Late Pleistocene-Holocene Amphibians Palaeontologia Electronica palaeo-electronica.org Late Pleistocene-Holocene amphibians from Okinawajima Island in the Ryukyu Archipelago, Japan: Reconfirmed faunal endemicity and the Holocene range collapse of forest-dwelling species Yasuyuki Nakamura and Hidetoshi Ota ABSTRACT Amphibian fossils excavated from two Late Pleistocene-Holocene fissures in the southern part of Okinawajima Island in the Ryukyu Archipelago were identified through detailed comparisons with skeletal specimens of extant taxa. The identified species (eight frogs and two newts) were confined to extant elements on the island and were mostly endemic to either the island or the central part of the archipelago including it. Previous Late Pleistocene records of extralimital frogs (two mainland Japanese spe- cies [Glandirana rugosa and Rana tagoi] and one Southeast Asian frog currently natu- ralized in the Ryukyus [Polypedates leucomystax]) from one of the study sites (the Minatogawa Fissure) are therefore considered misidentifications. This may also be the case with Fejervarya kawamurai, a synanthropic frog currently widely distributed in East Asia, including the Central Ryukyus. Of the species obtained from the Late Pleis- tocene deposits at these sites, five frogs (Limnonectes namiyei, Babina holsti, Odor- rana ishikawae, O. narina, and Rana ulma) are forest-dwellers currently confined to the northern forested area of Okinawajima. Additionally, none of these species were detected in the middle Holocene deposits from one of the sites, the Sashiki Fissure. Our findings indicate that the southern limestone area of Okinawajima in the Late Pleistocene harbored a dense and humid forest with diverse lotic habitats, similar to the northern forested area of the island at present, and that these environmental condi- tions were lost by the midpoint of the Holocene Period. The taxonomy of Babina sensu lato was controversial in the past decades. In this study, the phylogeny of genus Babina sensu lato was re-constructed based on genetic analysis, morphological comparison and advertisement call analysis. We found that Babina sensu stricto and previous subgenus Nidirana should be two distinct genera in the family Ranidae. N. caldwelli is confirmed to be a synonym of N. adenopleura because of the small genetic divergence and the lack of distinct morphological differences. A new species, Nidirana nankunensis sp. nov. is described based on a series of specimens collected from Mt. Nankun, Guangdong Province, China, which can be distinguished from other known congeners by having a behavior of nest construction, distinctive advertisement calls, significant divergence in the mitochondrial genes, and a combination of morphological characters. Genetic differentiation between populations of R. amurensis from Sakhalin and the Maritime Territory was found to be minimal. The resultant phylogenetic tree indicates monophyly of brown frogs and earliest divergence of R. amurensis among all the brown frogs studied. For this reason, separation of R. amurensis from the R. japonica group is suggested, but separation of the R. sauteri complex as a distinct ge- nus or subgenus Pseudorana is not supported. One group including populations of Alishan (central Taiwan) and Sanyi (western Taiwan) seemed to be closest to R. tagoi and the presumptive common ancestor of these frogs is thought to have diverged very early.   • Nansei Islands Biological Diversity Evaluation Project Report 1 Chapter 1
    Introduction WWF Japan’s involvement with the Nansei Islands can be traced back to a request in 1982 by Prince Phillip, Duke of Edinburgh. The “World Conservation Strategy”, which was drafted at the time through a collaborative effort by the WWF’s network, the International Union for Conservation of Nature (IUCN), and the United Nations Environment Programme (UNEP), posed the notion that the problems affecting environments were problems that had global implications. Furthermore, the findings presented offered information on precious environments extant throughout the globe and where they were distributed, thereby providing an impetus for people to think about issues relevant to humankind’s harmonious existence with the rest of nature. One of the precious natural environments for Japan given in the “World Conservation Strategy” was the Nansei Islands. The Duke of Edinburgh, who was the President of the WWF at the time (now President Emeritus), naturally sought to promote acts of conservation by those who could see them through most effectively, i.e. pertinent conservation parties in the area, a mandate which naturally fell on the shoulders of WWF Japan with regard to nature conservation activities concerning the Nansei Islands. This marked the beginning of the Nansei Islands initiative of WWF Japan, and ever since, WWF Japan has not only consistently performed globally-relevant environmental studies of particular areas within the Nansei Islands during the 1980’s and 1990’s, but has put pressure on the national and local governments to use the findings of those studies in public policy. Unfortunately, like many other places throughout the world, the deterioration of the natural environments in the Nansei Islands has yet to stop.

Our results supported a sister group relationship of Amolops ricketti and Amolops hainanensis. However, the grouping of Amolops mantzorum and Amolops monticola needs to be resolved with more data. Amolops tormotus was nested in genus Odorrana. Thus, recognition of the A. tormotus group is unwarranted and A. tormotus should be referred to genus Odorrana as O. tormota. This species is the sister group of O. nasica plus O. versabilis. The new classification implies that the genus Wurana is to be considered as junior subjective synonym of Odorrana.

Clade A contained brown frogs with 24 chromosomes and was split into two distinct subclades (Subclade A-1: R. chensinensis and R. huanrenensis; Subclade A-2: R. dybowskii). Polytomous relationships among populations of R. chensinensis and R. huanrenensis suggested the necessity of further taxonomic assessment.

Both the gastro­ myzophorous tadpoles and adults have distinct geographical variations in morphology. Clinal variation in the shape of the upper jaw sheath and the number of labial tooth rows occurs from the foothills of western Taiwan eastward through the Central Mountain Range to the eastern slopes of the Central Mountain Range. This geographical pattern is supported and validated by multivariate analyses of adult morphology. Ac­ cording to the dispersal hypothesis, this geographical pattern may suggest that the dispersal center (= initial range of the ancestral group) was located in the western foothills, and it may indicate that the population expanded radially through the clinally variable transition zone to areas adjacent to the ridge of the Central Mountain Range (= range of the derived group). However, the establishment of the geographical pattern of R. sauteri seems to be better explained by the vicariance hypothesis which supports the occurrence of isolation-based dichotomous divergence within this grossly defined taxon.

The discovery of novel cathelicidins is important research; however, despite the identifcation of many cathelicidins in vertebrates, few have been reported in amphibians. Here we identifed a novel cathelicidin (named cathelicidin-OA1) from the skin of an amphibian species, Odorrana andersonii. Produced by posttranslational processing of a 198-residue prepropeptide, cathelicidin-OA1 presented an amino acid sequence of ‘IGRDPTWSHLAASCLKCIFDDLPKTHN′ and a molecular mass of 3038.5 Da. Functional analysis showed that, unlike other cathelicidins, cathelicidin-OA1 demonstrated no direct microbe-killing, acute toxicity and hemolytic activity, but did exhibit antioxidant activity. Importantly, cathelicidin-OA1 accelerated wound healing against human keratinocytes (HaCaT) and skin fbroblasts (HSF) in both time- and dose-dependent manners. Notably, cathelicidin-OA1 also showed wound-healing promotion in a mouse model with full-thickness skin wounds, accelerating re-epithelialization and granulation tissue formation by enhancing the recruitment of macrophages to the wound site, inducing HaCaT cell proliferation and HSF cell migration. This is the frst cathelicidin identifed from an amphibian that shows potent wound-healing activity. These results will help in the development of new types of wound-healing agents and in our understanding of the biological functions of cathelicidins. Cathelicidins, which belong to a group of cationic peptides with amphipathic properties, play critical roles in host defense1.
