We demonstrate that in R. prolixus following infection with epimastigotes of Trypanosoma cruzi clone Dm28c and, in comparison with uninfected control insects, the midgut contained (i) fewer bacteria, (ii) higher parasite numbers, and (iii) reduced nitrite and nitrate production and increased phenoloxidase and antibacterial activities. In addition, in insects pre-treated with antibiotic and then infected with Dm28c, there were also reduced bacteria numbers and a higher parasite load compared with insects solely infected with parasites. Furthermore, and in contrast to insects infected with Dm28c, infection with T. cruzi Y strain resulted in a slight decreased numbers of gut bacteria but not sufficient to mediate a successful parasite infection. We conclude that infection of R. prolixus with the T. cruzi Dm28c clone modifies the host gut immune responses to decrease the microbiota population and these changes are crucial for the parasite development in the insect gut. -

The Chagas disease parasite, Trypanosoma cruzi, was found in all three species. Their natural infection rates were similar in the first two species (20.6%; 19.1%) and slightly lower in T. nitida (13.8%). However there was no significant difference in the infection rates in the three species (p = 0.131). T. dimidiata males have higher infection rates than females (p = 0.030), whereas for R. prolixus there is no difference in infection rates between males and females (p = 0.114). The sex ratios for all three species were significantly skewed. More males than females were found inside houses for T. dimidiata (p < 0.0001) and T. nitida (p = 0.011); a different pattern was seen for R. prolixus (p = 0.037) where more females were found. Sex ratio is proposed as an index to show the mobility of T. -

SELECTION, EXPRESSION AND VACCINATION WITH RECOMBINANT RHODNIUS PROLIXUS ANTIGENS by ALICE CHARLOTTE SUTCLIFFE (Under the Direction of Donald E. Champagne) ABSTRACT Triatomine (Hemiptera:Reduviidae) control is an important aspect of reducing the spread of Trypanosma cruzi, the causative agent of Chagas disease, for which no vaccine currently exists. The development of a triatomine vaccine would increase the number of prevention and control methods available. Here, I describe the selection of twelve Rhodnius prolixus proteins, their recombinant expression and use in vaccine trials in mice as well as the investigation of function of one of these protein targets. Overall, my results do not establish biological function for this protein target and they indicate that induction of an antibody responses can occur in mice vaccinated with recombinant R prolixus proteins but this was not a sufficient protective response. Six out of 84 dwellings (7%) surveyed in a rural village were infested (78 bugs/infested domicile; 279 bugs were collected in a single dwelling). Precipitin tests revealed R. ecuadoriensis fed on birds (65%), rodents (31%), marsupials (8%), and humans (15%) – mixed bloodmeals detected in 37.5% of individual samples. Trypanosoma cruzi from opossums and rodents may thus be introduced into the domestic cycle. Wasp parasitoidism was detected in 6.5% of 995 R. ecuadoriensis eggs (only in peridomestic habitats). Control strategies should integrate insecticide spraying (in- doors and peridomestic), better management of poultry, and housing improvements. A possible inefficacy of Malathion is reported. As important vectors, triatomine bugs have attracted ongoing attention, and, thus, various aspects of their systematics, biology, ecology, biogeography, and evolution have been studied for decades. In the present paper the authors summarize the current knowledge on the biology, ecology, and systematics of these vectors and discuss the implications for human health. Studies conducted in river Ererê located in the left margin of Negro River, municipality of Barcelos, state of Amazonas, have confirmed that Rhodnius brethesi has as its natural habitat the palm tree Leopoldinia piassaba. By scanning electron microscopy, sensillum type was studied on the antennae of R. brethesi. The specimens used come from the field and laboratory colony. No differences were observed between R. brethesi and other Triatominae studied. In the R. brethesi antennas, differences were observed only between the antennal segments and in the dorsal and ventral portions. -

Levels of genetic variation and population structure were determined for 181 Triatoma sordida insects from four populations of southeastern Brazil, through the analysis of 28 allozyme loci. None of these loci presented fixed dif- ferences between any pair of populations, and only two revealed polymorphism, accounting for low levels of heterozygos- < ity ( He = 0.027), and low genetic distances ( D 0.03) among populations. FST and Contingency Table results indicated the existence of genetic structure among populations ( FST = 0.214), which were incompatible with the isolation by distance model (Mantel test: r = 0.774; P = 0.249).
