  • An Integrative DNA Barcoding Framework of Ladybird Beetles (Coleoptera: Coccinellidae)
    www.nature.com/scientificreports OPEN An integrative DNA barcoding framework of ladybird beetles (Coleoptera: Coccinellidae) Weidong Huang1,2, Xiufeng Xie3, Lizhi Huo4, Xinyue Liang1,2, Xingmin Wang2 & Xiaosheng Chen1,2 ✉ Even though ladybirds are well known as economically important biological control agents, an integrative framework of DNA barcoding research was not available for the family so far. We designed and present a set of efcient mini-barcoding primers to recover full DNA barcoding sequences for Coccinellidae, even for specimens collected 40 years ago. Based on these mini-barcoding primers, we obtained 104 full DNA barcode sequences for 104 species of Coccinellidae, in which 101 barcodes were newly reported for the frst time. We also downloaded 870 COI barcode sequences (658 bp) from GenBank and BOLD database, belonging to 108 species within 46 genera, to assess the optimum genetic distance threshold and compare four methods of species delimitation (GMYC, bPTP, BIN and ABGD) to determine the most accurate approach for the family. The results suggested the existence of a ‘barcode gap’ and that 3% is likely an appropriate genetic distance threshold to delimit species of Coccinellidae using DNA barcodes. Species delimitation analyses confrm ABGD as an accurate and efcient approach, more suitable than the other three methods. Our research provides an integrative framework for DNA barcoding and descriptions of new taxa in Coccinellidae. Our results enrich DNA barcoding public reference libraries, including data for Chinese coccinellids. This will facilitate taxonomic identifcation and biodiversity monitoring of ladybirds using metabarcoding. As species are a fundamental biological category, accurately identifying them is an essential premise of biological studies.

Dr. Weis hat seine Unterrichtsverpflichtung sehr ernst genommen und war ein ausge- zeichneter Biologielehrer. So hat ihm die Unterrichtsvorbereitung kaum Zeit gelassen, sich weiter der besonders arbeitsaufwendigen Untersuchung der Aphiden zuzuwenden. Die Verbundenheit mit der Natur, der Flora und Fauna, ganz besonders der Arthropoden- fauna, war ihm aber stets eine Selbstverständlichkeit. So hat er sich sowohl im Unter- richt, als auch besonders bei Exkursionen, Schulausflügen und Schullandwochen der Tierwelt, besonders der Kleintierwelt Oberösterreichs angenommen. Seine besondere Zuwendung galt dabei den Käfern, sodaß aus dieser, sich über mehrere Jahrzehnte er- streckenden Sammeltätigkeit schließlich eine Käfersammlung erwuchs, die u.a.

In many countries, they are well established for environmental impact assessment, but even in the well-studied Central European fauna, species identification can be very difficult. A comprehensive and taxonomically well-curated DNA barcode library could remedy this deficit and could also link hundreds of years of traditional knowledge with next generation sequencing technology. However, such a beetle library is missing to date. This study provides the globally largest DNA barcode reference library for Coleoptera for 15 948 individuals belonging to 3514 well-identified species (53% of the German fauna) with representatives from 97 of 103 families (94%). This study is the first comprehensive regional test of the efficiency of DNA barcoding for beetles with a focus on Germany. Sequences ≥500 bp were recovered from 63% of the specimens analysed (15 948 of 25 294) with short sequences from another 997 specimens.

Saproxylic insects represent an important part of forest biodiversity and have functionally significant role in the forest ecosystems. They are species-rich organisms that depend upon the dead or dying wood or upon the presence of other saproxylic species. In this study we investigated whether stump harvesting affects saproxylic beetle assemblages in the clear-cut of managed forest in Latvia. We used pitfall traps and window traps for sampling beetles in the clear-cut sites with removed and retained stumps. Overall, these two sampling sites shared a similar number of beetle species. However, there were significant differences between two sampling methods with more species from window traps. Saproxylic species were more abundant in site with retained stumps.     Proceedings of the XXIII Nordic Meeting of Entomology The XXIII Nordic Meeting of Entomology was held in Turku, Finland, on 24--27 July 1994. The participants who gave a presentation in the meeting were informed that they could publish papers based on these presentations in a special issue of Entomolo gica Fennica. This is the issue which includes the papers submitted and accepted within the allowed time. As these articles are based on the meeting presentations, the dates of receipt and acceptance of the manuscripts are not given (i.e. the dates of "publication" are, in fact, the meeting presentation dates). During the research periods (2000-2006) in Naujene rural municipality Daugavpils district 165 beetles species were found that belon to 28 families, from which 5 species are protected in Latvia. We had seen an incredible total of 928 identified species (a record for this tour) including 390 higher plants and 371 invertebrate species, of which a superb 110 were butterflies (tour record) and 73 moths (tour record) as well as 35 dragonflies (another tour record), 36 bugs, 29 beetles etc. It is always difficult to find birds on this tour, being rather late in the season. The extremely high temperatures at the start of the week did not help as birds sort cover, but we nonetheless managed a creditable total of 97 species, including some not often seen on this tour. Day 1 Saturday 27th July Fly Heathrow to Sofia; drive to Melnik Sunny on arrival in Sofia, hot, with maximum temperature 27°C It was not the most auspicious of starts as our flight was delayed for more than an hour 'due to overbooking'. In actual fact it transpired that this was because the aircraft being used was smaller than the one originally scheduled and luggage also had to be moved.
