Abstract: This study aimed at determining the in vitro antibacterial activity of a clinically achievable marbofloxa- cin (MAR) concentration against the clinical isolate S. pseudintermedius in an in vitro dynamic model simulat- ing the in vivo pharmacokinetics of dogs. The in vitro PK/PD (pharmacokinetic/pharmacodynamic) model that mimics the single daily doses of MAR (half-life, 8 h) was simulated. An inoculum (108 cfu/ml) of clinical isolate S. pseudintermedius (MIC = 0.0625 μg/ml) was exposed to monoexponentially decreasing concentrations of MAR with simulated AUC24 h/MIC varied from 34.81 h to 696.15 h. Every two hours, the multiple sample colony forming units were determined. The result of this study demonstrated that the clinically achieved MAR concentrations at AUC24 h/MIC ratios of 348.08 and 696.15 h produced a pronounced reduction in the bacterial counts and pre- vented the re-growth of the clinical isolate S. pseudintermedius. However, further study, considering the strains with different susceptibility levels, is recommended. ZENIQUIN- marbofloxacin tablet Zoetis Inc. ---------- Zeniquin® (marbofloxacin) Tablets For oral use in dogs and cats only CAUTION: Federal law restricts this drug to use by or on the order of a licensed veterinarian. Federal law prohibits the extralabel use of this drug in food-producing animals. DESCRIPTION Marbofloxacin is a synthetic broad-spectrum antibacterial agent from the fluoroquinolone class of chemotherapeutic agents. Marbofloxacin is the non-proprietary designation for 9-fluoro-2,3-dihydro-3- methyl-10-(4-methyl-1-piperazinyl)-7-oxo-7H-pyrido[3,2,1-ij][4,1,2] benzoxadiazine-6-carboxylic acid. The empirical formula is C17H19FN4O4 and the molecular weight is 362.36. The compound is soluble in water; however, solubility decreases in alkaline conditions. The N-octanol/water partition coefficient (Kow) is 0.835 measured at pH 7 and 25°C. Figure 1: Chemical structure of marbofloxacin CLINICAL PHARMACOLOGY Marbofloxacin is rapidly and almost completely absorbed from the gastrointestinal tract following oral administration to fasted animals. Divalent cations are generally known to diminish the absorption of fluoroquinolones. The effects of concomitant feeding on the absorption of marbofloxacin have not been determined. (See Drug Interactions.) In the dog, approximately 40% of an oral dose of marbofloxacin is excreted unchanged in the urine1. Excretion in the feces, also as unchanged drug, is the other major route of elimination in dogs. Ten to 15% of marbofloxacin is metabolized by the liver in dogs. In vitro plasma protein binding of marbofloxacin in dogs was 9.1% and in cats was 7.3%. In the cat, approximately 70% of an oral dose is excreted in the urine as marbofloxacin and metabolites with approximately 85% of the excreted material as unchanged drug.

ABSTRACT Pseudomonas aeruginosa has been determined to be the distinct cause of a number of different infections in both humans and animals. Apart from its high intrinsic resistance, which makes it difficult to treat, it also has a great capacity to acquire further resistance mechanisms. These mechanisms can be present simultaneously in one cell, conferring a multiresistant phenotype. Taking into account that the known risk factor for selection of resistant strains is excessive and inappropriate drug use, especially fluoroquinolones, the aim of this study was to determine the susceptibility of P. aeruginosa isolates to these antibacterial agents. The susceptibility of 90 canine isolates was determined by disk diffusion susceptibility test and the broth microdilution method. Both methods showed that isolates were significantly more sensitive to ciprofloxacin than to marbofloxacin or enrofloxacin, and more sensitive to marbofloxacin than to enrofloxacin. The results of microdilution and disk diffusion testing were in 98.9% agreement for ciprofloxacin and in 91.1% agreement for marbofloxacin, with no statistically significant difference between the two methods (P>0.05) for these two antibiotics.

Food Safety Commission of Japan Risk Assessment Report on Marbofloxacin (Veterinary Medicines) Food Safety Commission of Japan (FSCJ) August, 2007 Summary A risk assessment was conducted on marbofloxacin, a new quinolone antibacterial agent effective for gram-negative bacteria and many species of gram-positive bacteria. The assessment was conducted based on data obtained from test and experiments on animal metabolism and residual properties (in rats, dogs, swine animals and bovine animals), acute toxicity (in mice and rats), subchronic toxicity (in rats and dogs), reproductive development toxicity (2-generation reproduction in rats), teratogenicity (in rats and rabbits), and from studies on genotoxicity and microbiological effects. Genotoxicity, effects on reproduction, and teratogenicity were not found. Although chronic toxicity/carcinogenicity studies have not been conducted, no carcinogenicity has been reported in quinolone agents in general, and marbofloxacin is considered to have no genotoxicity that can be a concern for human health. Accordingly, it is determined that it is possible to set an ADI without conducting carcinogenicity studies. New quinolone antibacterial agents are used for human clinical treatments. Distinctive side effects include joint disturbance in immature animals and phototoxicity. A value was settled for the NOAEL for the joint effects of marbofloxacin in a study using beagle dogs. Phototoxicity is considered not strong due to its structural properties. Accordingly, intake of marbofloxacin through food is considered to have negligible phototoxicity to human health provided that it is used under proper management. The lowest NOAEL obtained from various studies was 4 mg/kg bw/day in a 13-week subchronic toxicity study using rats or dogs.

ABVP (Reptiles and Amphibians) Abstract The standard of today0s reptile practice calls on clinicians to use an ever-increasing array of diagnostic tools to gather information and obtain a definitive diagnosis. Few, if any, pathognomonic signs exist for reptile diseases, and for most clinical syndromes there is a lack of information regarding pathophysiology for one to define standard therapeutic protocols based solely on clinical signs without objective diagnostic information. For example, in the relatively distant past, clinicians treating reptile patients would routinely administer parenteral calcium to green iguanas (Iguana iguana) with the primary presenting clinical sign of muscle tremors. Today, veterinarians who treat reptiles recognize that the risk of soft tissue mineralization and permanent damage to arteries, renal tubules, and other tissues usually outweighs the potential short-term benefit of calcium therapy. Before calcium therapy is initiated, it is best to know the patient0s ionized calcium concentration to reduce the risk of potential adverse therapeutic side effects. A problem-oriented diagnostic approach directed toward minimizing risk and maximizing therapeutic benefit is now the standard of reptile practice. Copyright 2014 Elsevier Inc. All rights reserved.
