    PROFILE Profile of Nahum Sonenberg PROFILE Ann Griswold, Science Writer Five years after the final shots of World War II rang out, a child and his father stood on the coast of the Mediterranean Sea. From their vantage point in a cemetery in the town of Jaffa, Israel, the boy gazed across the sea and envisioned stepping onto the sands of a distant shore. He pointed to the horizon and asked his father what lay beyond. “I remember my father said, ‘There is America, the country where everything is possible,’” says Nahum Sonenberg, the Gilman Cheney Professor of Biochem- istry at McGill University in Montreal. Sonenberg, elected as a foreign associate to the National Academy of Sciences in 2015, has spent nearly five decades looking beyond the horizon to map un- explored territory in molecular biology. His research has uncovered the cellular control knobs of protein synthesis and revealed how this process drifts off-course in cancer, obesity, diabetes, and neurological diseases. His achievements have garnered numerous hon- ors. Sonenberg is a fellow of the Royal Society of London and Canada, Foreign Member of the Amer- ican Academy of Arts and Sciences and the American Association for the Advancement of Science, Asso- ciate Member of European Molecular Biology Orga- nization, and an Officer of the Order of Canada. He has received numerous prizes in the biological sciences, including the Lewis S. M5G 1X8, Canada b Center for Metabolic Disease, University of Leuven, Leuven, Belgium Received 8 February 1999; accepted 3 May 1999 Abstract The carbohydrate-deficient glycoprotein syndromes (CDGS) are a group of autosomal recessive multisystemic diseases characterized by defective glycosylation of N-glycans. This review describes recent findings on two patients with CDGS type II. In contrast to CDGS type I, the type II patients show a more severe psychomotor retardation, no peripheral neuropathy and a normal cerebellum. The CDGS type II serum transferrin isoelectric focusing pattern shows a large amount (95%) of disialotransferrin in which each of the two glycosylation sites is occupied by a truncated monosialo-monoantennary N-glycan. Fine structure analysis of this glycan suggested a defect in the Golgi enzyme UDP-GlcNAc:K-6-D-mannoside L-1,2-N-acetylglucosaminyltransferase II (GnT II; EC 2.4.1.143) which catalyzes an essential step in the biosynthetic pathway leading from hybrid to complex N-glycans. GnT II activity is reduced by over 98% in fibroblast and mononuclear cell extracts from the CDGS type II patients. Direct sequencing of the GnT II coding region from the two patients identified two point mutations in the catalytic domain of GnT II, S290F (TCC to TTC) and H262R (CAC to CGC).

    Biography: David Naylor ● President, University of Toronto, since October 1, 2005. ● Dean of Medicine and Vice Provost, Relations with Health Care Institutions, at the University of Toronto since 1999. As dean, academic and administrative leadership role in Canada's largest multi-disciplinary health science faculty (2,000 full-time academic appointees). As Vice Provost, responsible for relations with nine fully-affiliated and eleven partly-affiliated hospitals. Also facilitated partnerships with various community and governmental partners. ● Formerly a Senior Scientist of the Medical Research Council of Canada (MRC), co-author of approximately 300 scholarly publications. Scholarly work spans social history, public policy, epidemiology and biostatistics, and health economics, as well as clinical and health services research in most fields of medicine. ● MD with honours from the University of Toronto in 1978 with scholarships in medicine, surgery and paediatrics. ● Rhodes Scholar, 1979 at Hertford College, Oxford University. Earned D.Phil. in 1983, for thesis work in the Sub-Faculty of Sociology, Department of Social and Administrative Studies. ● Trained in general internal medicine at the University of Western Ontario (Fellow of the Royal College of Physicians and Surgeons of Canada, 1986), followed by a year in Toronto as an MRC fellow in clinical epidemiology. ● Joined the academic staff of the Faculty of Medicine at the University of Toronto in 1987-88. ● From 1990 to 1996 developed and led a new research program in clinical epidemiology at Sunnybrook Health Science Centre in Toronto. ● From 1991 to 1998 developed, and was inaugural Chief Executive Officer of, the Institute for Clinical Evaluative Sciences (ICES), a non-profit corporation that continues to support Canada's largest multi-disciplinary grouping of academics and trainees dedicated to healthcare research.
