La realització d’aquesta tesi va acompanyada de moltes circumstàncies i persones. Quan contactes amb persones proposant-los idees i projectes, moltes vegades un es pot trobar que la porta es tanca. A totes aquelles portes que es van tancar agraeixo la seva honestedat perquè gràcies a elles s’ha pogut fluir vers la consolidació d’aquest projecte. Gràcies, moltes gràcies. Més enllà de les idees calen persones que en facilitin la seva realització i en aquest punt han estat determinants certes persones. Aquesta tesi respon a la voluntat d’una persona que quan la vaig contactar no va dir que NO. Aquesta persona és el Dr Joan Vendrell i Ortega, sense ell no hi hagués hagut projecte, sense ell no hi hauria respostes. Gràcies, moltes gràcies. UNIVERSITAT ROVIRA I VIRGILI EVIDÈNCIES EN TEIXIT ADIPÓS D'ALTERACIONS GÈNIQUES DELS VASOS LIMFÀTICS EN L'OBESITAT Alfons Horra Pueyo Les raons que ens fan viure són moltes i potser moltes són, a ulls d’alguns, irrellevants: un cafè amb llet en una cafeteria, un somriure entre amics, un llibre que dóna respostes a qüestions no resoltes, una abraçada, un petó, unes paraules... Un llibre, el cervell i el sentit de la vida del Paul Thagard, em va il·lustrar sobre que el sentit de la vida estava en l’amor, la feina i el joc. Reflexions que comparteixo. L’amor: l’amor a la vida, a les persones, a la ciència, a la feina, al plaer de l’art, de la música, de la pintura... Possiblement l’amor no defineix el seu sentit en l’objecte del nostre amor sinó en la voluntat de sentir amor. Agraeixo a la meva família per haver-me fet créixer en l’amor a la llibertat; llibertat de pensar, estimar i de viure. M’heu ensenyat el més important que es pot ensenyar a ningú, possiblement des de la inconsciència d’haver-ho fet. M’heu ensenyat que poc importa el que pensis; el més important és que ho facis des de l’honestedat i el respecte. La llibertat no s’ensenya sinó que s’exerceix, i ho heu fet molt bé. M’heu ensenyat l’amor en la seva vessant humana, m’heu ensenyat voluntat, perseverança, raó i dubte. M’heu ensenyat tantes coses ! Gràcies, moltes gràcies. La feina: la meva feina ha donat i dóna sentit a la meva vida en tant que em fa créixer. A nivell personal, pel diàleg amb els meus pacients i, professionalment, pel que aprenc de les seves dolences. Dels meus pacients, n’aprenc els matisos de la natura, del seu sentit de desordre i d’equilibri. Saviesa continguda que sempre és un gran gaudi descobrir. Gràcies, moltes gràcies. El joc: el joc és el sentit més bàsic del nostre creixement com a individus. El vivim en la infantesa des de la seva espontaneïtat i el vivim en tant que adults des de la seva racionalitat. To date, there are no ideal drugs for treating obesity. Studies have shown that activation of brown adipose tissue (BAT) can promote energy consumption and inhibit obesity, which makes browning of white adipose tissue (WAT) a potential therapeutic target for obesity. Our objective was to identify genes and molecular pathways associated with WAT and the activation of BAT to WAT browning, by using publicly available data and computational tools; this knowledge might help in targeting relevant signaling pathways for treating obesity and other related metabolic diseases. Results: In this study, we used text mining to find out genes related to brown fat and white fat browning. Combined with biological process and pathway analysis in GeneCodis and protein-protein interaction analysis by using STRING and Cytoscape, a list of high priority target genes was developed. The Human Protein Atlas was used to analyze protein expression. Candidate drugs were derived on the basis of the drug-gene interaction analysis of the final genes. Our study identified 18 genes representing 6 different pathways, targetable by a total of 33 drugs as possible drug treatments. The final list included 18 peroxisome proliferator-activated receptor gamma (PPAR-γ) agonists, 4 beta 3 adrenoceptor (β3-AR) agonists, 1 insulin sensitizer, 3 insulins, 6 lipase clearing factor stimulants and other drugs.

This was done using various approaches such as IDH1 and IDH2 mutation analysis, MGMT methylation analysis, copy number analysis using array comparative genomic hybridisation and identification of differentially expressed miRNAs using miRNA microarray analysis. IDH1 mutation was present at a frequency of 71% in low grade glioma and was identified as an independent marker for improved OS in a multivariate analysis, which confirms the previous findings in low grade glioma studies.

Programming of several brain regions in males depends upon aromatase conversion of testosterone to estrogen. However, it is not clear the direct contribution that Y chromosome associated genes, especially sex- determining region Y (Sry), might exert on brain sexual differentiation in therian mammals. Two species of spiny rats: Amami spiny rat (Tokudaia osimensis) and Tokunoshima spiny rat (T. tokunoshimensis) lack a Y chromosome/Sry, and these individuals possess an XO chromosome system in both sexes. Both Tokudaia species are highly endangered. To assess the neural transcriptome profile in male and female Amami spiny rats, RNA was isolated from brain samples of adult male and female spiny rats that had died accidentally and used for RNAseq analyses. Results: RNAseq analyses confirmed that several genes and individual transcripts were differentially expressed between males and females. In males, seminal vesicle secretory protein 5 (Svs5) and cytochrome P450 1B1 (Cyp1b1) genes were significantly elevated compared to females, whereas serine (or cysteine) peptidase inhibitor, clade A, member 3 N (Serpina3n) was upregulated in females. Victoria University of Wellington 2017. i ABSTRACT Non-alcoholic fatty liver disease (NAFLD) is a manifestation of the metabolic syndrome in the liver. It is marked by hepatocyte accumulation of triacylglycerol (TAG) rich lipid droplets. In some patients, the disease progresses to non-alcoholic steatohepatitis (NASH), characterized by cellular damage, inflammation and fibrosis. In some cases, cirrhosis and liver failure may occur. However, the pathogenesis of NAFLD is still unclear. The present project is based on the hypothesis that hepatocytes are equipped with mechanisms that allow them to manage lipid accumulation to a certain extent. Continued or increased lipid accumulation beyond this triggers molecular mechanisms such as oxidative stress, lipid peroxidation and cell death that aggravate the condition and cause disease progression. The aim of this project is to study the effects of lipid accumulation on the cells using proteomics approach to identify proteins involved in the disease progression. A cell culture model was used in the study. HepG2 cells, a human liver carcinoma cell line, were treated with a mixture of fatty acids (FA) to induce lipid accumulation. The lipid accumulation in HepG2 cells was measured with Oil red O assay and the effect of lipid accumulation on the proliferation of the cells was measured using an MTT cell proliferation assay. HepG2 cells treated with 1 mM FA mixture for 6 hours induced lipid accumulation 1.4 times of control with 90% of cell proliferation capacity of the control cells.

Among these, “extended haplotype homozygosity” (EHH) is a reliable approach to detect genome regions under recent selective pressure. The objective of this study was to use this approach to identify regions that are under recent positive selection and shared by the most representative Italian dairy and beef cattle breeds. Results: A total of 3220 animals from Italian Holstein (2179), Italian Brown (775), Simmental (493), Marchigiana (485) and Piedmontese (379) breeds were genotyped with the Illumina BovineSNP50 BeadChip v.1. After standard quality control procedures, genotypes were phased and core haplotypes were identified. The decay of linkage disequilibrium (LD) for each core haplotype was assessed by measuring the EHH. Since accurate estimates of local recombination rates were not available, relative EHH (rEHH) was calculated for each core haplotype. Genomic regions that carry frequent core haplotypes and with significant rEHH values were considered as candidates for recent positive selection. Candidate regions were aligned across to identify signals shared by dairy or beef cattle breeds. Overall, 82 and 87 common regions were detected among dairy and beef cattle breeds, respectively.

Ernst3, Hasan Khatib1 and Guilherme JM Rosa1,4* Abstract Background: Joint modeling and analysis of phenotypic, genotypic and transcriptomic data have the potential to uncover the genetic control of gene activity and phenotypic variation, as well as shed light on the manner and extent of connectedness among these variables. Current studies mainly report associations, i.e. undirected connections among variables without causal interpretation. Knowledge regarding causal relationships among genes and phenotypes can be used to predict the behavior of complex systems, as well as to optimize management practices and selection strategies. Here, we performed a multistep procedure for inferring causal networks underlying carcass fat deposition and muscularity in pigs using multi-omics data obtained from an F2 Duroc x Pietrain resource pig population. Results: We initially explored marginal associations between genotypes and phenotypic and expression traits through whole-genome scans, and then, in genomic regions with multiple significant hits, we assessed gene-phenotype network reconstruction using causal structural learning algorithms. One genomic region on SSC6 showed significant associations with three relevant phenotypes, off-midline10th-rib backfat thickness, loin muscle weight, and average intramuscular fat percentage, and also with the expression of seven genes, including ZNF24, SSX2IP,andAKR7A2. The inferred network indicated that the genotype affects the three phenotypes mainly through the expression of several genes.
