B-2 B cells expressing IgM of the suppressed allotype were evident in the spleens of suppressed mice 4 to 6 weeks after cessation of the suppression regimen, whereas B-1 B cells of the suppressed allotype were undetectable for up to 9 months. Although serum IgM of the suppressed allotype was initially depleted in mice suppressed for either allotype, by 7 months of age, there were detectable levels of IgM of the suppressed allotype in the serum; however, the levels were significantly below that found in nonsuppressed mice. When mice were immunized with either the T-independent or T-dependent form of phosphorylcholine, those suppressed for either allotype, and consequently depleted of B-1 B cells of that allotype, did not respond with phosphorylcholine-specific IgM of the suppressed allotype. In contrast, when mice were immunized with 1-3 dextran, the Igh6a allotype-suppressed mice were able to produce dextran-specific IgM of that allotype. -

Epitopes, Isotypes, Allotypes, Idiotypes
Epitope • Epitope or antigenic determinant- is a portion of a foreign protein, or antigen, that is capable of stimulating an immune response. • An epitope is the part of the antigen that binds to a specific antigen receptor on the surface of a B cell (BCR). • Binding between the receptor and epitope occurs only if their structures are complementary. • If they are, epitope and receptor fit together like lock and key. This binding is necessary to activate B-cell for the production of antibodies. • The antibodies produced by B cells are targeted specifically to the epitopes that bind to the cells’ antigen receptors. • Thus, the epitope also is the region of the antigen that is recognized by specific antibodies, which bind to and remove the antigen from the body. Isotype • Each antibody has only one type of (γ, or α, or μ, or ε, or δ) heavy chain and one type of (k or λ) light chain. • The structural differences in the constant region of a heavy chain or light chain determine immunoglobulin (Ig) class and sub- class, types and subtypes within a species. • These constant region determinants are called isotypic determinants or isotypes. Allotype • Based on the genetic difference among individuals. • Although all members of a species inherit the same set of isotype genes, multiple alleles exist for some of the genes. • These alleles encode minor amino acid differences, known as allotypic determinants. • Occurs in some but not all members of a species. • The sum of the individual allotypic determinants displayed by an antibody determines its allotype. Detailed structural analyses of these protein– protein complexes may contribute to our understanding of the pathophysiology of this disease. We analyzed the structure of the IgA-albumin complex using various electrophoresis, mass spectrom- etry, and in silico techniques. The data based on the electrophoresis and mass spectrometry showed that IgA in the sera of patients was dimeric, linked via the J chain. Only dimeric IgA can bind to albumin molecules leading to IgA-albumin complexes, although both monomeric and dimeric forms of IgA were present in the sera. -
Allergen-Specific Igg1 and Igg3 Through Fc Gamma RII Induce Eosinophil Degranulation

Research Article Evidence suggests that eosinophils contribute to inflammation in bronchial asthma by releasing chemical mediators and cytotoxic granule proteins. To investigate the mechanism of eosinophil degranulation in asthma, we established an in vitro model of allergen-induced degranulation. We treated tissue culture plates with short ragweed pollen (SRW) extract and sera from either normal donors or SRW-sensitive patients with asthma. Eosinophils were incubated in the wells and degranulation was assessed by measurement of eosinophil-derived neurotoxin in supernatants. We detected degranulation only when sera from SRW-sensitive patients were reacted with SRW. Anti-IgG and anti-Fc gamma RII mAb, but not anti-IgE or anti-Fc epsilon RII mAb, abolished the degranulation. IgG-depleted serum did not induce degranulation; IgE-depleted serum triggered as much degranulation as untreated serum. Furthermore, serum levels of SRW-specific IgG1 or IgG3 correlated with the amounts of released eosinophil-derived neurotoxin. When eosinophils were cultured in wells coated with purified IgG or IgE, eosinophil degranulation was observed only with IgG. Finally, human IgG1 and IgG3, and less consistently IgG2, but not IgG4, induced degranulation. Thus, sera from patients with SRW-sensitive asthma induce eosinophil degranulation in vitro through antigen-specific IgG1 and IgG3 antibodies. These antibodies may be responsible for degranulation of eosinophils in inflammatory reactions, such as bronchial asthma. The monomodular form normally comprises four functional genes, but in duplicated RCCX segments generally only the gene-encoding complement component C4 produces a protein. C4 genes can code either for a C4A or a C4B isotype protein and exhibit dichotomous size variation. Distinct RCCX variants show association with numerous diseases; however, identification of the basis of these associations is often challenging, not least because the RCCX is localized in the major histocompatibility complex (MHC) region, a genomic area characterized by exceedingly long-range linkage disequilibrium. Here we present a detailed analysis on RCCX variants and their relationship with so-called ‘ancestral’ or ‘conserved extended’ MHC haplotypes in healthy Caucasians. In addition to former investigations, precise order and size of all C4A and C4B genes were determined even in trimodular RCCX structures. Considering C4 copy numbers, length, isotype specificity and CYP21A2 copy numbers, we have identified 15 distinct RCCX variants and described the RCCX structures involved in 29 repeatedly occurring MHC haplotypes. The etiology is covid-19 ,the problem at the moment is the lack of cure for the disease.In this study five continents where of interest and they include North and South America,Europe,Africa and Asia.Data was obtained from the internet from worldometer.info/coronavirus/country and cases from February to 15 december 2020 were considered and analysed statistically using analysis of variants to determine monthly incidence and prevalence,case fatality,morbidity ,mortality and population at risk.The reason for this survey is to establish ways to decrease the high morbidity and mortality rates , reduce the devastating economic impact of this disease and increase daily socialization among Humans and curb hunger and boredom that covid-19 has caused.More so the it is a global pandemic needing urgent attention.In conclusion the morbidity and mortality where highest in the months of July and October and the population at risk are 95%-98%.Prophylactic measure to help alleviate the incidence of this disease include daily administration of blood tonic and ascorbic acid in their daily recommended dosages pre- infection to enhance growth of tissues and cellular epithelization and boost energy generation and build . -
