It is also commonly known as HCC-1 and is produced as a protein precursor that is processed to generate a mature active protein containing 74 amino acids with a 46% identical amino acid composition to CCL3 and CCL4. This chemokine is expressed in various tissues including spleen, bone marrow, liver, muscle, and gut. CCL13 activates monocytes, but does not induce their chemotaxis. Human CCL13 is located on chromosome 17 within a cluster of other chemokines belonging to the CC family. Recombinant human HCC-1 produced in E. coli is a single, non-glycosylated polypeptide chain of 72 amino acids and a molecular mass of 8,41 Dalton. The rHu HCC-1 has purified by proprietary chromatographic techniques. The biological activity is calculated by its ability to chemoattract human monocytes at a concentration of 5-20 ng/ml. Specific Activity see below Formulation White, lyophilized (freeze-dried) powder. Lyophilized from a sterile-filtered protein solution in 20 mM PBS (pH 7.4) and 150 mM NaCl. Reconstitution Please Note: Always centrifuge product briefly before opening vial. The lyophilized protein should be reconstituted in sterile, ultra-pure water to a concentration of 0.1 - 1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at -20°C for future use. Purity Greater than 99% (determined by SDS-PAGE, FPLC and RP-HPLC analysis). Endotoxin Level < 0.1 ng per µg of rHu HCC-1 (1EU/µg). Storage The lyophilized protein, though stable at room temperature for up to 3 weeks, is best stored desiccated at -20°C. Reconstituted rHu HCC-1 should be used immediately or stored long-term in undiluted working aliquots at -20°C. For long-term storage it is recommended to add a carrier protein (0.1% endotoxin-free HSA or BSA; e.g. Cat.No. C-69500A). Avoid repeated freeze-thaw cycles.
