    CHARACTERIZATION OF MALUS GENOTYPES WITHIN THE USDA-PGRU MALUS GERMPLASM COLLECTION FOR THEIR POTENTIAL USE WITHIN THE HARD CIDER INDUSTRY A Thesis Presented to the Faculty of the Graduate School of Cornell University In Partial Fulfillment of the Requirements for the Degree of Master of Science by Nathan Carey Wojtyna August 2018 © 2018 Nathan Carey Wojtyna ABSTRACT In the United States, hard cider producers lack access to apple genotypes (Malus ×domestica Borkh. and other Malus species) that possess higher concentrations of tannins (polyphenols that taste bitter and/or astringent) and acidity (described as having a sharp taste) than what is typically found in culinary apples. Utilizing the USDA-PGRU Malus germplasm collection, two projects were conducted to address these concerns. The first project characterized fruit quality and juice chemistry for a target population of 308 accessions with the goal of identifying accessions with desirable characteristics for hard cider production. The second project used the same sample population to explore the use of the Ma1 and Q8 genes as potential markers to predict the concentration of titratable acidity of cider apples. An initial target population of 308 accessions were identified and 158 accessions were assessed in 2017 for external and internal fruit characteristics along with juice chemistry. As per the Long Ashton Research Station (LARS) cider apple classification system where apples with tannin concentration (measured with the Löwenthal Permanganate Titration method) greater than 2.0 g×L-1 are classified as bitter, and those with a malic acid concentration greater than 4.5 g×L-1 are classified as sharp, 29% of the 158 accessions would be classified as bittersweet, 13% bittersharp, 28% sweet (neither bitter nor sharp), and 30% sharp.

Apple cultivars can be maintained for centuries in plant collections through grafting, and some are thought to date as far back as Roman times. Molecular markers provide a means to reconstruct pedigrees and thus shed light on the recent history of migration and trade of biological materials. The objective of the present study was to identify relationships within a set of over 1400 mostly old apple cultivars using whole-genome SNP data (~ 253 K SNPs) in order to reconstruct pedigrees. Results: Using simple exclusion tests, based on counting the number of Mendelian errors, more than one thousand parent-offspring relations and 295 complete parent-offspring families were identified. Additionally, a grandparent couple was identified for the missing parental side of 26 parent-offspring pairings. Among the 407 parent-offspring relations without a second identified parent, 327 could be oriented because one of the individuals was an offspring in a complete family or by using historical data on parentage or date of recording. Parents of emblematic cultivars such as ‘Ribston Pippin’, ‘White Transparent’ and ‘Braeburn’ were identified.

    Southern Plant Lists Southern Garden History Society A Joint Project With The Colonial Williamsburg Foundation September 2000 1 INTRODUCTION Plants are the major component of any garden, and it is paramount to understanding the history of gardens and gardening to know the history of plants. For those interested in the garden history of the American south, the provenance of plants in our gardens is a continuing challenge. A number of years ago the Southern Garden History Society set out to create a ‘southern plant list’ featuring the dates of introduction of plants into horticulture in the South. This proved to be a daunting task, as the date of introduction of a plant into gardens along the eastern seaboard of the Middle Atlantic States was different than the date of introduction along the Gulf Coast, or the Southern Highlands. To complicate maters, a plant native to the Mississippi River valley might be brought in to a New Orleans gardens many years before it found its way into a Virginia garden. A more logical project seemed to be to assemble a broad array plant lists, with lists from each geographic region and across the spectrum of time. The project’s purpose is to bring together in one place a base of information, a data base, if you will, that will allow those interested in old gardens to determine the plants available and popular in the different regions at certain times. This manual is the fruition of a joint undertaking between the Southern Garden History Society and the Colonial Williamsburg Foundation. In choosing lists to be included, I have been rather ruthless in expecting that the lists be specific to a place and a time.

Apple trees that have fruit on spurs or semi-spurs are more dwarfing. They also require special pruning techniques. Tip and Partial-tip Apple Varieties – Over 350 varieties listed. Fruit are borne on the tip of the branches, and are weeping and require little to no pruning. ________________________________________________________________________ Apple Fruiting 1 12/8/06 SPUR-TYPE FRUITING APPLES FOR THE HOME ORCHARD For home orchardists there are several advantages in growing spur–type trees. As the name indicates, the fruit is borne on spurs. Spurs are slow growing leafy shoots and have a mixed terminal bud. A mixed terminal bud will produce shoot and flowers. In apples, spurs develop on two–year old shoots from axillary buds located at the base of each leaf. Axillary buds on a spur can give rise to shoots or new spurs. A branched spur system forms after several years when new spur form on old spurs. Spur–type strains are more dwarfing than the standard stain. When spur and standard strains were compared in Washington rootstock trials, the spurs were 25% smaller than standard stains. Spur–type apples have a growing and fruiting characteristic in which lateral (axillary) buds on two year old wood gives rise to a higher portion of spurs and fewer lateral shoots than occur with standard growth habits. This gives the tree a more open canopy and compact growth habit than standard trees. Research indicates that they have approximately half the canopy volume of standard strains.
