and several cellular stressors. To analyse the implication and contribution of some of these cellular processes, we altered the expression of the; classical apoptotic genes namely Buffy and Debcl; autophagy genes Atg6 and Pi3K59F; the antiapoptotic transmembrane Bax inhibitor-1 containing motif (TMBIM) family Bax inhibitor-1, Lifeguard and GHITM; mitochondrial genes such as HtrA2, MICU1, porin/VDAC, Pink1 and parkin; and an increased disease risk gene pyridoxal kinase in DA neurons and in the developing eye. We show that the altered expression of key genes can either be detrimental or beneficial to the health of DA neurons as determined by lifespan and locomotor function, in addition to supportive results obtained from biometric analysis of phenotypes from the neuron rich eye.

ABSTRACT Background Although AKI lacks effective therapeutic approaches, preventive strategies using precondi- tioning protocols, including caloric restriction and hypoxic preconditioning, have been shown to prevent injury in animal models. A better understanding of the molecular mechanisms that underlie the enhanced resistance to AKI conferred by such approaches is needed to facilitate clinical use. We hypothesized that these preconditioning strategies use similar pathways to augment cellular stress resistance. Methods To identify genes and pathways shared by caloric restriction and hypoxic preconditioning, we used RNA-sequencing transcriptome profiling to compare the transcriptional response with both modes of preconditioning in mice before and after renal ischemia-reperfusion injury. Results The gene expression signatures induced by both preconditioning strategies involve distinct com- mon genes and pathways that overlap significantly with the transcriptional changes observed after ischemia-reperfusion injury. These changes primarily affect oxidation-reduction processes and have a major effect on mitochondrial processes. We found that 16 of the genes differentially regulated by both modes of preconditioning were strongly correlated with clinical outcome; most of these genes had not previously been directly linked to AKI.

In experimental sepsis there is prominent apoptosis of various cell types, and genetic manipulation of death and survival pathways has been shown to modulate organ injury and survival. Methodology/Principal Findings: We investigated the effect of extracellular administration of two anti-apoptotic members of the BCL2 (B-cell lymphoma 2) family of intracellular regulators of cell death in a murine model of sepsis induced by cecal ligation and puncture (CLP). We show that intraperitoneal injection of picomole range doses of recombinant human (rh) BCL2 or rhBCL2A1 protein markedly improved survival as assessed by surrogate markers of death. Treatment with rhBCL2 or rhBCL2A1 protein significantly reduced the number of apoptotic cells in the intestine and heart following CLP, and this was accompanied by increased expression of endogenous mouse BCL2 protein. Further, mice treated with rhBCL2A1 protein showed an increase in the total number of neutrophils in the peritoneum following CLP with reduced neutrophil apoptosis. Finally, although neither BCL2 nor BCL2A1 are a direct TLR2 ligand, TLR2-null mice were not protected by rhBCL2A1 protein, indicating that TLR2 signaling was required for the protective activity of extracellularly adminsitered BCL2A1 protein in vivo.
