  • Evidence for Male XO Sex-Chromosome System in Pentodon Bidens Punctatum (Coleoptera Scarabaeoidea: Scarabaeidae) with X-Linked 18S-28S Rdna Clusters
Genes Genet. Syst. (2003) 78, p. 427–432 Evidence for male XO sex-chromosome system in Pentodon bidens punctatum (Coleoptera Scarabaeoidea: Scarabaeidae) with X-linked 18S-28S rDNA clusters Roberto Vitturi1*, Mariastella Colomba2, Nicola Volpe1, Antonella Lannino1 and Mario Zunino2 1Dipartimento di Biologia Animale, Università di Palermo, Via Archirafi 18, 90123 Palermo, Italy 2Istituto di Ecologia e Biologia Ambientale, Università di Urbino, via Sasso 75, 61029 Urbino (PU), Italy (Received 24 December 2002, accepted 21 Novembr 2003) In scarab beetle species of the genus Pentodon, the lack of analysis of sex chro- mosomes in females along with the poor characterization of sex chromosomes in the males, prevented all previous investigations from conclusively stating sex determination system. In this study, somatic chromosomes from females and spermatogonial chromosomes from males of Pentodon bidens punctatum (Coleoptera: Scarabaeoidea: Scarabaeidae) from Sicily have been analyzed using non-differential Giemsa staining. Two modal numbers of chromosomes were obtained: 2n = 20 and 19 in females and males, respectively. This finding along with other karyological characteristics such as the occurrence of one unpaired, het- erotypic chromosome at metaphase-I and two types of metaphase-II spreads in spermatocytes demonstrate that a XO male/XX female sex determining mecha- nism – quite unusual among Scarabaeoidea - operates in the species investigated here. Spermatocyte chromosomes have also been examined after a number of banding techniques and fluorescent in situ hybridization with ribosomal sequences as a probe (rDNA FISH). The results obtained showed that silver and CMA3 staining were inadequate to localize the chromosome sites of nucleolus organizer regions (NORs) due to the over-all stainability of both constitutive heterochroma- tin and heterochromatin associated to the NORs.

Zoosystema 35 (4): 537-549. http://dx.doi.org/10.5252/z2013n4a7 ABSTRACT Augosoma centaurus Fabricius, 1775 (Melolonthidae: Dynastinae), one of the largest Scarabaeoid beetles of the Ethiopian Region, is classified in the tribe Dynastini MacLeay, 1819, principally on the basis of morphological characters of the male: large frontal and pronotal horns, and enlargement of fore legs. With the exception of A. centaurus, the 62 species of this tribe belong to ten genera grouped in Oriental plus Australasian and Neotropical regions. We performed cytogenetic studies of A. centaurus and several Asian and Neotropical species of Dynastini, in addition to species belonging to other sub-families of Melolonthidae Leach, 1819 and various tribes of Dynastinae MacLeay, 1819: Oryctini Mulsant, 1842, Phileurini Burmeister, 1842, Pentodontini Mulsant, 1842 and Cyclocephalini Laporte de Castelnau, 1840. The karyotypes of most species were fairly alike, composed of 20 chromosomes, including 18 meta- or sub-metacentric autosomes, one acrocentric or sub-metacentric X-chromosome, and one punctiform Y-chromosome, as that of their presumed common ancestor.

  • Abstract Introduction Objective
The Journal of Agriculture and Environment Vol:13, Jun.2012 ... .............Technical paper DISTRIBUTION OF WHITE GRUBS IN THREE ECOLOGICAL DOMAINS OF NEPAL Dipak Khanal1*, Yubak Dhoj GC2, Marc Sporleder3 and Resham B. Thapa1 ABSTRACT A survey was conducted to study the abundance and distribution of white grubs in three districts representing different ecological domines in the country during June-July 2010. Two light traps were installed for two nights in two locations each of Makawanpur, Tanahu and Chitwan districts, and a season long light trap was installed at Mangalpur of Chitwan district from April to September 2010 for assessing scarab beetles flight activity. The ‘simple matching coefficient' revealed high similarity >70% between two sites in each of the districts, while a similarity of 29-50% was observed between sites of different districts. The Jaccard coefficient revealed the same trend. However, coefficients were much lower, above 40% when comparing sites within a district, and below 20% when compared sites among the districts. The dominant species in Chitwan were Anomala dimidiata Hope (24%) followed by Maladera affinis Blanchard (23.75%), Anomala varicolor (Gyllenhal) Rutelinae (23%), Heteronychus lioderus Redtenbacher (14%) and Holotrichia sp (7%). The flight activity and species composition of scarab beetles in the three districts appeared to be different. Key words: Abundance, distribution, dominant species, white grub INTRODUCTION White grubs (Coleoptera: Scarabaeidae) are the soil-living and root feeding immature stages of scarab beetles, of which both adult and larval stages are destructive in nature. The white grub family is the second largest omnipresent family, which includes over 30,000 species (Mittal, 2000).

Department of Entomology, Faculty of Science, Ain- Shams University, Cairo, Egypt Abstract In Egypt, The white grub, Pentodon algerinum (Coleoptera: Scarabaeidae) damages various economic important plants recently strawberry. This study aimed to DNA identification and first evaluation of the Spodopteda littoralis Nuclear polyhedrosis virus (SpliNPV) and Pentodon algerinum Nuclear polyhedrosis virus (PNPV) against Pentodon algerinum third instar larvae in the laboratory to find successful safe alternative control method against this pest. Bioassay was made by two experiments, one used five SpliNPV concentrations, 6.7X106, 3.11X107, 1.1X108, 3.64X109 and 3.64X1010 Polyhedral inclusion bodies (PIB/ml) achieved 80, 100, 100, 80 & 90% mortality respectively during thirteen days. Another experiment used four PNPV concentrations, 8.2x107, 8.2x 108, 2.6x109 and 8.2x109 PIB/ml caused 100, 70, 67 & 72.7% mortality respectively during ten 14 9 9 days. SpliNPV Lethal concentrations LC50 6.2x10 & LC90 8.9x10 PIB/ml were more than PNPV LC50 2.8X10 & LC90 1.8X108 PIB/ml. Thus PNPV and SpliNPV were effective against Pentodon larvae but PNPV was effective and specific more than SpliNPV. DNA of SpliNPV and PNPV was identified by RAPD PCR using four RAPD primers, Operon A9, A20, B17 & B7 (Operon Technologies, Inc., Alameda, CA, USA) using automated (Bio Rad, USA) thermal cycler.

Here we compare the morphology of the antennal surface between major pleurostict lineages, including Cetoniinae, Dynastinae, Melolonthinae, Rutelinae, and a few outgroups, including Scarabaeinae and Hybosoridae. We identifed various types of antennal sensilla morphologically and searched for phylo- genetic patterns of sensilla within the Scarabaeidae. Sensilla were examined using SEM micrographs of 36 species and the occurrence of the diferent types of antennal sensilla was studied for each species. We observed a high diversity of sensilla, including multiple transitional forms. There were also a number of other interesting structures on the antennal surface with adaptive value, such as elongate elevations, serial bags, and felds of setae. Our results confrm earlier fndings that within pleurostict scarabs there has occurred a clear diferentiation of sensilla composition and patterns. Their morphological development over phytophagous beetle groups comprising about 25,000 spe- millions of years resulted in divergence in body shape, size, cies (Scholtz and Grebennikov 2005).
