BACTERIAL CELL MEMBRANE Dr. Rakesh Sharda Department of Veterinary Microbiology NDVSU College of Veterinary Sc. & A.H., MHOW CYTOPLASMIC MEMBRANE ➢The cytoplasmic membrane, also called a cell membrane or plasma membrane, is about 7 nanometers (nm; 1/1,000,000,000 m) thick. ➢It lies internal to the cell wall and encloses the cytoplasm of the bacterium. ➢It is the most dynamic structure of a prokaryotic cell. Structure of cell membrane ➢The structure of bacterial plasma membrane is that of unit membrane, i.e., a fluid phospholipid bilayer, composed of phospholipids (40%) and peripheral and integral proteins (60%) molecules. ➢The phospholipids of bacterial cell membranes do not contain sterols as in eukaryotes, but instead consist of saturated or monounsaturated fatty acids (rarely, polyunsaturated fatty acids). ➢Many bacteria contain sterol-like molecules called hopanoids. ➢The hopanoids most likely stabilize the bacterial cytoplasmic membrane. ➢The phospholipids are amphoteric molecules with a polar hydrophilic glycerol "head" attached via an ester bond to two non-polar hydrophobic fatty acid tails. ➢The phospholipid bilayer is arranged such that the polar ends of the molecules form the outermost and innermost surface of the membrane while the non-polar ends form the center of the membrane Fluid mosaic model ➢The plasma membrane contains proteins, sugars, and other lipids in addition to the phospholipids. ➢The model that describes the arrangement of these substances in lipid bilayer is called the fluid mosaic model ➢Dispersed within the bilayer are various structural and enzymatic proteins, which carry out most membrane functions. ➢Some membrane proteins are located and function on one side or another of the membrane (peripheral proteins). -

Interpretation and evaluation of precursor ion scanning chromatograms (urine profile recognition) In the field of anti-doping control, chromatograms from target steroid-analysis are generally evaluated and interpreted visually by the analyst which is trained for this purpose. A similar approach will be adopted for evaluating the precursor scanning chromatograms. To get acquainted with the chromatograms, around 50 blank samples will be analysed. In a second step precursor ion scan chromatograms obtained from controlled administration studies (e.g. methyltestosterone, methanedienone,…) and from adverse analytical findings from routine target screening will also be investigated to get acquainted with positive samples. The applicability of instrument software for this purpose will be evaluated. B. Application to real samples Urine-samples will be analysed. These samples will include all out of competition samples and both out of competition samples and in competition samples C. Study of the influence of different population factors in the detectability of different markers for several anabolic steroids A single dose of the previously studied steroids (stanozolol and methyltestosterone) will be administered to six volunteers belonging to different population groups. Urine will be collected for three weeks and a method including all feasible markers will be applied. -

PHYTOSTEROLS, PHYTOSTANOLS AND THEIR ESTERS Chemical and Technical Assessment 1 Prepared by Richard Cantrill, Ph.D., reviewed by Yoko Kawamura, Ph.D., for the 69th JECFA 1. Summary Phytosterols and phytostanols, also referred to as plant sterols and stanols, are common plant and vegetable constituents and are therefore normal constituents of the human diet. They are structurally related to cholesterol, but differ from cholesterol in the structure of the side chain. Commercially, phytosterols are isolated from vegetable oils, such as soybean oil, rapeseed (canola) oil, sunflower oil or corn oil, or from so-called "tall oil", a by-product of the manufacture of wood pulp. These sterols can be hydrogenated to obtain phytostanols. Both phytosterols- and stanols, which are high melting powders, can be esterified with fatty acids of vegetable (oil) origin. The resulting esters are liquid or semi-liquid materials, having comparable chemical and physical properties to edible fats and oils, enabling supplementation of various processed foods with phytosterol- and phytostanol esters. The most common phytosterols and phytostanols are sitosterol (3β-stigmast-5-en-3ol; CAS Number 83- 46-5), sitostanol (3β,5α-stigmastan-3-ol; CAS Number 83-45-4), campesterol (3β-ergost-5-en-3-ol; CAS Number 474-62-4), campestanol (3β,5α-ergostan-3-ol; CAS Number 474-60-2), stigmasterol (3β- stigmasta-5,22-dien-3-ol; CAS Number 83-48-7) and brassicasterol (3β-ergosta-5,22-dien-3-ol; CAS Number 474-67-9). Each commercial source has its own typical composition. Dietary intake of phytosterols ranges from 150-400 mg /day in a typical western diet. -

We present a comparative study of the effects of cholesterol and lanosterol on molecular conformational order and phase equilibria of lipid-bilayer membranes. By using deuterium NMR spectroscopy on multilamellar lipid-sterol systems in combination with Monte Carlo simulations of microscopic models of lipid-sterol interactions, we demonstrate that the evolution in the molecular chemistry from lanosterol to cholesterol is manifested in the model lipid-sterol membranes by an increase in the ability of the sterols to promote and stabilize a particular membrane phase, the liquid-ordered phase, and to induce collective order in the acyl-chain conformations of lipid molecules. -

ABSTRACT Extraction of phytosterols as lipid substances from various plant sources, including corn, is a subject of many foreign publications. However, studies that identify the behavior and influence of phytosterols in the beer making process were not found in the literature. Phytosterols present in cereals as free sterols, fatty acid esters and phenolic acids, glycosides and acylated glycosides. The study showed that phytosterols’ content is entirely dependent on the raw material used. The more a malt part in the grain, the higher is phytosterol content. Phytosterol content accumulates in beer due to the extraction of raw materials from unmalted grain products, malt and hops.
