The current need to develop new antifungal,anticancer, antibiotic and antiviral drugs has led to an intenseresearch effort into the discovery, isolation and structure determinationof potential medicinal agents from marine algae. In the past two years, I have participatedin a drug discovery program designed forantitumor, antifungal and other agents of potential pharmaceutical utility from themarine cyanobacterium Lyngbya majuscula. This research utilized modernchromatographic and spectrochemical techniques including2D NMR spectroscopy. Brine shrimp toxicity guided the fractionationthat led to the discovery of the biologically activecompound kalkitoxin from a Curacao Lyngbya majuscula extract.The structure of this new thiazoline ring-containing lipid wasdetermined spectroscopically by interpretation of 2D-NMR experiments,including heteronuclear multiple quantum coherence (HMQC),heteronuclear multiple-bond coherence spectroscopy (HMBC) and'H-'H COSY at room temperature and elevated temperature.Kalkitoxin shows modest molluscicidal toxicity, good brine shrimptoxicity and extremely potent ichthyotoxicity. A. van Leeuwenhoek, November 4, 1681 lthough the term “protozoa” has been around for nearly 200 years, and was used in the context of a phylum for about 100 years, it is now ap- parent that it defines no more than a loose assemblage, or grade, of pri- Amarily single-celled, heterotrophic, eukaryotic organisms—it does not define a monophyletic assemblage meriting single-phylum status. The kingdom Protista (or “Protoctista”) includes those organisms traditionally called protozoa, as well as some autotrophic groups. But the kingdom Protista itself is not united by any unique distinguishing features or synapomorphies, and the boundary between autotrophy and heterotrophy is blurred in these organisms. Protists are definable only as a confederation of eukaryotes lacking the tissue level of organization seen in plants, animals, and fungi. Invertebrate biology texts have traditionally treated the “pro- tozoa,” and for many biology students this coverage will be their only detailed exposure to this important group of organisms. In this chapter we include those groups traditionally lumped under the name “protozoa” (i.e., the heterotrophic protists), and for completeness, we also briefly cover the commonly recognized autotrophic protist phyla (Box 5A).
