Editor in charge: Ralf Britz Abstract Psilorhynchus kamengensis, new species, is described from a stream of the Kameng River, a northern tributary to the Brahmaputra drain- age, Arunachal Pradesh, northeast India. The new species belongs to the P. balitora species group and can be distinguished from all other members of this group by the presence of a longer post-epiphyseal fontanelle, 3 unbranched anal-fn rays, 9 + 8 caudal-fn rays, 35 vertebrae and fn coloration. The validity of the species is also corroborated by molecular phylogenetic analysis based on the mitochondrial cyt b gene. Key words Torrent minnow, Kameng, morphology, cyt b, phylogeny. Introduction Cypriniform fshes of the genus Psilorhynchus McClel- The Kameng River is the major drainage in East and land, 1838 are characterized by having an arched dor- West Kameng districts of Arunachal Pradesh, northeast sum with fattened ventral surface, horizontally-placed India. One hundred and five species belonging to nine order and 29 families were recorded during the study. It has been recorded that 52 species have ornamental value of which 31 are consumed by the local people as food and other 21 species have only the ornamental importance. After extensive ex-situ study in the laboratory 25 species were recorded as the potential ornamental fishes and other 27 were excluded because of various factors that need extensive study to acclimatize them for the aquarium habitat. 12 exotic fishes were also recorded during the survey from the Coochbehar district of which 3 were ornamental fishes. Molecular Diversity of Freshwater Fishes of Bangladesh Assessed by Dna Barcoding

Sagir Ahmed 1*, Sujan Kumar Datta 2 and Ayesha Akhter Zhilik 1 Department of Zoology, University of Dhaka, Dhaka1000, Bangladesh ABSTRACT: This study represents the comprehensive molecular identification of freshwater fishes of Bangladesh based on a fragment of the cytochrome c oxidase subunit I (COI) gene in the mitochondrial genome. A total of 315 mitochondrial COI barcode sequences were obtained from 153 species,114 genera, 49 families and 16 orders of fishes. The mean length of the sequences was 652 base pairs. For all the samples, %G was significantly lower compared to the other nucleotides and %GC was lower compared to %AT ( p-value ˂ 0.05). Also, a significantly lower %GC content was observed in second and third codon position compared to the first one in all the samples (1 st >2 nd >3 rd , p-value ˂ 0.05). The average K2P distances within species, genera, families and orders were 0.38%, 7.02%, 12.75% and 18.68%, respectively. The mean interspecific distance was 18-fold higher than the mean intraspecific distance. The K2P neighbor-joining (NJ) trees based on the sequences generally clustered species according to their taxonomic position. A total of 12 species were newly recorded in Bangladesh. High efficiency in species identification were demonstrated in the present study by DNA barcoding, and concluded that COI sequencing can be used as an authentic identification marker for freshwater fish species. Kullander 2 We sequenced the standard DNA barcode gene fragment in 694 newly collected specimens, Received: 21 November 2018 representing 243 species level Operational Barcode Units (OBUs) of freshwater fshes from Bangladesh. Accepted: 3 June 2019 We produced coi sequences for 149 out of the 237 species already recorded from Bangladesh. Another Published online: 28 June 2019 83 species sequenced were not previously recorded for the country, and include about 30 undescribed or potentially undescribed species. Several of the taxa that we could not sample represent erroneous records for the country, or sporadic occurrences. Species identifcations were classifed at confdence levels 1(best) to 3 (worst). We propose the new term Operational Barcode Unit (OBU) to simplify references to would-be DNA barcode sequences and sequence clusters. We found one case where there were two mitochondrial lineages present in the same species, several cases of cryptic species, one case of introgression, one species yielding a pseudogene to standard barcoding primers, and several cases of taxonomic uncertainty and need for taxonomic revision. Large scale national level DNA barcode prospecting in high diversity regions may sufer from lack of taxonomic expertise that cripples the result. Consequently, DNA barcoding should be performed in the context of taxonomic revision, and have a defned, competent end-user. Fish and fsheries play an important role in Bangladesh’s economy, nutrition and culture. With 47 609 km2 of inland water bodies, it is the third inland fsh producing country in the world afer China and India1. -
