Bilaterians have eight functionally distinct Voltage-gated K+ channel subfamilies: Shaker, Shab, Shaw, Shal, KCNQ, Eag, Erg and Elk. These subfamilies are defined by sequence conservation, functional properties as well as subfamily-specific assembly. Genome searches revealed metazoan-specificity of these gene families and the presence of prototypic voltage-gated K+ channels in a common ancestor of ctenophores (comb jellies) and parahoxozoans (bilaterians, cnidarians and placozoans). Establishment of the gene subfamilies, however happened later in a parahoxozoan ancestor. Analysis of voltage- gated K+ channels in a cnidarians species Nematostella vectensis (sea anemone) unveiled conservation in functional properties with bilaterian homologs. Phosphoinositide (most notably PIP2) regulation of ion channels is universal in eukaryotes. PIP2 modulates Shaker, KCNQ and Erg channels in distinct manners, while PIP2 regulation of Elk channels has not been reported.

Gao 1,2,4 ✉ Antibody-dependent enhancement (ADE) is an important safety concern for vaccine development against dengue virus (DENV) and its antigenically related Zika virus (ZIKV) because vaccine may prime deleterious antibodies to enhance natural infections. Cross-reactive antibodies targeting the conserved fusion loop epitope (FLE) are known as the main sources of ADE. We design ZIKV immunogens engineered to change the FLE conformation but preserve neutralizing epitopes. Single vaccination conferred sterilizing immunity against ZIKV without ADE of DENV-serotype 1–4 infections and abrogated maternal–neonatal transmis- sion in mice. Unlike the wild-type-based vaccine inducing predominately cross-reactive ADE-prone antibodies, B cell profiling revealed that the engineered vaccines switched immunodominance to dispersed patterns without DENV enhancement. The crystal structure of the engineered immunogen showed the dimeric conformation of the envelope protein with FLE disruption. We provide vaccine candidates that will prevent both ZIKV infection and infection-/vaccination-induced DENV ADE. IKV is a mosquito-borne pathogen belonging to the genus models12,13,19,20. More importantly, a recent study of pediatric cohorts Flavivirus in the family Flaviviridae1. The 2015–2016 ZIKV in Nicaragua clearly shows that previous ZIKV infection enhanced epidemic spread to 84 countries worldwide, including future risk of DENV2 disease and severity, as well as DENV3 sever- Z2,3 21,22 China .

Neurosci. Abstr. 5:585; 1980:Nature, 283;673). This was seen as a voltage-de- pendent, subthreshold, non-inactivating potassium current that had a dramatic braking action on repetitive action potential discharges. It was dubbed “M-current” because it was inhibited by muscarine, acting on muscarinic ace- tylcholine receptors (mAChRs), thereby increasing action potential discharges. M-current is an imperfect descriptor because the current can also be inhibited by activating other Gq-type G protein-coupled receptors (GPCRs) and mus- carine itself only works if the cell has M1, M3 or M5 receptors, not M2 or M4 (Robbins et al., 1991: Eur J Neurosci. 3: 820). The molecular composition of the M-channel was revealed some two decades later by David McKinnon and his colleagues as a combination of theKCNQ2 and KCNQ3 gene products Kv7.2 and Kv7.3 (Wang et al., 1998: Science. 282: 1890), probably as a 2+2 heteromer (Hadley et al, 2003: J Neurosci. 23: 5012).     Geochemistry and Transport of Uranium- Bearing Dust at Jackpile Mine, Laguna, New Mexico By Reid Douglas Brown Submitted in Partial Fulfillment of the Requirements for the Masters of Science in Hydrology New Mexico Institute of Mining and Technology Department of Mechanical Engineering Socorro, New Mexico (August 2017) ABSTRACT Closed mines pose significant risks to the environment and human health. Uranium mine contamination of surface water, groundwater and soil have received moderate attention, but few studies have investigated dust transport of uranium. The latter has immediate implications for remediation efforts and environmental/human health regulators. Frequent dust storms intensify aeolian transport of uranium in arid settings. At the Jackpile Mine in Laguna Pueblo, New Mexico, 15 sets of dust traps have been installed at heights of 0.25 m, 0.5 m, 1.0 m and 1.5 m above the soil surface. Some of these traps are within the mine pit, while others are up to 4 km away; dust from these sites was collected every two months. In addition, soil samples from each site were collected and sieved into eight size classes. All samples were acid digested, and the uranium content analyzed using Inductively Coupled Plasma Mass Spectrometry. We investigate whether uranium has an affinity for a particular particle size class, with interest centered on particles small enough to be completely inhaled by humans. Results show that surface concentrations of uranium vary substantially across the landscape. Distance from the pit shows no correlation with uranium in the upper 5 cm of soil. Other factors appear to control accumulation, such as vegetation height and density and topographic relief, which are known to have a significant impact on wind speeds, soil erosion and dust deposition.

We studied a human Kv10.2 channel mutation (R327H) recently identified in a child with epileptic encephalopathy and autistic features. Using multistate structural modeling, we demonstrate that the Arg327 residue in the S4 helix of voltage-sensingdomainhasstrongionicinteractionswithnegativelychargedresidueswithintheS1–S3helicesintheresting(closed)and early-activation state but not in the late-activation and fully-activated (open) state. The R327H mutation weakens ionic interactions between residue 327 and these negatively charged residues, thus favoring channel opening. Voltage-clamp analysis showed a strong hyperpolarizing(ϳ70mV)shiftofvoltagedependenceofactivationandanaccelerationofactivation.Ourresultsdemonstratethecritical role of the Arg327 residue in stabilizing the channel closed state and explicate for the first time the structural and functional change of a Kv10.2 channel mutation associated with neurological disease.
