ABSTRACT This work studied the species composition of the phytoplankton community in three major ports of the Colombian Caribbean (Coveñas, Cartagena and Santa Marta) and three adjacent areas (Cispatá, Baru and Rodadero) during the fourth quarter of 2010 (october). Special attention was given to document the presence of non-native species that currently reside in ports and adjacent areas, in order to relate the degree of invasion to the degree of shipping activity and to collect baseline information for monitoring and management of invasive species. For this purpose, phytoplankton samples were collected by surface tows (23 m mesh size) from 17 replicate sampling points (6 from Cartagena, 6 from Coveñas and 5 from Santa Marta). Species were identified to the lowest taxonomic level possible and they were classified as being either native or non-native. In total, 60 phytoplankton species were identified. These were divided into the following groups: Centric diatoms (31), pennate diatoms (13), dinoflagellates (12), Chlorophyta (1), Cyanophyta (1), Euglenophyta (1) and silicoflagellates (1). It was noted that the most abundant species in both ports and adjacent areas were Skeletonema costatum and several species of the genera Coscinodiscus, Chaetoceros, and Ceratium, which occurred abundantly throughout the study region. No significant differences were found with respect to species richness between ports and their adjacent zones, nor between the three general areas, using the nonparametric Mann Whitney test. Notwithstanding, eight non-native species were identified; the most abundant ones being Bacteriastrum hyalinum, Ceratium furca, Ceratium hirundinella and Ditylum brightwellii. However, these species were found to be present not only in port areas but also in adjacent areas. This could be an indication of a wider effect generated by ballast water discharged in port areas which rapidly spreads to adjacent areas, Furthermore, it is noteworthy that many of the species found in this study corresponded to toxic and harmful species. Key words: Phytoplankton, Port areas, non-native species, ballast water. CONTENIDO 1. INTRODUCCIÓN JUSTIFICADA 1 2. MARCO TEÓRICO Y ESTADO DEL ARTE 5 3. • Ultrastructural Studies of Microalgae
Ultrastructural Studies of Microalgae Alanoud Jaber Rawdhan A thesis submitted for the degree of Doctor of Philosophy (Integrated) October, 2015 School of Agriculture, Food and Rural Development, Newcastle University Newcastle upon Tyne, UK Abstract Ultrastructural Studies of Microalgae The optimization of fixation protocols was undertaken for Dunaliella salina, Nannochloropsis oculata and Pseudostaurosira trainorii to investigate two different aspects of microalgal biology. The first was to evaluate the effects of the infochemical 2, 4- decadienal as a potential lipid inducer in two promising lipid-producing species, Dunaliella salina and Nannochloropsis oculata, for biofuel production. D. salina fixed well using 1% glutaraldehyde in 0.5 M cacodylate buffer prepared in F/2 medium followed by secondary fixation with 1% osmium tetroxide. N. oculata fixed better with combined osmium- glutaraldehyde prepared in sea water and sucrose. A stereological measuring technique was used to compare lipid volume fractions in D. salina cells treated with 0, 2.5, and 50 µM and N. oculata treated with 0, 1, 10, and 50 µM with the lipid volume fraction of naturally senescent (stationary) cultures. There were significant increases in the volume fractions of lipid bodies in both D. salina (0.72%) and N. oculata (3.4%) decadienal-treated cells. However, the volume fractions of lipid bodies of the stationary phase cells were 7.1% for D. salina and 28% for N. oculata. Therefore, decadienal would not be a suitable lipid inducer for a cost-effective biofuel plant. Moreover, cells treated with the highest concentration of decadienal showed signs of programmed cell death. This would affect biomass accumulation in the biofuel plant, thus further reducing cost effectiveness.

  • Molecular Profiling of 18S Rrna Reveals Seasonal Variation and Diversity of Diatoms Community in the Han River, South Korea
Journal46 of Species Research 10(1):4656, 2021JOURNAL OF SPECIES RESEARCH Vol. 10, No. 1 Molecular profiling of 18S rRNA reveals seasonal variation and diversity of diatoms community in the Han River, South Korea Buhari Lawan Muhammad, YeonSu Lee and JangSeu Ki1,* Department of Biotechnology, Sangmyung University, Seoul 03016, Republic of Korea *Correspondent: [email protected] Diatoms have been used in examining water quality and environmental change in freshwater systems. Here, we analyzed molecular profiling of seasonal diatoms in the Han River, Korea, using the hypervariable region of 18S V1V3 rRNA and pyrosequencing. Physicochemical data, such as temperature, DO, pH, and nutrients showed the typical seasonal pattern in a temperate region. In addition, cell counts and chlorophylla, were recorded at high levels in spring compared to other seasons, due to the diatom bloom. Metagenomic analysis showed a seasonal variation in the phytoplankton community composition, with diatoms as the most frequently detected in spring (83.8%) and winter (69.7%). Overall, diatom genera such as Stephanodiscus, Navicula, Cyclotella, and Discostella were the most frequent in the samples. However, a large number of unknown Thalassiosirales diatoms were found in spring (35.5%) and winter (36.3%). Our molecular profiling revealed a high number of diatom taxa compared to morphological observation. This is the first study of diatoms in the Han River using molecular approaches, providing a valuable reference for future study on diatomsbasis environmental molecular monitoring and ecology. Biodiversity Data Journal 9: e59898. https://doi.org/10.3897/BDJ.9.e59898 Abstract Background Freshwater diversity, and diatoms in particular, from Desertas Islands (Madeira Archipelago, Portugal) is poorly known, although the Islands are protected and became a Natural Reserve in 1995. During two field expeditions in 2013 and 2014 to Deserta Grande Island, several freshwater and terrestrial habitats were sampled. The analysis of these samples aims to contribute to the biodiversity assessment of the freshwater biota present in Deserta Grande Island.
