Gryglewski Jagiellonian Medical Research Centre, Kraków, S³awkowska 17, PL 31-016 Kraków, Poland Correspondence: Ryszard Gryglewski, e-mail: [email protected] Abstract: Prostanoids are cyclic lipid mediators which arise from enzymic cyclooxygenation of linear polyunsaturated fatty acids, e.g. arachidonic acid (20:4 n 6, AA). Biologically active prostanoids deriving from AA include stable prostaglandins (PGs), e.g. PGE , PGF =, PGD , PGJ as well as labile prostanoids, i.e. PG endoperoxides (PGG , PGH ), thromboxane A (TXA ) and prostacyclin (PGI ). A “Rabbit aorta Contracting Substance” (RCS) played important role in discovering of labile PGs. RCS was discovered in the Vane’s Cascade as a labile product released along with PGs from the activated lung or spleen. RCS was identified as a mixture of PG endoperoxides and thromboxane A . Stable PGs regulate the cell cycle, smooth muscle tone and various secretory functions; they also modulate inflammatory and immune reactions. PG endoperoxides are intermediates in biosynthesis of all prostanoids. Thromboxane A (TXA ) is the most labile prostanoid (with a half life of 30 s at 37°C). It is generated mainly by blood platelets. TXA is endowed with powerful vasoconstrictor, cytotoxic and thrombogenic properties. Again the Vane’s Cascade was behind the discovery of prostacyclin (PGI ) with a half life of 4 min at 37°C. It is produced by the vascular wall (predominantly by the endothelium) and it acts as a physiological antagonist of TXA .

KORBUT PHARMACOLOGY AT THE JAGIELLONIAN UNIVERSITY IN KRAKOW, SHORT REVIEW OF CONTRIBUTION TO GLOBAL SCIENCE AND CARDIOVASCULAR RESEARCH THROUGH 400 YEARS OF HISTORY Chair of Pharmacology Jagiellonian University Medical College, Krakow, Poland Pharmacology at the Jagiellonian University was taught since the foundation of the University in 1364 under then used names of medical botany and pharmacognosy. The first in Poland Division of Pharmacology and Pharmacognosy was created in 1857. Modern era in the history of pharmacology in Krakow starts in 1929 when Prof. Janusz Supniewski became the Head of the Department. He was the relentless researcher whose scientific interests were as diverse as ranging from anticancer chemotherapeutics, antibiotics, and oral hypoglycemic drugs to lipid-lowering agents. Skills of organic chemistry synthesis were of paramount importance for the scientific achievements. Prof. Supniewski died in 1964, leaving the Department well equipped in instruments. He raised in his laboratory many eminent scientists who later beacame heads of pharmacology departments throughout Poland. In 1964, the Head of the Department of Pharmacology became Prof. Ryszard Gryglewski. Under his leadership the Department focused scientific efforts on various aspects of cardiovascular pharmacology. Prof. Gryglewski established collaborations with many of the best pharmacology researchers in the world, including Sir John Vane, Nobel Prize laureate. Prof. Gryglewski's assistants had ample opportunity to train in these laboratories and to bring these skills back to Krakow. Prof. Gryglewski and his team published many articles in the most prestigious scientific journals. The most important themes included discovery of prostacyclin, role of nitric oxide and of free radicals for vascular biology.
