Pennington Dr. Jason McNeill i ABSTRACT Inorganic selenium, oxo-sulfur, and polyphenol compounds are found in foods and dietary supplements, and are recognized for their nutritional benefits and their potential to treat or prevent diseases caused by oxidative stress. In our experiments to determine the effects of inorganic selenium compounds on iron-mediated DNA damage, Na2SeO3 and SeO2 exhibit antioxidant and pro-oxidant activities depending on concentrations of both the compound and hydrogen peroxide. Additional experiments demonstrate that iron coordination is a novel mechanism responsible for the observed + activities. In similar experiments, oxo-sulfur compounds prevent Cu /H2O2-mediated 2+ DNA damage significantly more than DNA damage from Fe /H2O2. UV-vis and gel electrophoresis experiments also confirm that copper coordination is primarily responsible for the DNA damage inhibition, a novel mechanism that extends to all tested sulfur and selenium antioxidants. Electrospray ionization mass spectroscopy indicates that these sulfur and selenium compounds generally bind Cu+ in a 1:1 ratio. Combinations of bioactive components in foods can affect activity of antioxidants. For example, adding one equivalent of caffeine to polyphenols has no effect on DNA damage prevention by epigallocatechin gallate, but significantly decreases the antioxidant ability of quercetin. In addition, DNA damage prevention studies on peach extracts indicate that genetically-modified peach cultivars prevent more DNA damage than unmodified cultivars. In contrast, tetraphenyl-porphyrin-doped conjugated polymer dot nanoparticles cause DNA backbone and base damage upon irradiation, suggesting that these nanoparticles may be efficient photosensitizers for photodynamic therapy i (PDT). Our studies also show H2O2 formation by and iron association with polyethylene glycol (PEG) and PEG-functionalized beads at biologically-relevant concentrations. 2+ Since Fe and H2O2 react to form damaging hydroxyl radical, use of PEG-functionalized nanoparticles in medical applications may cause oxidative stress. Overall, this work has elucidated of antioxidant and pro-oxidant mechanisms of inorganic selenium, oxo-sulfur, and polyphenol compounds, as well as the potential toxicity of functionalized nanomaterials used for PDT and other medical applications. ii DEDICATIONS I dedicate this dissertation to my parents, Phagoo and Gowra Ramoutar, and sisters Reshma and Asha for their encouragement, love and support that allowed me to achieve this measure of academic success. Antimicrobial activity of two enriched saponin fractions obtained from the fruits were also evaluated. The radical scavenging activity of the fractioned extracts was determined spectrophotometrically using 1,1-diphenylpicrylhydrazyl free radical (DPPH). The IC50 of l-ascorbic acid, ethyl acetate and n-butanol fractions from the leaves and ethyl acetate fraction from the fruits were 6.48 mg/mL, 13.26 mg/mL, 27.22 mg/mL, and 285.78 mg/mL, respectively. Total methylxanthine content was 1.16 Æ 0.06 mg/g dry weight in the fruits and 8.78 Æ 0.01 mg/g in the leaves.

Global redox colorimetric assays have commonly been used to estimate the total polyphenol content in foods. However, these assays lack specificity. Contents of individual polyphenols have been determined by chromatography. These data, scattered in several hundred publications, have been compiled in the Phenol-Explorer database. The aim of this paper is to identify the 100 richest dietary sources of polyphenols using this database. Subjects/Methods: Advanced queries in the Phenol-Explorer database (www.phenol-explorer.eu) allowed retrieval of information on the content of 502 polyphenol glycosides, esters and aglycones in 452 foods. Total polyphenol content was calculated as the sum of the contents of all individual polyphenols. These content values were compared with the content of antioxidants estimated using the Folin assay method in the same foods. These values were also extracted from the same database. Amounts per serving were calculated using common serving sizes.

Abstract The antihypertensive effects of aqueous polyphenol-rich extracts of Amaranthusviridis (AV) and Telfairiaocciden- talis (TO) leaves in spontaneously hypertensive rats (SHR) were investigated. The dried vegetable leaves were extracted using 1:20 (leaves:water, w/v) ratio for 4 h at 60 °C. Results showed significantly (P < 0.05) higher polyphenol contents in TO extracts (80–88 mg gallic acid equivalents, GAE/100 mg) when compared with the AV (62–67 mg GAE/100 mg). Caffeic acid, rutin and myricetin were the main polyphenols found in the extracts. The TO extracts had significantly (P < 0.05) higher in vitro inhibition of angiotensin I-converting enzyme (ACE) activity while AV extracts had better renin inhibition. Oral administration (100 mg/kg body weight) to SHR led to significant (P < 0.05) reductions in systolic blood pressure for the AV (−39 mmHg after 8 h)and TO (−24 mmHg after 4 and 8 h).The vegetable extracts also produced significant (P < 0.05) reductions in diastolic blood pressure, mean arterial blood pressure and heart rate when compared to the untreated rats.

Methods: A 40 healthy adult female Wistar albino rats were used in this study. Rats were randomly divided into four groups to determine the effects of curcumin and capsaicin against Cyclophosphamide side effects on the uterus (n=10 in each group); Group 1 was the control group (sham-operated), Group 2 was the cyclophosphamide group, Group 3 was the cyclophosphamide + curcumin (100mg/kg) group, and Group 4 was the cyclophosphamide + capsaicin (0.5 mg/kg) group. Results: Increased tissue oxidative stress and histological damage in the rat uterus were demonstrated due to the treatment of systemic cyclophosphamide chemotherapy alone. The level of tissue oxidant and antioxidant markers and histopathological changes were improved by the treatment of curcumin and capsaicin.

Se(IV) exhibited the lowest antioxidant properties in comparison to other selenium compounds in all assays. The highest reducing power was obtained for SeMet, while the highest ability to scavenging DPPH radicals for MeSeCys. The results obtained experimentally for the mixtures containing selenium species and green or black tea infusion were compared with theoreti- cal values calculated by adding up the effects of both individual components analyzed separately. The results obtained from each assay clearly show that observed effect is not additive. In almost every case the theoretical value of antioxidant capac- ity was significantly higher from that obtained from the activity of the binary mixture of black tea infusion with selenium compound decreased in the order: SeMet > Se(IV) > Se(VI) > MeSeCys, while for similar mixtures with green tea infusion: MeSeCys > Se(VI) > SeMet ~ Se(IV). Colorectal cancer is the third most prevalent cancer worldwide and the most common diet-related cancer, influenced by diets rich in red meat, low in plant foods and high in saturated fats. Observational studies have shown that fruit and vegetable intake may reduce colorectal cancer risks, although the precise bioactive components remain unclear. This review will outline the evidence for the role of polyphenols, glucosinolates and fibres against cancer progression in the gastrointestinal tract. Those bioactive compounds are considered protective agents against colon cancer, with evidence taken from epidemiological, human clinical, animal and in vitro studies. Various mechanisms of action have been postulated, such as the potential of polyphenols and glucosinolates to inhibit cancer cell growth and the actions of insoluble fibres as prebiotics and the evidence for these actions are detailed within. In addition, recent evidence suggests that polyphenols also have the potential to shift the gut ecology in a beneficial manner. Such actions of both fibre and polyphenols in the gastrointestinal tract and through interaction with gut epithelial cells may act in an additive manner to help explain why certain fruits and vegetables, but not all, act to differing extents to inhibit cancer incidence and progression.

There are also no reports available on the polyphenols content of rubber-processing effluent. The objective of this study is to determine the presence of polyphenol compounds in latex C-serum and effluent using Fourier-transform infrared spectroscopy (FTIR) analysis. This study also aims to profile specific phenolics using High Performance Liquid Chromatography (HPLC). Results from the FTIR analysis showed the presence of polyphenols in both latex and effluent. The optimal method for determining polyphenol by HPLC was determined which uses methanol and 0.5% acetic acid as the mobile phases. Several polyphenol peaks, including gallic acid, naphtoic acid, quercetin, chlorogenic acid and rutin, were detected in both latex and effluent when compared to authentic polyphenol standards.

A better knowledge of the factors that influence the action of PPO is imperative in order to control and manipulate its detrimental activity in plant products. This paper presents an overview of the current understanding of the reaction properties, biochemical characteristics and potential physiological roles of PPO in plants. Reaction properties will include general PPO reactions, specificities and molecular mechanisms of these reactions, and methods available to assess PPO activity. Physicochemical properties will evaluate substrate specificity, environ- mental influences such as pH and temperature, multiplicity, latency, and activators and inhibitors of PPO.
