6 2016 Report to Our Donors Dr. Tak Mak changed the world of medical research in 1984 when he published a landmark scientific paper on the cloning of the T-cell receptor gene, considered the ‘holy grail’ of the human immune system. He is the Director of The Campbell Family Institute for Breast Cancer Research, and was invited to the Vatican this year, along with several other world-leading cancer researchers, to take part in a conference that focused in part on immunotherapy.

Dr. Pamela Ohashi is the Co-Director of the Tumor Immunotherapy Program (TIP). Her laboratory has made key contributions to our understanding of the immune system, and she spearheaded the establishment of the first Adoptive T-cell Therapy clinical trial in Canada.

Dr. Lillian Siu is a senior staff medical oncologist and a recognized world leader in the management of Phase I and II clinical trials. She is Co-Director of the Tumor Immunotherapy Program, Co-Director of the BRAS Drug Development Program, and Executive Director of the Cancer Genomics Program. Together, the three programs of tumor immunotherapy, drug development and cancer genomics form a triad of synergy that supports The Princess Margaret’s core vision to deliver Personalized Cancer Medicine.

Dr. Marcus Butler is a senior medical oncologist and Director of the Immune Monitoring Laboratory. Before joining The Princess Margaret, Dr. Butler was an Instructor in Medicine at Harvard Medical School and a Clinical Fellow in Medicine at the Dana-Farber Cancer Institute. As a clinician-scientist, Dr. Butler plays a leadership role in translating the discoveries in our laboratories into clinical trials that have the potential to improve outcomes for patients.

A gift of $50 million pledged in early 2013 by Emmanuelle Gattuso, Allan Slaight and the Slaight family was a huge boost to our BILLION DOLLAR CHALLENGE. It has enabled Princess Margaret Cancer Centre to continue building the scientific team needed to lead the way in defining and implementing Personalized Cancer Medicine.

“Recruiting scientists in new fields like bioinformatics, epigenetics and immunotherapy has enabled us to expand the breadth of our research program,” explains Dr. Bradly Wouters, Research Director at Princess Margaret Cancer Centre. “Even more importantly, with these highly- specialized scientists collaborating with the world-renowned cancer researchers already here, we are uniquely equipped to tackle challenging research problems that haven’t been attempted anywhere before. There is an incredible level of synergy and excitement that has been created through these new recruits.”

Dr. David Brooks is the Research Chair and a senior scientist specializing in immunotherapy. In particular, he is focused on what causes our immune system to fail in attacking cancer or persistent viral infections. Before joining The Princess Margaret, Dr. Brooks was a tenured Associate Professor and led a laboratory at the University of California, Los Angeles. “Moving my laboratory and my life to a new country was an exciting (and daunting) endeavor. The Gattuso Slaight gift has made my transition into cancer research a reality, and I am truly excited about what the future holds.”

Dr. Steven Chan is a clinician-scientist who is also part of the Leukemia Program at The Princess Margaret. Dr. Chan’s research focuses on how mitochondria (which produce energy for cells) functions differently in cancer cells than in normal cells. Dr. Chan received his medical and research training at Stanford University in California. “Philanthropic support has allowed me to set up a new research program in a top-ranked research institution. The generosity of Emmanuelle Gattuso and the Slaight Family is a source of inspiration and motivation for my team to work harder to achieve the goal of personalized medicine.”

“We believe that immunotherapy is and will continue to be a giant leap forward in the treatment of cancer,” says Tony Traub. Tony is an entrepreneur who has spent much of his career in the financial services industry. Together with his wife Wendy, they recognized that The Princess Margaret has a world class team in the area of immunotherapy, and wanted to help move their initiatives forward. Tony and Wendy are excited about the potential that lies ahead with Personalized Cancer Medicine, and they want to be part of making it a reality. They decided this year to make a significant investment to help accelerate the Tumor Immunotherapy Program at Princess Margaret Cancer Centre, bringing their total giving to $1 million. Wendy is currently a patient at the cancer centre. She was diagnosed with melanoma in July 2014—the same month in which they lost a dear friend to brain cancer. Wendy’s melanoma was quickly removed, but it recurred. Today, she is in the care of Dr. Marcus Butler, a medical oncologist, and his team. Wendy is receiving immunotherapy and being cared for by the staff at The Princess Margaret. The Princess Margaret Cancer Foundation 11 Investing in more effective treatment for ovarian cancer patients

Janet Cottrelle worked as a high school history teacher for many years and then made a successful transition to the financial services industry. As an investment advisor for RBC Dominion Securities, her priority was to design unique wealth solutions for her clients with their lifestyle in mind. Janet’s own lifestyle includes riding horses, working in the garden and enjoying rural life. She was a serious horse jumping competitor, and is in the Jump Canada Hall of Fame. But three cancer diagnoses have made Janet very aware that our own personal vision for the future can be rudely interrupted. She has been treated twice for breast cancer, and is now being treated for ovarian cancer. Having had both breast and ovarian cancer, Janet is keenly aware of the difference in public awareness and the level of research funding between the two diseases. She says, “Given a five-year survival rate of 45 percent, I was disappointed to find out ovarian cancer research was seriously underfunded. There is an urgent need for procedures to diagnose ovarian cancer earlier and for more effective treatments.” In recognition of the important work going on at The Princess Margaret, the Janet D. Cottrelle Foundation is making a donation of $1.26 million to support ovarian cancer research. The goal of the Foundation is to fund initiatives that will improve women’s health, and the first initiative to be supported is a study led by Janet’s physician, Dr. Amit Oza, that will create a framework for anticipating newly-diagnosed ovarian cancer patients’ resistance to platinum-based chemotherapy drugs. Dr. Oza is a world leader in clinical research for ovarian cancer, and the efforts of his team have led to two new drug therapies being approved in the past five years.

Making ovarian cancer treatment more effective by making it more personal

One of the current challenges in treating ovarian cancer is understanding a patient’s resistance to platinum-based chemotherapy drugs. Dr. Oza believes a biological ‘snapshot’ of each patient’s resistance pattern to platinum chemotherapy is urgently needed. This snapshot would offer physicians very valuable information on which to base their treatment recommendations. He believes that this snapshot could be formulated in three ways: • Creating a genetic profile of each patient’s tumor sample • Conducting liquid biopsies that test for circulating tumor DNA in a patient’s blood over the course of their treatment • Building an immune profile to assess a patient’s suitability for immunotherapy treatment

In 1937, two brothers, Sam and Sid Sniderman, created a record department in their family’s store, Sniderman Radio Sales & Service, on College Street. That little department was the spark that led to the creation of a Toronto landmark—Sam the Record Man—that lasted over four decades at 347 . The Princess Margaret now has a very special connection to ‘Sam’s’ thanks to a transformational gift from Sid’s daughter Lana. Like her father, Lana worked in the back office specializing in franchising, leasing and investments. She had a very strong work ethic, and she also had passions that she pursued avidly outside of working hours. She owned a farm in Nobleton, and there she became a Lana Sniderman successful dog breeder, specializing in Dobermans. And then there was horse racing and horseback riding. Lana’s love of thoroughbred horses and her compassion for animals was evident through her long-time and generous support of the LongRun Thoroughbred Retirement Society—an organization devoted to arranging good care for retired racehorses. Her long-time friend, Barbara Jacobs, spoke about how loyal Lana was to her close friends and staff. “She was a private person, but very generous to the people and causes that were important to her.” Now people with cancer are benefiting from Lana’s compassion and generosity. She had her own battles with cancer, and, in gratitude for wonderful care that she received at Princess Margaret Cancer Centre, she made a generous bequest in her will of over $2 million. Just as Lana worked hard to build and sustain a successful business, her legacy will be hard at work in the quest to conquer cancer.

First estate planning seminar delivered in Cantonese

In March 2016, at the encouragement of David Dai and Lydia Cheung, financial consultants with Investors Group, The Princess Margaret Cancer Foundation held its first estate planning seminar in Cantonese. Held at the Hilton Toronto/ Markham Suites, over 60 attendees heard from four different advisors—a lawyer, an accountant, and two financial consultants—about the importance of having a Canadian will and a Power of Attorney. The speakers also discussed ways to minimize tax, while protecting and distributing assets to one’s estate. Several people attending shared with us that they did not know they could leave a gift to a charity in their will, and they were very grateful for the opportunity to hear about the benefits of doing so in their own language. 掌握未來,還看今天。

Fighting cancer with viruses (or agents that can act like viruses) Dr. Daniel De Carvalho and his team are doing pioneering work using therapeutic agents that mimic viruses, in order to trigger a response from the immune system that will destroy cells infected with the virus. There is tremendous potential for these agents to work in combination with another class of drugs called immune checkpoint inhibitors. Immune checkpoint inhibitors remove the brakes from our immune system to allow it to attack ‘foreign’ substances in the body, including viruses.

14 2016 Report to Our Donors Invest in Research provides cancer researchers with ‘seed funding’

In 2007, Tom Ehrlich asked an important question of his physician, Dr. Mary Gospodarowicz: What can I do to help that will have impact? She said there was a real need for seed funding to support new cancer research projects. In response, Tom (currently the Chair of the Board of Directors for The Princess Margaret Cancer Foundation) started the Invest in Research program. This program brings together people interested in making directed investments to help conquer cancer. With boardroom-style accountability, the group annually considers a number of proposals and awards one or two researchers with grants of $100,000 each, to move their ideas forward. Each member of Invest in Research has a vote and helps to choose the projects that are funded. The grant Tom Ehrlich and Michael Wilson recipients later report back to the group on their progress. To date, Invest in Research has raised more than $2 million for projects that improve our understanding of cancer at the molecular level and advance new drug, radiation and surgical therapies. Recipients of these grants have leveraged their seed-funding into more than $30 million of additional philanthropic, government or corporate funding for their projects. “I look for meaning in my investments and Invest in Research really struck an emotional chord. GIPPEC has a productive first year The Global Institute of Psychosocial, Palliative & End-of-Life Care (GIPPEC) is a partnership formed in 2014 between University Health Network, Princess Margaret Cancer Centre and the University of Toronto. The Director of GIPPEC is Dr. Gary Rodin, Head of Supportive Care at Princess Margaret Cancer Centre. The past year and a half has been very productive for GIPPEC’s executive team as they have: • Built up the academic faculty to foster collaborative international research teams, including accomplished leaders in end-of-life care from Africa, Italy, Germany, Great Britain and China • Organized a seminar series and a large conference that brought together thought leaders and investigators contributing to public policy regarding the care of patients with life-threatening or terminal disease • Launched an online educational platform that supports graduate training and enhances research skills The work and ambitions of GIPPEC were highlighted and supported this year at an event organized by Prime Quadrant, a firm providing financial investment services for affluent Canadians.

Stem-cell scientists, led by Dr. John Dick, have discovered a completely new understanding of how human blood is made. For people with blood disorders and diseases, including leukemia and lymphoma, the potential benefits of this discovery are significant, unlocking a distinct route to personalizing therapy.

Dr. Rama Khokha and her team discovered that blocking the master regulator of bone renewal stops osteosarcoma, the most common primary bone cancer in children and teens. Osteosarcoma is the disease that proved fatal for Canadian icon Terry Fox.

In March 2016, Dr. Jonathan Irish, Chief of Surgical Oncology, gave a tour of Canada’s most technologically-advanced cancer surgical suite via Periscope—a social media tool that connects to Twitter accounts and allows viewers to ask questions in real-time. Over 550 participants tuned in and asked questions about the technology demonstrated and the benefits to patients.

In April 2016, Dr. Gang Zheng, a Senior Scientist and expert in nanoparticles connected with over 3,200 interested participants on the discussion website Reddit. His science forum on the site’s ‘Ask me Anything’ page ranked 14th that day for ‘most talked about things on the Internet’. His lab has discovered how to get colourful porphyrins (used by plants in photosynthesis) to self-assemble into biodegradable nanoparticles called ‘porphysomes’ which target cancer. Once they invade the cancer, the now- coloured tumors can absorb laser light, heating and killing the tumor, and sparing healthy cells.

In May 2016, Dr. Geoffrey Liu and his team of cancer researchers made a discovery in the treatment of colorectal cancer. They identified a marker, found in blood tests, that determines which patients would benefit from receiving a drug called cetuximab. The research solved an ongoing mystery of why receiving cetuximab was ineffective for up to half of incurable colorectal cancer patients who did not respond to treatment.

Dr. Amit Oza is the new Head of Medical Oncology and Hematology for Princess Margaret Cancer Centre. He is an internationally-renowned clinician scientist and also holds the positions of Medical Director of the Cancer Clinical Research Unit and Co-Director of the BRAS Drug Development Program at Princess Margaret Cancer Centre. Dr. Oza was instrumental in establishing the Princess Margaret Phase II Consortium—a pan-Canadian cancer research enterprise that has conducted more than 120 clinical trials. The 4,853 participants in the 2015 Enbridge Ride to Conquer Cancer raised an incredible $19.3 million.

The Enbridge Ride to Conquer Cancer provides everyone with the opportunity to positively impact future outcomes for cancer patients. That is why Team Erin has been riding since year one, and has raised over $2 million. Erin was a mother, wife, daughter and beloved sister who beat back pancreatic cancer for 6.5 years. She received aggressive treatment at The Princess Margaret and went into remission for several years. In 2008, just as the Ride to Conquer Cancer was being launched, she relapsed. But Erin showed her spirit and found the strength to complete the Ride for two years along with her sister Denise and brother Noah. The Ride will always be about Erin for Denise and Noah, but they have grown their team to over 60, with many others having stories just like theirs. Sharing stories motivates and inspires their fundraising efforts. “No one ever thought our ‘little team that could’, which started with only six, would be one of the top performing Ride teams year after year,” says Denise. “I think our success comes from a collection of passion, hard work, grief and hope. We believe we can make an impact.”

Helping the Ride move

“We can do more.” These are the words that fundraisers dream about hearing from their donor community. And when Cameron Fowler, Group Head, Canadian Personal & Commercial Banking for BMO Financial Group, said ‘we’, he didn’t mean just BMO; he meant all the banks, and, in fact, the entire financial services industry. In his leadership role as Honorary Chair of this year’s Enbridge Ride to Conquer Cancer, Cam led the charge with the financial services industry. First, he held a Banker’s Breakfast to bring together leaders from the major banks, and he challenged them to ramp up their fundraising efforts for The Princess Margaret by exceeding their rider and fundraising goals from the prior year.

Alison looked forward to riding with her own yellow flag

Everyone has a story, and many of them bring tears to your eyes. That’s a huge part of what binds participants in the Enbridge Ride to Conquer Cancer. And some stories, by virtue of their timing or their telling, take on the quality of a legend and, ultimately, become part of the folklore. The story of Alison Salinas is going to be one of those. Alison—young, full of life and thoughtful of others—was diagnosed in 2013 with stage 1 cervical cancer, and her boyfriend, Tyler, participated in the 2014 Ride to support her fight. Alison was determined to participate herself in the 2015 Ride sporting a yellow flag along with other cancer survivors, but a recurrence of her cancer took that opportunity from her. Instead, she shared her story on stage at camp. In her speech, Alison spoke about how she remained hopeful that thanks to the funds raised by the Ride, better treatment options would become available. Sadly, she passed away five months later. Even riders who love cycling need inspiration to set ambitious goals and become fundraisers for cancer research. For the thousands of people at camp that night, Alison Salinas provided it. An inspiring presentation at this event by the head of surgical oncology for The Princess Margaret, Dr. Jonathan Irish, helped everyone appreciate how the Ride funds are improving outcomes for cancer patients. Cam Fowler also called the industry back together prior to the Ride to inspire them to do more. He offered a personal incentive to teams, and challenged them to beat their fundraising goals, and he engaged Air Miles and MasterCard to bring great prize incentives to all riders fundraising for the event. The success of Cam’s call to action was evident in the increased participation and inspiration to do more. And under Cam’s leadership, Team BMO raised more than $500,000. Cam noted that cancer touches everyone in some way, directly or indirectly, and that “Princess Margaret Cancer Centre is doing outstanding work and leading us to a place where cancer is controlled and ultimately cured.”

4,523 people participated in OneWalk This year, we were delighted to welcome to Conquer Cancer in 2015 and raised Rexall as the new title sponsor of OneWalk over $7.7 million (gross). to Conquer Cancer. Like The Princess Margaret, Rexall cares deeply about the ongoing health of the community, and continues to evolve to meet the changing needs of their customers. “Our pharmacists and pharmacy staff treat, counsel and manage the health of thousands of customers every day, including cancer patients. It seemed entirely appropriate that we team up with one of the most recognized names in cancer research worldwide—Princess Margaret Cancer Centre—to help with this tremendously successful event,” says Jürgen Schreiber, CEO of Rexall Health. The Rexall OneWalk to Conquer Cancer is the result of continuous innovation that began with the Weekend to End Breast Cancer in 2003. Today, participants walk 25 kilometres in a single day and raise funds either individually or as part of a team for the cancer that matters most to them. To date, cummulatively, 58,000 walkers have raised a staggering $156 million for cancer research and care at The Princess Margaret. The event will be 15 years strong in 2017.

These GI Joes are in the trenches fighting some of the toughest types of cancer

The first thought for most of us when we hear ‘GI’ is a soldier on a mission, and the GI Joe’s of the Rexall OneWalk to Conquer Cancer are on a mission too. ’GI’, in this case, stands for Gastro-Intestinal cancers which includes colon, rectal, esophageal, gastric, liver, pancreas, bile duct, gall bladder and small bowel cancers. In 2015, the GI Joe team of 45 walkers, led by Dr. Jennifer Knox, raised over $134,500 through a variety of fundraising activities. It was a very emotional day for many on the team which included patients, cancer centre staff and caregivers walking together. Dr. Knox is passionate about making a difference by developing more and better treatment options for GI patients. She is inspired daily by the amazing courage she sees in her patients, their families and the dedicated team she works with.

Julia inspires a team of cancer warriors

Rosemary Bolognese is the captain of Julia’s Warriors, a team of friends and family who love hockey and originally came together for the 2013 Scotiabank Road Hockey to Conquer Cancer to support Rosemary’s niece, Julia Lisena. “Creating Julia’s Warriors was just one way we showed her how much we care and how we are fighting right alongside her.” Julia was first diagnosed with leukemia when she was only three years old. She spent considerable time in hospitals, but in 2012, the family was happy to celebrate her five-year cancer free milestone. About eight months later, however, they noticed something wrong with her vision. A biopsy confirmed that she had an extremely rare tumor that occurs in the bones of the skull and spine. Doctors said this second diagnosis of a completely different—and rare—type of cancer was the equivalent of being struck by lightning twice. Fortunately, Julia’s last MRI showed that everything is stable, and she is feeling great. Rosemary Bolognese and Julia Lisena Over three years, the team has raised just under $70,000.

On October 15, 2015, The Princess Margaret Cancer Foundation brought together representatives from Toronto’s Chinese community for a special evening called Champion of Hope. It was an exclusive ‘up close and personal’ opportunity for attendees to mingle with and hear from some of Canada’s leaders in cancer care and research, and pay tribute to one of their highest achievers, Dr. Tak Mak. He is one of the world’s most accomplished research scientists who has spent most of his career at The Princess Margaret. Dr. Mak is also beloved by a large community of people who have helped to raise funds for his research into women’s cancers at The Princess Margaret. He has become a face Dr. Tak Mak, Mrs. Helen Ching-Kircher and Dr. Ming-Tat Cheung associated with medical progress and innovation. The evening, generously hosted by BMO Financial Group, included remarks from leaders of the Chinese and Princess Margaret communities, and concluded with a wonderful surprise for Dr. Mak—a performance by one of his favourite musical artists, Canadian guitar legend Liona Boyd. The Foundation is extremely grateful to Mrs. Helen Ching-Kircher and Dr. Ming-Tat Cheung, the co-Chairs of the event. Preparing for a wedding is a time when brides and grooms often reflect on who will be missing from their important day. It might be a parent, a sibling, or a friend. When Helen Sweet was planning her wedding 11 years ago, she thought about her best friend Lisa, who she lost to sarcoma (the same cancer as Terry Fox) when she was just nine-years-old. Cancer later took her cousin, several grandparents and friends. The experience of planning her wedding planted a seed in Helen that led her to start The Bride’s Project. It provides an environmentally, socially and fiscally- conscious option for brides seeking a wedding dress, and for brides seeking the benefits of recycling and reusing after they no longer need their wedding dress. Helen has donated $40,000 over the past two years from the proceeds of The Bride’s Project to support cancer research at Princess Margaret Cancer Centre. “When they hear about The Bride’s Project, many brides see it as a wonderful way they can honour the people they have lost to cancer, as well as those who have survived the disease,” explains Helen. Spreading holiday cheer in a meaningful way

Holidays are a traditional time of saying thank you to customers, suppliers, colleagues and loved ones, and both individuals and organizations sometimes struggle to find the most appropriate and meaningful way to do that. For nine years, Tolin Enterprises— a framing construction contracting firm in Concord, Ontario—has said thank you to their clients and suppliers by making donations in their honour in support of Princess Margaret Cancer Centre. Tony Gargaro, President of Tolin Enterprises donates to The Princess Margaret to support great care, research and its exceptional staff. He has experienced firsthand, the loss of close family members and friends, so this cause is dear to his heart. Marisa Messina, Office Manager at Tolin also shares great admiration for The Princess Margaret, with the loss of her mother and father to cancer. Her sister was recently diagnosed with multiple myeloma, and is in remission at the present time. “Dr. Donna E. If, as many say, Generation X embodies and celebrates individual effort, entrepreneurial spirit and risk- taking, then Jennifer Huggins is right in the bull’s eye. As a teenager, she was a competitive figure skater and then she went on tour as a magician’s assistant. She has learned what it takes to put on a show and engage an audience! Then Jennifer found her real passion— boxing—and she has turned it into a successful business and a way to give back to her community. She now owns Kingsway Boxing Club with two locations in Etobicoke, offers personal training, and is accredited as a boxing referee at the Olympic level. In 2011, she created Fight To End Cancer. Each year, she auditions and selects ten people with no prior boxing experience and trains them for six months in preparation for fight night—a black tie charity gala held in the ballroom of the Old Mill. In five years, Jennifer, her fighters and supporters, have raised over $500,000 for The Princess Margaret.

Palmer Bros. Charity Golf Tournament now aims to improve care for leukemia patients

Five years ago, the Palmer Bros. Charity Golf Tournament began directing the event proceeds to Princess Margaret Cancer Centre after one of their own was diagnosed with leukemia. By then, the event had already been running for more than two decades. Headquartered in Pickering, Ontario, Palmer Bros. has been providing heating and air conditioning services to their community for over 40 years. They have raised over $30,000 for leukemia research at The Princess Margaret. Because they wanted to help leukemia patients and nursing staff in a more direct way, they also purchased furniture and more than a dozen televisions for the cancer centre’s hematology department. Heather Harris and her daughter Jessie More than 160 golfers take part in the tournament each year.

B-Strong achieves a $1-million milestone

In 2015, the B-Strong initiative celebrated its 11th anniversary and the achievement of an incredible milestone. B-Strong has now raised over $1 million for brain tumor research. Its tremendous success is deeply rooted in Markham community outreach and the legacy left behind by Lindsay Bolger. Lindsay started B-Strong when she was first diagnosed with a brain tumor, and she continued to be a passionate advocate for brain cancer awareness and research. She designed and sold B-Strong baseball caps in recognition of patients who lose their hair from cancer treatment, and ran her first fundraiser—the B-Strong Bash—which raised $65,000 in one night. Lindsay was just 26 years old when she passed away in 2009 after a four and a half year battle with brain cancer. In 2011, the City of Markham proclaimed April 22nd (Lindsay’s birthday) as B-Strong Day, and that same year, Lindsay’s family created the B-Strong Walk to raise funds for research at the Pencer Brain Tumor Centre at The Princess Margaret. To date more than 2,500 people have taken part in the five B-Strong Walks.

n November 5, 2015, members gathered at The Omni OKing Edward Hotel for a special dinner to celebrate the inauguration of the Till & McCulloch Society. Honorary member Dr. James Till attended with senior staff from Princess Margaret Cancer Centre. The keynote speaker, Dr. John Dick, spoke about the legacy of Drs. Till and McCulloch and how their work continues to influence cancer research around the world today. As further proof, Dr. Dick described his team’s groundbreaking research published that same day in the journal Science. This research toppled the textbook view that has been held since the 1960s of how human blood is formed. e have also held two Sandwiches & Scientists lunch- Wtime presentations where members have met with several of our top scientists and learned about the progress they are making thanks to philanthropic support. Two of our leaders in immunotherapy, Drs. Pamela Ohashi and Lillian Siu, spoke about the promising strategies their teams are developing and testing that will allow our immune system to be more effective in killing cancer. Dr. Mathieu Lupien described how the study of our ‘epigenome’ is already leading to new cancer therapies.
