HOCKEY ABSTRACT A method for the isolation of grey sterile fungi from brown root rot infected tomato root systems was developed. Semi-sel ecti ve media significantly reduced the growth of Colletotriohum ooooodes and catyptetla campa~ula with little effect on the growth of grey sterile fungi • Pycni di a characteri sti c of pyrenochaeta lyoopel'sioi were formed on VS-Juice agar (VSA) by twelve of the 19 grey sterile fungal i sol ates tested. A method for the routi ne producti on of pycni di a/ coni di a was developed: P. lyoopersioi cul tures, i nocul ated onto V8A are i ncubated at 220 C with a 16h bl ack light photoperi od • No vegetable constituent of V8-Juice, tested individually, could be shown to be solely responsible for sporulation on VSA. Conidi a requi re a temperature range of 20 to 26lC, pH range 5.0 to S.O and external nutrients to achieve germination levels qreater than 9at. Conidial germination decreased with age. Incubation of P. tycopersici coni di a ina dil ute ci rrus extract and under di fferent light regimes did not affect germination. Conidia were shown to swell prior to germination and produce either single or dual germ tubes. Using mycelial i nocul urn, grey steril e fungal 1501 ates caused lesions on tomato roots in vitro and i~ vivo. Conidia of P. tyoopersici produced 1esi ons on the roots of tomatoes grown in i nocul ated steril i sed compost and unsteril ised garden soil, withi n six weeks. Brown root rot and corky root symptoms were produced on tomatoes grown comnerct all yin soil s , infested with P. tyoopersioi coni di a, for up to 21 weeks. Root 1esi ons were produced on pl ants grown in conidia infested sterilised compost and unsterilised garden soil which had been stored at room temperature for up to 510 and 150 days respectively. Conidial germination was observed in soils, on cellophane and on polycarbonate membranes. On cellophane, coni di a germinated in unsterilised and sterilised composts forming sclerotia on a II frond" type myceli urn after three to seven days i ncubati on. Conidial germination was lower in washed sand and greatly reduced in peat. Germinati on on pol ycarbonate membranes in compost was slower and sclerotial formation reduced compared to that observed on cellophane. A soil assay has been developed to estimate soil infestation 1evel s of brown root rot (BRR). Using the BRR assay, various species of bacteria and Streptomycetes were shown to reduce the number of 1esi ons on tomato pl ants grown in soil infested with brown root rot. Three locations within Gezira State were selected namely the vicinity of Almusallamih Tayiba, Wad Al ataya and Hamdalnil and located at North, central and south of the State respectively. The results showed that the local variety was found to be highly susceptible to the disease than the exported of the hybrid ones. The highest disease incidence was recorded in Hamdalnil (16.8%) while the lowest disease incidence was recorded at Wad Al ataya(9.23%). Koch’s postulates were performed to prove that the fungus isolated Pyrenochaeta terrestris was the causal agent of the pink root rot on onion plants.

They are implicated in the production of antibiotic and other compounds of therapeutic importance. Ecologically they provide several benefits to plants, including protection from plant pathogens. There have been numerous studies on the biodiversity and ecology of endophytic fungi. Some taxa dominate and occur frequently when compared to others due to adaptations or capabilities to produce different primary and secondary metabolites. It is therefore of interest to examine different fungal species and major taxonomic groups to which these fungi belong for bioactive compound production. In the present paper a list of endophytes based on the available literature is reported. More than 800 genera have been reported worldwide. Dominant genera are Alternaria, Aspergillus, Colletotrichum, Fusarium, Penicillium, and Phoma. Most endophyte studies have been on angiosperms followed by gymnosperms. Among the different substrates, leaf endophytes have been studied and analyzed in more detail when compared to other parts. Most investigations are from Asian countries such as China, India, European countries such as Germany, Spain and the UK in addition to major contributions from Brazil and the USA.

In order to stabilize the placement of the genus and family, an epitype for the type species was designated after obtaining ITS sequence data and conducting maximum likelihood and Bayesian phylogenetic analyses. Iodosphaeria foliicola occurring on overwintered Alnus sp. leaves is described as new. Five species in the genus form a well-supported monophyletic group, sister to thePseudosporidesmiaceae in the Xylariales. Selenosporella-like and/or ceratosporium-like synasexual morphs were experimentally verified or found associated with ascomata of seven of the nine accepted species in the genus.

Fungal pathogens that infect roots receive less attention than those infecting the aerial parts of crops despite their significant impact on plant growth and fruit production. Results: We assembled a 54.9Mb P. lycopersici draft genome sequence based on Illumina short reads, and annotated approximately 17,000 genes. The P. lycopersici genome is closely related to hemibiotrophs and necrotrophs, in agreement with the phenotypic characteristics of the fungus and its lifestyle. Several gene families related to host–pathogen interactions are strongly represented, including those responsible for nutrient absorption, the detoxification of fungicides and plant cell wall degradation, the latter confirming that much of the genome is devoted to the pathogenic activity of the fungus. We did not find a MAT gene, which is consistent with the classification of P. lycopersici as an imperfect fungus, but we observed a significant expansion of the gene families associated with heterokaryon incompatibility (HI). Conclusions: The P. lycopersici draft genome sequence provided insight into the molecular and genetic basis of the fungal lifestyle, characterizing previously unknown pathogenic behaviors and defining strategies that allow this asexual fungus to increase genetic diversity and to acquire new pathogenic traits.

Phoma heteromorphospora, type species of Phoma section Heterospora, and its allied species Phoma dimorphospora, are transferred to the genus Heterospora stat. nov. The Phoma acuta complex (teleomorph Leptosphaeria doliolum), is revised based on a multilocus sequence analysis of the LSU, ITS, small subunit 18S nrDNA (SSU), β-tubulin (TUB), and chitin synthase 1 (CHS-1) regions.

This species has recently come under threat from the disease white pine blister rust (WPBR). White pine blister rust is caused by the pathogenic fungus Cronartium ribicola, which was inadvertently introduced into North America from Europe in the early 1900’s, and has since spread widely with devastating impacts (Burns et al., 2008). In North America, WPBR is largely lethal to five-needle pine species. In Colorado, WPBR has been found in stands of Rocky Mountain bristlecone pine and limber pine (Pinus flexilis), and efforts have been made to identify trees with increased resistance to the disease. The USDA Forest Service Rocky Mountain Research Station has identified specific trees that harbor some level of heritable resistance to WPBR, versus those appearing fully susceptible (Schoettle, 2004; Schoettle et al., 2012; Schoettle et al., 2014). Essentially all plants in the wild harbor endophytic bacteria and fungi, which are defined as co-existing in plant tissues without causing evidence of disease, and it is increasingly appreciated that endophytes can alter plant responses to both biotic and abiotic stresses ii (Rodriguez et al., 2008).
